Analysis of gene polymorphisms in the MBL2 gene and its diagnostic significance

MBL2 gene codes a protein called mannose binding lectin, a component of our immune system. This protein takes a part in a nonspecific humoral immune response, opsonizing pathogenic microorganisms and providing activation of the lectin pathway in the complement system, leading to elimination of pathogens and simultaneously inducing the inflammation. MBL deficiencies are widely researched in cases of recurrent infections, autoimmune diseases and others. MBL-protein serum level is affected by polymorphisms in MBL2 gene, located in three codones marked as '52', '54' and '57'. A mutation of the '52' codon is referred to as an allele D, for the '54' codon as an allele B and for the '57' codon as an allele C. In many cases the alleles might be referred to as the alleles zero (0), or as an 'A' in the case of wildtype alleles. Other polymorphisms are located in non-translated locations. The first one may be found in the promotor 1, in a position -550 (variants H/L); the second one in a position -221 (variants X/Y); and the third one in a non translated 5'-end-part of a locus in a position +4 (variants P/Q). The practical part of this bachelor thesis was executed in the genetic lab in the company GENLABS s.r.o., České Budějovice. I focused on the analysis of MBL2 polymorphisms located in exon 1 (alleles B, C, D). I examined 30 patient samples, 25 of them were provided by patients with ongoing dementia and 5 of the samples were taken from patients with no signs of dementia. The protocol of the analysis consisted of an isolation of DNA, a measurement of DNA concentration, a preparation and an execution of a PCR method followed by a control of the products in a gel electrophoresis. These PCR products were then purified and sequenced. A precise description of the analysis including an overview of the obtained results are summed in the practical part of this bachelor thesis.
Musical activities in nursery school focused on children at two and three years

The thesis deals with the musical activities of two-year and three-year-old children in kindergarten. The theoretical part describes the characteristics of two-year and three-year-old children, the inclusion of two-year-old children in kindergarten, the importance of music and the child's musical development. It also deals with musical activities focusing on two-year and three-year-old children. The practical part of the work was focused on the research of particular musical activities in the kindergarten of the above mentioned age group from the point of view of the teachers and also on the musical environment in family. The research was conducted in the form of an interview with kindergarten teachers, a questionnaire survey among the parents of these children, and an analysis of songs and songbooks for the youngest children. The main objective was to find out how teachers perform musical activities with children, and what they see as the biggest differences between two- and three-year-old children. Next goal was to find out if parents work with children in this area and how. Whether the selected sample of songbooks contains songs of the appropriate range and pitch and whether they contain examples of possible instrumental and musical involvement of the children. Procedural Periods in Roman Law Submitted thesis is focused on chosen questions of procedural periods in Roman law. It is too focused on chosen institutes of roman procedural law. Firstly the thesis dissertates about general information and about and progress of roman procedura law. The relation of roman procedural law to singular procedurals periods was emphasized in the introduction. The crucial part of this opening leans in analysis of singular judicial magistrates. Further the contexture for individual procedural parties was mentioned. The main part of thesis describes separates procedura periods in detail. It makes through particular roman-law institutinos which are alone influenced by procedural periods. The single chapter disserts on prescription of rights, preclusion and temporal character of some actions. These law institutions are very important. It is also crucial in contemporary law. This chapter follows on discourse about executory periods. The final part of thesis is created by question of time and its computation, especially in law and in standing roman life. Convergence of the Czech Republic to the European Union

This thesis deals with an analysis of the convergence of the Czech Republic to the first fifteen member states of the European Union. The aim of this thesis is to evaluate different types of resources that are available for the Czech Republic from the European Union and if and to what extent the development levels of the Czech Republic and the EU 15 countries come closer. The first part of this thesis describes resources provided by the European Union to its member states and funds used to their allocation. The second part focuses on drawing of these resources by the Czech Republic. Specifically, the amount of resources allocated to the Czech Republic is described and the ability of the Czech Republic to use them is analyzed. The third part, core of this work, concentrates on an analysis of several fundamental development indicators. Main indicators of both nominal and real convergence are evaluated, as well as the Human Development Index. These indicators are applied on both the Czech Republic and the EU 15 countries, they are compared and the level of the convergence between the two units is evaluated. Speech Politeness Strategy in Czech and Spanish Language: pragmalinguistic and anthropolinguistic comparative research

The thesis is concerned with the definition of the speech politeness as a unique pragmatic system which is specific for each language community and which creates in speakers a pragmatic habitus or an abstract model for culturally speech behavior, manifested as a polite speech norm in geographically and time-limited context of a linguistic market. In the practical part, the thesis deals with the comparison of the language devices used by Czech and Spanish speakers to maintain the safe relationship with the addressee in order to achieve a successful socio-economic exchange. The study focuses on speech courtesy on one hand and comparison of two ethnolinguistic areas from the point of view of linguistics, pragmatics, sociology and cultural anthropology.
