New regulatory hormones of breast milk

Background: Breastfeeding is supposed to exert long-term protection of the infant from the development of civilisation diseases later in life. Adiponectin, leptin and AFABP are candidate proteins in nutritional programming of the infant. Aim: To determine intraindividual changes of breast milk levels of regulatory hormones (adiponectin, leptin, AFABP) and adipophilin in breast milk during 12 months of lactation. Methods: Proteins were measured using high sensitive ELISA method in 72 healthy mothers within 2 days of beginning of lactation (day 0) and 1, 3, 6 and 12 months following delivery. Results: Adiponectin levels in breast milk on day 0 (D0) were 22.8 ± 0.8 (mean ± S.E.M.), in 1 month (M1) 22.0 ± 0.6, in 3 months (M3) 20.5 ± 0.6, in 6 months (M6) 21.4 ± 0.8, and in 12 months (M12) 25.7±1.4 ng/ml. AFABP levels were 12.3 ± 2.0, 6.2 ± 1.3, 1.3 ± 0.2, 2.5 ± 1.0 and 4.6 ± 1.9 ng/ml, respectively. Leptin levels were 0.3 ± 0.04, 0.2 ± 0.03, 0.1 ± 0.01, 0.1 ± 0.02 and 0.2 ± 0.04 ng/ml, respectively. Adipophilin levels in D0 were 1.98 ± 0.12, in M1 2.83 ± 0.21, in M3 2.39 ± 0.17, in M6 2.57 ± 0.16, and in M12 3.25 ± 0.21 μg/ml. A simple method for detection of Czech bolting garlics (Allium sativum L.) using microsatellite analysis: methodology for practice

The aim of the methodology was to verify and compile a set of microsatellite markers for the purpose of verification to be used for the undisputed identification of selected types of Czech bolting garlics. The object of submitted methodology is to simplify procedure that allow to determine Czech bolting garlic using microsatellite analysis (SSR- Single Sequence Repeats), which is applicable in testing laboratories. The described method allows to identify the variety and breeding material of bolting garlic of Czech origin using DNA markers (microsatellite alleles) that are present only for this group of varieties and breeding lines. By the amplification of the genome containing the microsatellite locus by polymerase chain reaction (PCR) using specific, fluorescently labeled primers, and subsequent analysis of the lengths of PCR products. Methodology newly brings a unique set of diagnostic SSR markers, describe the evaluation and interpretation of the results.

Detection of five garlic viruses using real- time PCR analysis: methodology for practice

The objective of the assay is detection of five viruses in garlic using real- time PCR analysis. . This assay allows to identify five of the viruses in the varieties of garlic - Onion yellow dwarf virus (OYDV), Leek yellow stripe virus (LYSV), Garlic common latent virus (GCLV), Shallot latent virus (SLV) and Garlic mite-borne filamentous virus (GMbFV) based on the detection of gene sequences for the coat protein by SYBR Green real-time PCR. This method was shown in our work to be a suitable tool for the detection of highly variable pathogens, such as garlic viruses. The method brings a new procedure for detection of these viruses.

The sampling methodology for GM plants unintentionally present in the environment

In this methodology, sampling of plants from agricultural crops(corn, soybean, canola and potato) is defined to assess the presence of unauthorized GM plants in the environment.The methodology is applied mainly in the field of inspection controls of the compliance with Act no.78/2004. The sampling and collected samples must represent to a maximum extent the crops of the controlled part of the field (land parcel or land parcel portion). The basic applications of the methodology include:(a) low portion of unauthorized GM plants(seed contaminated with GM plants unauthorized for cultivation), (b) monitoring of the presence of unauthorized GM plants on land parcel, where there was such an occurence recorded in previous years (for example, plants growing on the field from seeds or tubers from the so called "soil´s seed bank").

The object of the methodology is the process characterizing the autenticity of varieties of onion put through microsatellite analysis (SSR- Single Sequence Repeats) for the characterization of varieties of garlic into the practice. This method allows to identify varieties of onion and characterize genetic resources using DNA markers and determine their genetic similarity based on the length variability of short repeating sequences (microsatellites) in the characterization of varieties of onion, Allium cepa L. The principle of the method is amplification of the genome containing the microsatelite locus by polymerase chain reaction (PCR) using specific primers and subsequent analysis of the lengtht of the products and the identification of the DNA profile to the declared variety.

The nursing process in a child patient with portal hypertension

My work called "Nursing process at a child suffered from portal hypertension" have been devided into two basic parts. The first part is theoretical, the second one is practical. In the first half of my work I deal with anatomy and physiology of liver, gallbladder and gallbladder ways. In the second part I mention characteristics of the disease, a clinical picture of portal hypertension, a general assessement leading to diagnosis of this disease. In the first part I have mentioned basic information about the disease, course of hospitalization, investigative methods and therapy survey. In the practical part I use the investigative process model, which goes out from the "Functional health" by Majora Gordon. Thanks to gained infromation from the medical documentation, from the patient, patient's mother and other medical stuff I analyzed all problems and set out actual and potential nursing diagnosis before and after the surgical intervention. I describe realization and evaluation of nursing interventions. In the end of my work I delt with the patient's education and also with patient's mother'education. The aim of the education was to advice and to adhere to medical and nursing instructions, to hand over sufficient of information about consequences and answering all questions, that have been in my competence....
