Conjugates based on N-(2-hydroxypropyl)methacrylamide represent a new generation of polymeric anticancer drugs with improved therapeutic potential and considerably decreased nonspecific side effects. Antibody -targeted derivatives show specific active targeting and nonspecific passive accumulation in solid tumors.

Conjugates of N-(2-hydroxypropyl)methacrylamide with doxorubicin containing human IVIg were used for palliative treatment in patients with advanced cancer. More thena 116 biochemical and immunological parameters were tested. A decrease of some parameters with patological values was seen, and physiological values were occasionally reached, including some TAA.

Unwanted side effect of anticancer treatment could be eliminated by affinity therapy. It is necessary to find targeting system which deliver drugs specifically to tumor site. Polymeric conjugates based on N-(2-hydroxypropyl)methacrylamide are new generation polymeric anti-cancer drugs with enhanced cytostatic activity, but minimalized unwanted side effects.
