Biotyping of ascomycetous yeasts

In total, 84 yeast strains (originated from water, plants, fruits and soil) were selected for MALDI-TOF biotyping. All strains were cultivated on malt agar and YPD medium. Samples for biotyping were processed according to methods of Bruker Daltonik GmbH company, Institute of Chemistry of SAS and combination of these methods. Single strains were identified based on the analysis of intracellular ribozomal proteins using MALDI-TOF mass spectrometry. In case of ambiguous results, the DNA was isolated and the D1/D2 26S rRNA domain sequencing was performed. The strain identification was carried out by comparing its mass spectra with spectra of sequenced strains using MALDI Biotyper 3.0 software. The mutual similarity of strains was considered by score value, which was the result of the analysis. In total, 18 strains from 84 were previously sequenced and used as model strains for comparison with unknown isolates. Altogether 51 strains were definitely taxonomically categorized into 18 phylogenetic groups at the species level. The MALDI-TOF biotyping was repeated for overall 6 strains because of ambiguity of results. The taxonomic classification of 15 strains was not clearly determined and, therefore, these strains were suggested for D1/D2 26S rRNA domain sequencing. It was not possible to identify one strain, based on the results of sequencing, therefore, the DNA isolation was repeated. In the case of 8 strains, the results were identical with originally designed taxonomic classification. Conversely, the remaining 6 strains were identified as species. For 20 selected strains the basic characteristics were determined using microbiological methods. The shape of colonies growing on solid medium and appearance of cultures in liquid medium was assessed. Furthermore, the radial growth constant and the presence of urease were determined. Finally the microscopic observation of cells and the fermentation test for carbohydrate substrates were performed.

Security aspects of database systems implementation

The aim of this thesis is to provide a comprehensive overview of database systems security. Reader is introduced into the basis of information security and its development. Following chapter defines a concept of database system security using ISO/IEC 27000 Standard. The findings from this chapter form a complex list of requirements on database security. One chapter also deals with legal aspects of this domain. Second part of this thesis offers a comparison of four object-relational database systems - Oracle, IBM DB2, Microsoft SQL Server, and PostgreSQL. Comparative criteria are based upon the list of requirements on database security and reflect specific attributes of the specific type of the data model. The comparison aims at utilization possibilities of each database product as well as its limitations.

The relationship of Enterprise Architecture and strategic management

Enterprise Architecture is so far the highest development step in IT's architectural description of enterprises. At the beginning, just technological architecture existed. However, with the quickly growing complexity of IT other domains occurred, that had to be described. Information or application architectures can serve as examples. IT then realized, that it needed to add to its IT architectures a business view and consequently the discipline Enterprise Architecture came into being. Strategic management is crucial for the development of each enterprise. The thesis focuses on the strategic management of business and the strategic management of IT. The strategic management of business consists of a business strategy and an operating model. To the strategic management of IT then belongs an information strategy and IT governance. Business-IT alignment further explores and ensures the accord between the strategic management of business and the strategic management of IT. The aim of the thesis is to describe the relationship of Enterprise Architecture and the different forms of strategic management and to express to which rate Enterprise Architecture supports them. The explored relationships are: - The relationship of Enterprise Architecture and business strategy - The relationship of Enterprise Architecture and operating models - The relationship of Enterprise Architecture and information strategy - The relationship of Enterprise Architecture and IT governance - The relationship of Enterprise Architecture and business-IT alignment The goals of the thesis are reached via theoretical research and subsequent deduction. The author's personal asset consists of creating a hierarchical model of Enterprise Architecture, of defining Enterprise Architecture's reaction to business strategies, of modifying the methodology MMDIS in respect of information strategy, of depicting, how Enterprise Architecture can be used in IT governance, of illustrating, where Enterprise Architecture contributes to business-IT alignment and of assessing, to which rate Enterprise Architecture supports the different forms of strategic management.

Ongoing expansion of digital images requires new methods for sorting, browsing, and sear- ching through huge image databases. This is a domain of Content-Based Image Retrieval (CBIR) systems, which are database search engines for images. A user typically submit a query image or series of images and the CBIR system tries to find and to retrieve the most similar images from the database. Optimally, the retrieved images should not be sensitive to circumstances during their acquisition. Unfortunately, the appearance of natural objects and materials is highly illumination and viewpoint dependent. This work focuses on representation and retrieval of homogeneous images, called textu- res, under the circumstances with variable illumination and texture rotation. We propose a novel illumination invariant textural features based on Markovian modelling of spatial tex- ture relations. The texture is modelled by Causal Autoregressive Random field (CAR) or Gaussian Markov Random Field (GMRF) models, which allow a very efficient estimation of its parameters, without the demanding Monte Carlo minimisation. Water column separation under hydraulic turbine runner during unsteady operating regimes.

