Clostridium difficile - Importance and current options of laboratory diagnostics

This thesis mainly describes the question of Clostridium difficile diagnostic service. The first part briefly describes the characteristic of this bacterium. Further, the paper outlines common properties of the bacterium, the causes and manifestations of the diseases that it implicates and current opportunities of the laboratory service. The practical part of bachelors work deals with examination of toxin detection C.difficile in stool specimens, which was in case of positive result completed by cultivation. The cultivation and following Antimicrobial Susceptibility Testing (AST) is not routine task carried out in every clinical laboratory. PCR ribotyping is relatively new method that is provided rarely on the present. Generally, 195 patients have been tested in laboratory of Department of Medical Microbiology in Motol for Clostridium difficile toxin inherency in stool. Overall 30 samples have been tested as positive. The positive samples have been cultivated on special soil just for Clostridium difficile to acquire a clean culture. Certified methodic provides in depth information on cultivation technology, processing and use of maral root in the conditions of the Czech Republic. Both new research findings and practical recommendation for production of biologically active compounds are included in the publication. The methodic also describes utilization of the plant for the production of a biopesticide.
Production of cannabis in contidions of Czech Republic

The title of this bachelor work is Production of cannabis in contidions of the Czech Republic. There is describing growing of hemp, hemp production and hemp goods in the Czech Republic. Hemp production was reducing until 2008 in the Czech Republic but in Europe this plant is commonly used in industry. This Bachelor work proposes possibilities of using hemp in the industry. Work is focused of using industrial hemp in non-food and food industry. Bachelor thesis consists of two components - theoretical and practical parts. In theoretical part is describing industrial hemp - its cultivation, processing of hemp fiber, the use of hemp in the pharmaceutical industry, history of plant and hemp as a drug. In practical part is used analysis method that dealing production of industrial hemp especially in the Czech Republic.The information about hemp goods and producers I searched on the websites, from the professional books and some information I gained from experts whose I contacted. Second section is composed by survey that I handed around to anonyms. In this part I inquire the attitude to hemp of general public.
Factors influencing the quality of red wine

In my thesis, I focused on monitoring of microorganisms in the sample of red grape juice and on the interactions between yeasts, bacteria and filamentous fungi. Three different media were applied for the cultivation of microorganisms; firstly for monitoring of total volume of microorganisms, secondly for yeasts and third time for lactic acid bacteria. The indirect method was used for the determination of the amount of viable cells. This method consists in enumerating of visible macroscopic colonies grown up on agar plates. When the cells grew up, the forms of colonies were analyzed visually and the morphology of microorganisms was detected microscopically. The operating time of enzymes in grape juice in the production of red wine was monitored after application of commercial enzymatic preparation. The enzym action in grape juice was observed on the basis of the process of degradation of high – molecular substrate by enzymes through the use of Ubbelohd´s viscometer. The research findings provided a lot of knowledge about the occurance of microflora in the process of production of red wine. The commercial preparations added to grape juice played a significant role.
Infection of salmonelosis, their source and diagnosis.

This bachelor‘s thesis deals with infection of salmonelosis, it’s source, prevention and diagnosis. Theoretical part includes general information about Salmonella species and its morphology. It describes pathogenesis, source of infection, clinical symptoms, treatment and diet recommendations. It analyzes statistics of number of infection occurrences in Czech Republic. It also deals with prevention of infection origin on schools, population foreknowledge and climatic effects. Experimental part of this thesis contains description of optimization procedure of identification Enterobacteriaceae by adjusting of pH of prepared medium MIU.
The technology of cultivation and fertilization hemp (Cannabis) and ways of utilization

Contemporary is Hemp (Cannabis sativa L.) considered as a very promising plant whose parts can be used almost completely. Hemp is grown especially for fiber and seed. The plant is good biodegradable, so it causes no problems with waste disposal. The root system is deep and loosens the soil and prevents erosion. Hemp accumulate heavy metals from contaminated soils.To achieve the economic benefit are suitable fertile , deep soils with good nutrient supply and neutral or slightly alkaline reaction. Average seed yield in the Czech Republic is 0.6 t ha-1 and 6-7 t ha-1 stem. This study focuses on summarizing information about the cultivation, fertilization and the possibilities of using hemp.The first part is devoted to the origin, botanical characteristics, morphology and anatomy and principles of cultivation. The second part deals with the chemical composition, effective substances and use of cannabis in various industries. In this work was proposed five years crop rotation period for a variety Bialobrzeskie, grown for seed and stem. Than were selected pre-crops suitable for business without animal production and determined doses of nitrogen, phosphorous, potassium and magnesium fertilizers and suitable harvest mechanization.
Production efficiency of Miscanthus giganteus and possibilities of energy use

The thesis deals with renewable resources, biomass a its possible uses. The work is focused on Miscanthus sinensis. It described characteristics of plants, growing technology, harvesting and possible ways to use it - mainly burning. Attention is also focused on the yield potential of plants. The practical part of the thesis describes experiments with miscanthus at the experimental site in České Budějovice. There are reported experimental results and their evaluation and comparison with the literature.
Comparison of the effectivity of shovel cultivator and disc plough and their impact on the epigeic fauna

The diploma thesis is aimed at a comparison of tine and disc cultivators by several parameters which are most affecting the quality of a soil cultivation, and in the effect of the epigeic fauna, represented by ground beetles (Carabidae). Experiment was carried out twice during the season (after a harvest of wheat and rape) while soil was being cultivated with disc cultivator Lemken Rubin or tine cultivator Horsch Tiger. The experiment was carried out on the fields of ZD "Vysočina" Zbýšov and measured parameters were: plant residue decomposition, cultivation depth, diameter of aggregates, weed infestation and crop plant emergence, and abundance of ground beetles. Using the statistic tests, it was found, that almost every parameter describing the quality of the cultivation was influenced by the type of the cultivator. Tine cultivator had a higher decomposition of plant residues. This difference was more obvious when cultivating the field after rape harvest (P < 0.001) than field after wheat harvest (P < 0.01). Tine cultivator was more precise with keeping the cultivation depth as planned. Disc cultivator had a higher crumbling capacity. At the higher speed the machines were more inclining to come out of the ground at higher speeds (P < 0.05). A cultivation had a very negative influence on Carabidae population. On a rape stubble there was a 193 specimens caught in traps before the cultivation with two dominant species - Pseudoophonus rufipes and Pterostichus melanarius. Only 41 specimens was found in the traps after the cultivation.
Verification of commercial potential of the heat tolerant cyanobacteria Synechocystis sp. PCC6803 for biomass production in large volumes

The optimum culture conditions (culture temperature of 30°C, irradiance 100 µmol photons m-2.s-1) for potobioreactors up to 20 liters of working volume were specified. Verification of the growth potential heat tolerant cyanobacterium Synechocystis sp. PCC6803 was carried out in an open panel photobioreactor with the working volume of 140 liters.
