Differentiation of the stem cells, into the insulin producing beta-cells

Pancreaic stem cells are potent to differentiate into insulin producing -cells. Stem cells would be use for the cell therapy in the future. This diploma thesis is focused on this four transcription factors (LIF, noggin, TGF- a BMP-2) and their effects on the differentiation of the pancreatic stem cells into -cells. The results were analysed by evidential methods (RT-PCR, immunofluorescence and static incubation.
Growth of human adipose-derived stem cells on Ti-6Al-4V alloy with various surface modifications

Adipose-derived stem cells (ASCs) are of a great importance for bone tissue engineering. We compared ASCs obtained by liposuction under two pressures (-200 mmHg and -700 mmHg). The number and proliferation activity of cells isolated under -700 mmHg (ASC-H) were higher than in cells isolated under -200 mmHg (ASC-L). However, the ASC-L was more active in osteogenic differentiation, as manifested by a higher intensity of fluorescence of alkaline phosphatase and osteocalcin in these cells. The adhesion and growth of ASCs were then studied on Ti-6Al-4V samples either unmodified (K, roughness parameter R-a = 280 nm) or modified by shot blasting and tarnishing A, R-a = 200 nm), vibratory finishing (B, R-a = 100 nm) and vibratory finishing, shot blasting and polishing (C, R-a = 80 nm). All modified samples were more wettable than the K samples. On day 1 after seeding, the size of cell spreading area on some modified samples was lower than on K samples, namely on B samples in ASC-L, on B and C in ASC-H and on A in control MG-63 cells. On day 3, the cell number on some modified samples became higher than on K samples, namely on C samples (ASC-L), on B (ASC-H) and on B and C s (MG-63 cells). On day 7, the numbers of ASC-H and MG-63 cells on all modified samples evened out and became significantly higher than on K samples. On ASC-L samples, the highest cell numbers were obtained on A samples. Thus, all studied modifications of Ti-6Al-4V enhanced growth of ASCs and human osteoblast-like MG-63 cells.
Simulation of adhesion and transmigration imune-cells through capillary wall

Teoretická část této práce obsahuje biofyzikální popis endoteliální vrstvy a transmigrace buněk přes tuto vrstvu. Dále popisuje charakteristiky všech důležitých komponent pro vytvoření modelu cévy in vitro (endoteliální buňky, kmenové buňky, leukocyty, Calcein AM, PKH26). Praktická část práce je věnována sestavení in-vitro modelů dvojrozměrných a trojrozměrných endoteliálních vrstev, jejich zobrazení pomoci mikroskopu a testování interakce leukocytů a kmenových buněk s těmito endoteliálními vrstvami. V závěru je navrhnut nejlepší tvar a velikost mikrofluidních systémů simulujících kapiláry.

Cord blood - influence of cryopreservation and demographic data of mother and child on engraftment for the transplantation

The Master´s thesis gives an overview of cord blood, its use, processing and cryopreservation. The role of cord blood as an alternative resource for transplantation is being widely discussed. In the experimental part, 50 grafts of donor cord blood were processed and the effect of cryopreservation and demographic factors of mother and child were evaluated on the cellularity of cord blood. Based on the evaluation, a recommendation about the inclusion of additional tests in routine processing of cord blood were made.

MR tracking of transplanted stem cells labeled by iron-oxide nanoparticles

Our study demonstrates that grafted MSC and ESC migrate into an injured brain region. The commercially available contrast agent Endorem, based on iron-oxide nanoparticles, can be used a marker for long-term noninvasive MR tracking of implanted stem cells.
Stem cells and therapy of brain and spinal cord injury

Our studies demonstrate the potential of SC as a therapeutic tool in the treatment of injury and degenerative diseases, explain the effects of certain drugs on neurogenesis and regeneration and show that there may be various ways in which SC may interact with the host CNS tissue.