In this diploma thesis called Water column separation under the hydraulic turbine runner during unsteady operating regimes are solved the pressure pulsations of the reverse water hamer. In the thesis is deduced a mathematical relationship of elaboration the numerice model which is based on equations of continuity and equations of forces equilibrium. Numerical model is created in MS Excel uses for computation the numerical method Lax-Wendrof that allows consideration of variable sound speed as function of static pressure and allows variable lenght step in computation domain. Reverse water hammer is in the thesis solved with consideration of rotating flow behind shut-off valve, where we expect forming of vortex rope. This situation can be applied on the closing water turbine which has vertex rope under turbine runner. Specifically for this thesis was carried out the experiment of the reverse water hammer. Constants going into numerical solution are optimalized with using experiment and pressure pulsation are compared between numerical solution and experiment.

Nursing Care of the Patient after Transcatheter Aortic Valve Implantation

Resume This bachelor thesis is a case study of nursing care of 84-year-old patient hospitalised for aortic stenosis. The patient was treated in Cardiocenter of the University Hospital Královské Vinohrady in Prague using relatively new technique - transcatheter aortic valve implantation. In the first part of the thesis are given information from anatomy and physiology of the cardiovascular system, focusing on the heart and heart valves. In the section on pathophysiology are described cardiac valve diseases, with emphasis on aortic valve stenosis. Diagnosis and therapeutic procedures of the disease are described. The following are basic information about the patient and his health. Nursing part of the thesis describes the nursing process in accordance with the model Theory of basic nursing care by Virginia Henderson. According to this model nursing process for selected 84-year-old patient was compiled. This includes short and long term plan of care. Short-term plan of care is designed for the second day of hospitalisation, which is the day of the operation. Long-term care plan is set for a period of approximately 7 days. In the thesis is also mentioned patient 's social and psychology domain. The thesis deals with the education of selected patient, too.

Synthesis of model water-soluble azaphthalocyanine fluorescence quencher

1 Abstract Faculty of Pharmacy in Hradec Králové, Charles University in Prague Department of Pharmaceutical Chemistry and Drug Control Ljiljana Subara Synthesis of Model Water-Soluble Azaphthalocyanine Fluorescence Quencher Azaphthalocyanine (AzaPc) or tetrapyrazinoporphyrazine as they are often termed in literature, and its substituted derivatives have been investigated extensively for applications as photodynamic therapeutics, dyes, catalysts, liquid crystals, non-linear optical materials and as a red fluorophore. Quenching in general is a process that decreases the fluorescence of another compound. Azaphthalocyanine quenchers are synthetic dyes that decrease fluorescence by absorbing energy over a wide range of wavelengths to dissipate their absorbed energy as non fluorescence. However, in order to do so they must remain in close contact with the fluorescent compound. In this work, we describe the approaches used for the synthesis of azaphthalocyanines (AzaPc) and ways to increasing quenching properties while attaining water solubility. We synthesized a compound which had water solubility as a hydrochloride and no aggregation, however it lacked sufficient quenching properties, and remained difficult to purify on TLC. Mineralogy in the School curricula

This bachelor thesis is focused on teaching geology and mainly mineralogy on grammar schools. Questionnaire was made for purposes of this bachelor thesis. Objective of this questionnaire was to ascertain information about organization teaching geology and mineralogy on these types of schools and their inclusion in the school curricula. The questionnaire results showed that each school is very different in organization of teaching geology. Mineralogy is most often taught in first classes of four-year, or an equivalent multi-year study. Mineralogy is at some schools taught only in classes corresponding to an elementary school. Optional subject in which is the geology taught students may choose only to nearly forty percent of surveyed schools. Schools use a lot of sorts of material - textbooks for elementary and secondary schools, university textbooks, professional books and Internet resources. The most common tools used in the teaching of geology are collections of samples. Very frequent are also models of crystals and the geological maps. Fewer common are paintings, photographs and films with a geological theme. Research and development and valuable rights in Czech accounting legislation

The aim of the thesis is to create a manual for people who account for valuable rights and research and development in accordance with Czech legislation. The first part defines the terms intangible assets, fixed assets, research and development, valuable rights, capital improvements and goodwill. The second part specifies valuable rights, especially copyrights, patents, utility models, industrial designs, trademarks, know-how and domain names. This part is also focused on a valuation in owner's accounting, market valuation and valuation during business combinations, accounting for acquisition, amortization and decommissioning of valuable rights. The thesis mentions basic differences between accounting and tax legislation and between Czech and international accounting standards. The third part deals with research and development. Research and development is the driving force of the civilization and progress of whole society. It is evaluated most frequently by yield-based valuation methods, exceptionally by market or cost-based methods. It is supported by the state through grants, subsidies or tax reliefs. It is amortized by time or performance-based methods. The practical part deals with accounting for research and development and valuable rights in corporation when launching a new product.
