Optimization of N-glycopeptides analysis methods and their preliminary application to barley proteins study

N-glycosylation is the most frequently studied plant protein post-translational modification. The analysis of Nglycopeptides after protein proteolytic digestion offers information about the structure of both oligosaccharide and peptide moiety. However, this method has so far been less commonly used. Since glycopeptides hardly ionize during MS analysis in the presence of non-glycosylated peptides, they need to be separated from the complex peptide mixture. In this study, the glycopeptides enrichment, purification and analysis methods were successfully optimized on two standard N-glycoproteins. Concanavalin A (ConA) lectin tips were used for glycopeptide capturing, and obtained fractions were purified on carbon tips and analyzed using MALDI-TOF mass spectrometry. The differences in the CID fragmentation of certain types of glycopeptides were found. This technique was then applied to glycopeptide analysis of barley grain and malt proteins. Several barley glycopeptides were found, however, their identification was very difficult. More proteins separation techniques will be required before this enrichment procedure in further studies.
The efficient approaches for enzymatic digestion of plant proteins

In this work the fast and efficient methods applying ultrasound and infrared radiation were employed for in-gel digestion of proteins extracted from Hordeum vulgare. These techniques can be used instead of time consuming conventional protocol prior mass spectrometry analysis. The comparative study indicates that the examined protocols are able to enhance digestion efficiency and protein identification.
Proteomic evaluation of MCF-7 human breast cancer cells after treatment with retinoic acid isomers: Preliminary insights

The effects of 9-cis retinoic acid and all-trans retinoic acid in human MCF-7 breast cancer line have been investigated. The total cell proteins were extracted and separated on 1D SDS-PAGE. The proteins were subsequently in-gel digested by trypsin and identified by MALDI-TOF/TOF.
Sledování kvalitativních a kvantitativních změn proteomu ječmene na úrovni odrůdových rozdílů a změn v důsledku sladování

V této práci bylo analyzováno několik sladovnických odrůd ječmene jarního, které mají široké uplatnění ve sladovnictví a pivovarnictví a patří mezi doporučené odrůdy vhodné pro výrobu „Českého piva“. Relativní kvantifikace vybraných proteinů a jejich porovnání bylo realizováno pomocí kombinace isotopového značení peptidů (iTRAQ) a hmotnostní spektrometrie. Proteiny obilky ječmene byly separovány pomocí HPLC na koloně SEC-3 a vybrané identifikované proteinové frakce (beta-glukosidasa a beta-amylasa) byly následně použity pro isotopové značení. V obou případech lze sledovat značný nárůst zastoupení daného proteinu v důsledku sladování.
Changes of protein profile during the brewing process

Our work was focused on the protein identification in individual stages of brewing process. The greatest attention was paid to the proteins that resist the harsh conditions applied during brewing and therefore may influence various beer properties. These proteins (nsLTPs, protein Z and group of protease/alpha-amylase inhibitors) belong to the group of PRs.
Study on oligosaccharide composition of wort and beer samples by liquid chromatography/electrospray ionization tandem mass spectrometry

LC-MS technique was successfully applied to the description of oligosaccharides mixtures in wort and beer samples. Based on the important differences in MS/MS spectra the structural features of coeluted oligosaccharides were found.
Proteomická identifikace posttranslačních modifikací bílkovin v průběhu sladování

Proteomickou analýzou extraktu sladu byly identifikovány bílkoviny nespecifický lipid-transfer protein 1, nespecifický lipid-transfer protein 2 a protein Z (major endosperm albumin) a studovány jejich posttranslační modifikace.
