The development of microscopy techniques and their influence on the visualization of cell structures

The aim of this thesis is to review the literature on the topic of the visualization of objects in the realm of cell biology by means of various microscopy techniques (electron microscopy, confocal microscopy, etc.). It will describe the development of microscopy techniques during the 20th century and focus on discussions dealing with the objectivity of selected techniques and the trustworthiness of results gained by this means of observation.
Microscopy of leaves from different Bergenia species.

Plants of the genus Bergenia are part of remedies used in Ayurveda medicine. They also play an important part in traditional healing practice in China, India, Mongolia and Russia. Theoretical part of this thesis sums the newest findings and research results concerning three Bergenia species: Bergenia ciliata (Haw.) Sternb., Bergenia crassifolia (L.) Fritsch a Bergenia ornata Stein. The thesis mainly focuses on their current and potential use in medicine and pharmacy. Bergenia extract is traditionally used for dissolving kidney stones, treating respiratory tract illnesses and to stop bleeding. The most important active substances of these plants, their characteristics and main effects are also noted. Bergenia is an important source of arbutin and bergenin. Bergenin has antitussive, antiflogistic and gastroprotective effects. Arbutin is used to treat urinary tract diseases and in cosmetology to lighten the skin. The experimental part of this thesis includes methods of preparation of permanent microscope slides from leaves of chosen Bergenia species. Photographs have been taken from both permanent and native slides. Anatomy of the leaf and leaf epidermis is described including stomatal index. Microscopic characterization of flowers from culivars of Sambucus nigra L.

Sambucus Nigra belongs to the most favourite plants of our national folk medicine. It has important place in pharmacy and food industry. It is know in our environment mainly in wild form however increasing interest and demand for qualitative balanced drug contributed to cultivation of cultural varieties in plantations. Flower of Sambucus Nigra contains mostly flavonoids. For receiving of the most quality drug is necessary to follow the rules of harvesting and drying. Currently is drug obtained only just from the plants in wild nature. The goal of thesis was valorising of all characteristic features of flowers Sambacus Nigra and differentiation between cultural varieties and wild forms with using microscopic test of identity stated in Czech Pharmacopoeia 2009. Characteristic flower features of Albida, Alleső, Aurea, Bohatka, Dana, Haschberg, Juicy, Kőrsőr, Madona, Pregarten, Riese aus Vobloch, Tulbing and wild form of Sambuca Nigra were examined. The purpose of this work is created summary of laboratory methods, which are used for diagnosis of dermatophytosis. Beginning of thesis is given to common characteristics of dermatophytes, taxonomony, nomenclature and morphology of individual genera. The big part of thesis creates summary of laboratory methods. There are described individual types of microscopy, cultural techniques, molecular methods and immunological methods. The end of the thesis is dealted with infections, which are caused by dermatophytes and their treatment. Light microscopy and culture are used most frequently. Lithic function and its application in archeology

The goal of the thesis is to find the way how to apply use-wear analysis as well as functional analysis to archaeological assemblages of selected artefacts from Central European archaeological contexts, namely lithics (chipped stone artefacts) and obtain the greatest potential from the analysis. Thesis consists of: current state in the field of functional studies research worldwide, method of use-wear application in connection with material science knowledge, especially tribology. Use-wear analysis is applied to the selected lithic artefacts from Paleolithic to Eneolithic Periods. The results of this work are based on the following microscopic approaches: low power approach (LPA), high power approach (HPA), scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM). There are described and stated differences in potential of above mentioned approaches in connection with specific archaeological artefacts (assemblages of artefacts). The question answered in the conclusion is how to apply the above mentioned methodological approaches in application to various archaeological materials (period, number, context etc.) to obtain the greatest informational potential from the material analysed. Microscopy of Sambuci fructus from cultivars of Sambucus nigra Sambucus nigra L. is the important plant of the traditional folk medicine, its knowledge is accepted in the pharmaceutical and food industry. The elderberry fruits are used as diaphoretic, diuretic, during the migraine and neurological difficulties. On the grounds of the rising demand the collection of the wild-growing berries do not suffice and that is the reason for growing the elderberry and its cultivated varieties in plantations. The cultivated varieties of elderberry, which are grown for the use of the fruits in the food industry, could be exactly provided the quality drug for pharmaceutical use. Hana Engstová, Ph.D. Abstract: In this thesis the results are presented of a search for suitable PCFP markers for a visualization inner and outer mitochondrial membranes by unique biplane FPALM microscope. We participated in debugging the measuring software for the purpose of 3D vizualization mitochondrial nets and determine their parameters. For this purpose were developed two methods. The first one can be applied to determine the parametrs of outer membranes and the second one is applicable to determine parametrs of inner membranes. On smaller statistical file we managed to confirm dependence quality of mitochondrial net on the cell culture conditions. Experimentální studium ultrajemnozrnných slitin Ti pro využití v biomedicíně

In the present work the microstructure evolution of Ti-6Al-7Nb alloy prepared by high pressure torsion (HPT) and equal channel angular pressing (ECAP) was studied by scanning electron microscopy, microhardness measurements and electrical resistance measurements. Electron microscopy showed a bimodal structure of the alloy and deformed structure after HPT. Microhardness increased with the increasing number of turns of high pressure torsion. Electron back-scattered diffraction figured out that the grain misorientations are not random. Changes in the electrical resistance of the alloy prepared by ECAP showed irreversible process after heating above 450řC.

The theme of this diploma thesis is the design of a scanning electron microscope with a focused ion beam. Specifically, the thesis is focused on the design of the microscope covers and the adjacent workplace of the operator for Tescan Orsay Holding a.s.. Design is solved as the first proposal aimed at future innovation of the design of the entire product line.
