Optimization of the imaging spectrophotometer

Thesis deals with an optimization of an imaging spectrophotometer which consists of a monochromator, a CCD camera with extended detection range to UV spectra and an imaging system. Main benefits of the optimization are utilization of a new lightsource, a connection of the monochromator and the imaging system by a fibre bundle and upgrade of a sample angle adjustment system by use of a auxiliary CCD camera. More changes were made to increase accuracy of measurements and ease of use of the apparatus.
Design of microscope LED illumination system

The objective of this bachelor thesis was to design illumination assemblage in confocal microscope Confocal 2002. First part of this paper describes types and components of confocal microscopes. For correct function of the microscope, high-power illumination by a white light emmiter is required. Based on the analysis of the background research, suitable LED diode was selected. In experimental part of this work, eight optical systems are proposed, that has been assembled and evaluated. Based on the evaluation of obtained paramteres, the optimal assemblage was selected and designed for production.
Components for laser micromanipulation in a coherence-controlled digital holographic microscope

In the Bachelor's thesis, there was described the general principle of operation of the optical trap, along with various options for creating optical tweezers. The function and the main parameters of holographic microscope with the off-axis achromatic and spatially invariant interferometer were described, too. Two variations of laser tweezers connectable to coherence-controlled second generation holographic microscope were described in detail, calculated and designed at IPE VUT. The optimization of the two variations was performed in the software ZEMAX. A 3D model in the Autodesk Inventor PROFESSIONAL 2010 was created for the chosen design.
Flow-chambers for microscopy of living cells

The perfusion chamber for long term observing of live cells by the means the Coherence-Controlled Holographic Microscope (CCHM) was designed. CCHM was built and designed at the Laboratory of the optical microscopy at the Institute of Physical Engineering, Brno University of Technology. CCHM can quantitatively evaluate dynamical changes inside live cells thanks to the quantitative information about phase shift in each pixel of the image. In order to demonstrate advantages of CCHM experimentally, it is important to keep the live cells in the good conditions. This is made by adding the fresh cultivation medium for studied cells directly in the microscope. In contrast to the stationary chamber the perfusion chamber allows both the cultivation medium exchange and the application of biological reagents without the necessity of removing the chamber from the microscope. Therefore we can study the vital signs of cells before and after the application of reagents. An original perfusion system with accessories compatible with CCHM was designed. The design is based on the previously published perfusion system solutions that are referred to in this thesis. The flow characteristics and medium exchange process was discussed and a modification of the internal geometry, based on numerical simulations, was introduced. The applicability of this perfusion chamber has been proven for the CCHM and even for different types of microscopes. The reactions of tumor and epithelial cells during the change of the environment from the cultivation medium to the physiologically solution were studied.
Design of projection illuminator

The aim of this diploma thesis is to design a suitable reflector concentrating a light beam into a circular hole in a gobo. The light beam behind the screen should have good efficiency and at the same time good homogeneity. For the reflector design and analysis a software for designing optical systems called LightTools was used. The reflector was designed by six different methods. Several smooth and several faceted reflectors were designed. The selected reflector was analyzed in combination with a varifocal lens. For selected reflector was designed mechanical store with respect to the anticipated operating conditions. This thesis serves as the basis for the producing of the reflector.
Design of an active protection system LIDAR for the detection of subsonic missiles

The diploma thesis focuses on the design of an Active protection system LiDAR, with emphasis on the research for the operation of these systems and the components of which the system consists. It specifies the basic optomechanical parameters that the designed system must achieve. The thesis introduces a conceptual design of an optical system for ground surveillance and aviation surveillance. LiDAR systems are made of off-axis or Cassegrain telescopes. As a part of this thesis are also designs of an optical illuminating system and a zoom lens for infrared imaging.
Model creation and verification of imaging polarimeter in Mueller formalism

The aim of this master thesis is to create a model of imaging polarimeter developed by the company Meopta - optika, s.r.o. using Mueller formalism. The theoretical part describes Mueller formalism and polarimeter experimental setup. The practical part describes the created model and its software implementation in MATLAB environment. The model was verified by comparing measurement results with results obtained using the original model based on Jones formalism. In addition a sample holder design and its drawing documentation was created.
Mechanical design of measurement system for reactive ion etching system

The aim of this work is to design fully working measuring system for the reactive ion etching system (RIE). The Michelson interfometer, previously developed in Ústav přístrojové techniky, v.v.i., is used in this work. The theoretical part is aimed at description of interferometric methods for precise measuring of length. In next part the etching proces with RIE is described. In practical part the testing system was constructed from the parts of Thorlabs company . The functionality was controlled with this system and the results of measuring were compared with the profilometer. In next step technical drawings were created and the whole system was made.
Optical design of telecentric F-theta objective

This thesis deals with optical design of f-theta lens for purposes of precision micromachining using a laser beam. Optical design consists of creating a nominal design according to specifications given by Meopta – optika s.r.o company and a tolerance analysis, which describes manufacturability of given design. Specifications given are most importantly diffraction limited image quality, correction of distortion throughout the field and telecentricity of systém in image space. F-theta lens in this thesis was created using an optical design software called Zemax.
