Development and applications of ionization techniques in ambient mass spectrometry

Ambient mass spectrometry is a sharply developing subject area. Desorption electrospray ionization (DESI) and desorption atmospheric pressure photoionization (DAPPI) are two types of ionization techniques in the ambient mass spectometry. Herein, they were investigated in two research areas: (1) TLC analysis. (2) Localization of insect defense glands.

Anovel defensin from the mucus of the wood wasp Xiphydria camelus

We have isolated a novel defensin from the mucus of the wood wasp Xiphydria camelus. The Edman degradation in 50 cycles revealed a peptide sequence similar to other insect defensins. The complete 55 amino acid residues sequence of X. camelus defensin was proposed based on the mass spectrometry analysis and by the deduction based on the comparison with the sequences of other hymenopteran defensins. The defensin was tested against both Gram-positive and -negative bacteria.

Analysis of plant membrane lipids by RP-HPLC/HR-ESI-MS

Monogalactosyldiacylglycerols (MGDGs) and digalactosyldiacylglycerols (DGDGs) are glycolipids forming the major part of biological membranes in photosynthesizing organisms. The individual species differ from fatty acids linked to sn-1 and sn-2 possition on glycerol backbone. Complexity of their mixtures is increased by enzymatic or chemical oxidation of unsaturated species. The aim of this work was to develop and optimize RP-HPLC/HR-ESI-MS2 methods for analysis of oxidized and non-oxidized MGDGs and DGDGs in plant samples. MGDGs and DGDGs were isolated from Arabidopsis thaliana and Melissa officinalis.
Separation and identification of monogalactosyldiacylglycerols and digalactosyldiacylglycerols by RP-HPLC/ESI-MS

RP-HPLC/ESI-MS methods for analyzing complex mixtures of monogalactosyldiacylglycerols (MGDGs) and digalactosyldiacylglycerols (DGDGs) was developed. The methods allow separation and detection of more than 30 molecular species of MGDGs and more than 40 molecular species of DGDGs in plant samples.
New peptides with antimicrobial activity isolated from the venom of the bumblebee Bombus pascuorum (Hymenoptera: Apidae)

Six new heptadecapeptides were isolated and synthesized by standard Fmoc solid phase peptide synthesistogether with several structural analogues. Antimicrobial activities were correlated with the peptides hydrophobicity, hydrophobic moment, alpha-helicity and HPLC retention time.
Hmotnostní spektrometrie peptidů z jedových váčků čmeláků

Five novel structurally related peptides were isolated and characterized from the venom of the bumblebee Bombus terrestris. Some of these peptides consisting of 13-18 amino acids residues presented strong antimicrobial activity against Bacillus subtilis.
