Molecular mechanisms of formation and development of colonies of wild Saccharomyces cerevisiae strains

Yeasts are capable of forming a wide range of multicellular communities, which enable the survival in harmful and changing environment. Surface associated biofilms, often connected with infections in human body, and colonies can serve as an example of such populations. This work investigates formation and development of complex structured colonies of Saccharomyces cerevisiae, which can be considered as a distinctive feature of yeast strains isolated from the wild. Architecture and properties of such colonies are fundamentally different from the spatially undifferentiated colonies of most of laboratory strains and resemble in many ways rather natural biofilms of pathogenic yeasts. Yeast populations use specific developmental processes induced by communication mechanisms to synchronize the early stages of their development. Formation of specific three-dimensional colony architecture is enabled by the presence of extracellular matrix and adhesive protein Flo11p which provide stability and integrity of the whole structure. Protection of the colonies is accomplished by spatially differentiated cell subpopulations using various mechanisms such as expression of efflux pumps capable of removing toxic substances or production of extracellular matrix functioning also as selectively permeable barrier. Monitoring and analysis of individual periods of a business cycle dealing with relevant macroeconomical indicators in the Czech Republic

My bachelor work describes some empirical aspects of the problems of a business cycle. I am focused on monitoring, description and analysis of individual periods of a business cycle. Dealing with it I use some relevant macroeconomical indicators, which can have a role of detectors of cyclical changes in the economy of a particular country. First I describe and analyse the reasons of fluctuation of economical activity in form of supply and demand shocks. The fundamental indicator is a real production, approximative GDP, the rate of inflation, nominal interest rate and the deficit of a state budget. To show the fluctulation of economical activities more deeply I chose the Czech Republic from the year 1993. I am specially focused on the period when the Czech Republic was preparing for the entrance into EU. There is also descibed the context of stimulation demand of economics and its effect on former process of a business cycle.

Czech music journalism focused on rock music during the period of normalization

The thesis "Czech music journalism focused on rock music during the period of normalization" reflects the position and importance of Czech rock music journalism within the structure and development of normalization of the media system. It is brought back through the oral historical research based on authentic evidence of their protagonists. It defines the initial prerequisites (ideological, political and socio-cultural) which are important to understand the circumstances under which rock music could be created and developed into an institution. It all depended on the confrontational attitude of communist power towards rock music generally. The next methodic element how to fulfill the defined goals is descriptive and comparative perspective. On this basis there is confronted official and samizdat's area of magazine production on the example of chosen periodicals. There also belong other qualitative aspects of music journalism (personalization, professionalization and every day editorial job). At the end the accepted conclusions are supported by the analysis and interpretation of chosen cultural and music events. They are divided by the method of their media coverage from official and samizdat periodicals.

Characteristics of artistic expression in patients suffering from psychiatric illness

The bachelor thesis is about the development and characteristics in theart work of five people with psychotic illness in the period of three years. The cases were collected in the psychiatric ward at the hospital in Tábor. The thesis consists of theoretical and practical parts. The theoretical part collects information about psychotic illnesses and a description of the therapeutic approach to the mentally ill. It also describes the visual artwork characteristics of psychotic individuals, the work with them and the efforts necessary from the art therapists' side. The practical part describes each patient. Observing the individual stories, approachesused in each case, their artwork, changes and progress in their development. There is a description of approaches used in the acute stage of the disease and after stabilization, the patients features in the artwork and the arttherapists influence.

Complex evaluation of the best available techniques in selected company with pig farming and evaluation their economic impacts.

Thesis deals with complex evaluation of BAT techniques and their economic impacts in agricultural company Lihotošť which makes business in fattening pigs. Breeding of pigs is as well as the stud of other agricultural animals a producer of waste. In the European Union of production must be restricted gas emissions to a minimum. This range using the best available techniques (BAT), which help to reduce ammonia and greenhouse gases in the stable environment, landfill sites and in the application of slurry. The first part focuses on familiarizing with BAT techniques for pigs breeding. The next part deals with techniques applied in stud of pigs and their evaluation according to BREF. The third part is concentrated on evaluation of economic impact of applied BAT techniqueson monitored operation.

Application of sustainable practices of tourism development in hotel industry

This thesis concentrates on sustainable development of tourism and application of these sustainable practices in hotel industry. The aim of this thesis is analysis of concrete practices of the biggest hotel groups in the world and outline appropriate trend in this field. The theoretical part of this thesis concentrates on the definition of sustainable development of tourism as well as on its history. Furthermore, the thesis focuses on certification of sustainable development in hotels. In the analytical part there are already analyzed concrete practices and approaches of the biggest hotel groups sustainability reports. The practices focus on energy efficiency, waste production, water conservation and production of carbon emissions. At the end of this analytical part there is sum up of these concrete practices and approaches and outline appropriate trend.

Determination of sibiromycin by UPLC method.

Sibiromycin is produced by actinomycetes of Streptosporangium sibiricum and structurally belongs to family of pyrrolo-1,4-benzodiazepines. Sibiromycin is characterized by antibacterial and especially antitumor activity but due to its proved cardiotoxicity it cannot be used. The following research of possible therapeutic sibiromycin utilization is concerned on new nontoxic derivatives produced by genetic manipulated strains of S. sibiricum. For this purpose a new routine chromatographic UPLC-UV method with analyte preconcentration was developed and partially validated. A fermentation broth was extracted by solid phase extraction (SPE) on OASIS MCX columns with the sorbent based on a cation-exchange. Eluted extract was evaporated, reconstituted in methanol and loaded onto UPLC BEH C18 column and analyzed under gradient mode with mobile phases of methanol (A) and trifluoroacetic acid (B). Detection limit of the method was determined as 40 ng/ml, the recovery was 74.75 % and its reproducibility expressed as RSD was 5.18 %. The method was applied to comparison of sibiromycin production by S. sibiricum on 13 different fermentation media. Case study - biogas plant Úpice

In this work, a full-scale biogas plant (electrical installed capacity of 150 kW) located in Úpice near the city of Trutnov was monitored and assessed. The input materials of this plant are sewage sludge and different types of organic wastes. The samples of solid digestate were regularly taken between December 2008 and December 2009 and they were analysed for the content of toxic substances. Data concerning the amounts of particular input materials and the production of biogas were obtained directly from the operator of the plant. In order to reveal, if there is a relationship between the amount of particular input material and the production of biogas, a multiple linear regression was used. Finally, an overall assessment of the biogas plant was also implemented. The results of chemical analyses showed the digestate fulfills the norms given by the Czech legislation and thus can be certified and put on the market. Potentially problematic substances that should be monitored in the future are zinc and PAHs. A multiple linear regression identified "a gut fillings" and "silage" as the most suitable substrates for the biogas production in this plant. However, the value of r2 coefficient was quite low (r2 ~ 0,73), so we cannot say there is a clear relationship between the amounts of particular input...

Waste and Container Management Analysis at Selected Area

The goal of my bachelor´s work was the analysis of economical use of waste products and packing in the chosen area from the standpoint of both environment, economy and logistics. For my work I have chosen the town of Vodňany where refuse collection is provided by the company RUMPODL 01 {--} Vodňany L.t.d. The charge for the system working of gathering, collection, transport, sorting out, use and removing of the waste products was determined by the town of Vodňany to 480 Czech crowns. This charge was calculated from the real costs. If people sorted out more, this charge would fall. It follows from my questionnaire that most inhabitants of this town sort out only sometimes and that´s why the amount of waste increases. The amount of sorted waste also increases but not in sufficiency. People sort out mostly the plastics.
