Analysis of Organic Markers for Identification of Sources of Atmospheric Aerosols
Křůmal, Kamil ; Chýlková, Jaromíra (referee) ; Vávrová, Milada (referee) ; Smolík,, Jiří (referee) ; Večeřa, Zbyněk (advisor)
In this work the organic markers that serve for identification of sources of aerosols are monitored. Theoretic part deals with detailed survey of organic markers emitted from the most significant sources of atmospheric aerosols (biomass combustion, combustion of fossil fuels and traffic) as well as with sampling of aerosols and analysis of organic markers by analytical techniques. Monosaccharide anhydrides (emissions from biomass combustion) and polyaromatic hydrocarbons (emissions from traffic and incomplete combustion) were observed especially. Hopanes and steranes (traffic, coal combustion) and fatty acids (cooking) were next groups of monitored organic markers. Markers were studied mainly in size fraction PM1 because this fraction of aerosols is the most harmful to human health. Aerosols were sampled in two seasons (winter and summer) in two towns in 2009. Higher concentrations of aerosols and organic compounds were found in winter season, which resulted from increased combustion of biomass, coal and other organic material while traffic was the most significant source of aerosols in summer.

Water from 25 forest springs was characterized for basic physical parameters and chemical and bacteriological composition. Chemical composition was mostly within limits for drinking water, but bacteriological results indicate faecal pollution of water in majority studied springs.
Organic compounds and markers from wood combustion in boilers used for household heating
Křůmal, Kamil ; Mikuška, Pavel ; Horák, J. ; Jaroch, M. ; Hopan, F. ; Kuboňová, L.
Organic compounds (polycyclic aromatic hydrocarbons) and organic markers (monosaccharide anhydrides, diterpenoids) used for the identification of aerosol emission sources were measured in the emissions from the combustion of hard and soft wood. Wood was combusted in different boilers used for the residential heating in the Czech Republic, i.e., overfire boiler, boiler with down-draft combustion, gasification boiler and automatic boiler. Emission factors of particles and all measured organic compounds were the highest from the combustion of fuels in the oldest (overfire) boiler.
Organic compounds and markers from coal combustion in different types of boilers used for household heating
Křůmal, Kamil ; Mikuška, Pavel ; Horák, J. ; Hopan, F.
Organic compounds (polycyclic aromatic hydrocarbons) and organic markers (hopanes) used for the identification of aerosol emission sources were measured in emissions from the combustion of hard and brown coal. Coal was combusted in different\nboilers used for residential heating in the Czech Republic, i.e., overfire boiler, boiler with down-draft combustion and automatic boiler.
Polycyclic aromatic hydrocarbons in PM1 aerosol in 6 localities in the Czech Republic
Křůmal, Kamil ; Mikuška, Pavel
PAHs were measured in winter seasons in cities/towns Ostrava-Radvanice (industrial city), Brno, Mladá Boleslav, Kladno-Švermov, Čelákovice, Košetice (background town). Lifetime lung cancer risks of PAHs were calculated from concentrations of PAHs in PM1 aerosol.
Determination of fast changes in concentrations of volatile organic compounds using diffusion denuder
Křůmal, Kamil ; Mikuška, Pavel
Determination of fast changes in concentrations of volatile organic compounds was performed using wet effluent diffusion denuder, which allows collection of samples in 2-min intervals.

We analyzed gene expression changes in the lungs and the immunological response in splenocytes of mice exposed by inhalation of ZnO nanoparticles - NP. Adult female ICR mice were treated for three days and three months, respectively. Analysis of differential expression in genes involved in oxidative stress was conducted using quantitative RT-PCR. The potential immunotoxic and immunomodulatory effects of ZnO NP were analyzed by phenotyping and cytokine production by splenocytes after three months exposure. Three days exposure resulted in down-regulation of GCLC, GSR, HMOX-1, NQO-1, NF-kB2, PTGS2 and TXNRD1 mRNA expression, three months exposure increased the expression of these genes. Three months exposure caused a significant decrease in the percentage of granulocytes in the spleen cells, and affected the production of IL-10 and IL-6 by lipopolysaccharide-stimulated leukocytes. In summary, our study revealed changes in the expression of genes involved in the oxidative stress response following acute ZnO NP exposure. Subchronic ZnO NP exposure induced immunomodulatory effects in the spleen.
Chemical composition of aerosol particles in emissions from coal combustion in different boilers used for residential heating
Křůmal, Kamil ; Mikuška, Pavel ; Horák, J. ; Hopan, F.
Organic compounds (polycyclic aromatic hydrocarbons, alkanes) and organic markers (hopanes) used for the identification of aerosol emission sources were measured in emissions from the combustion of hard and brown coal. Coal was combusted in different boilers (overfire boiler, boiler with down-draft combustion and automatic boiler) used for residential heating in the Czech Republic.

Adult mice were continuously exposed to metal oxide nanoparticles for different time intervals. At the end of experiment, mice organs (lung, brain, kidney, liver and spleen) were collected for chemical, biochemical, histological and electron microscopical analyses.
