Development and validation of HPLC method for determination of impurity 2-ethylpyridine in topical gel containing dimetindene maleate

Title of graduation theses: Development and validation of HPLC method for determination of impurity 2-ethylpyridine in topical gel containing dimetindene maleate Petra Hudcová Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Analytical Chemistry A High Performance Liquid Chromatography method for the simultaneous determination of 2-ethylpyridine and dimetindene maleate in Fenistil® gel was developed and validated. Analytical column Supelco Discovery® Cyano (100 x 4 mm; 5 μm) and mobile phase composed of acetonitrile:ammonium secondary phosphate buffer 50 mM, pH 5.5, with addition of 0.5 ml of triethylamine in 100 ml buffer, 40:60 (v/v) were used for the chromatography. The flow-rate was 0.7 mL/min and the UV detection was accomplished at 258 nm. Diltiazem hydrochloride was used as internal standard. Substances were eluted in the following order: 2-ethylpyridine, diltiazem hydrochloride, dimetindene maleate. Validation parameters precision, accuracy, linearity, robustness, selectivity, limit of detection and limit of quantication were calculated. The system suitability test data (number of theoretical plates, asymmetry, resolution and repeatability of area and retention time) were tested.

Application of chromatographic methods for the separation of metabolites of cholesterol

Lucie Tetrevová Title: The application of chromatographic methods for separtion metabolites of cholesterol Language: czech The proposed diploma thesis deals with the development and validation of analytical method for free and esterified cholesterol separation from blood serum and adrenal tissue samples. This method is neccessary due to the different metabolism of free and esterified cholesterol in the body and it enables the research of these particular pathways using stable isotope tracers. The solid phase extraction method was chosen, optimised and validated with following results: precision values 2.14% for serum cholesterol ester, 6.98% for tissue cholesterol esters, 2.91% for serum free cholesterol, 7.48% for tissue free cholesterol, limit of detection 1 mol/l and linear range 1-25 mmol/l were found. The stability of the derivative was tested for 30 days at temperatures 4 C and -25 C. The derivative was found to be stable for the whole time period in both temperatures. Laboratory temperature was not tested due to a high volatility of the solvent used. The developed method was used in experimental studies evaluating the influence of dietary cholesterol and septic shock to cholesterol synthesis rate.

Statistical processing of the Hepatitis C virus genotype analysis results. Material and methods: In the period of 1 July 2005 - 30 June 2012, the Hepatitis C virus genotype analysis was performed in respect of 2 185 samples at the division of molecular biological diagnostics of the serology laboratory of the Institute of Clinical Biochemistry and Laboratory Diagnostics of the General Faculty Hospital and the First Faculty of Medicine of Charles University in Prague, using the reverse hybridization method. The testing results were statistically processed. Results: Of the set of 2 185 samples, 1 372 (62,8 %) originated from males and 813 (37,2 %) from females, with the average age of patients included in the set being 36,7 years. The majority of requests for the HCV genotype analysis originated from specialists in Prague (91,0 %). Chiral Separation of Biologically Important Compounds by High-Performance Separation Methods Using Macrocyclic Antibiotics Mgr. Marie Honetschlägerová-Vadinská Abstract The aim of this thesis was to study retention and enantioseparation behavior of selected biologically active compounds by high performance liquid chromatography and capillary liquid chromatography on chiral stationary phases based on glycopeptide antibiotics. The retention and enantioseleparation behavior of selected profen non-steroidal anti- inflammatory drugs (NSAIDs) on a teicoplanin aglycone (TAG) CSP in cLC under various mobile phase compositions was studied. As the chiral capillaries are not commercially available, three capillary columns, packed with TAG stationary phase using the same procedure, were tested to evaluate the repeatability and reproducibility. The cLC results for a set of profen NSAIDs were compared with the HPLC results obtained on the TAG CSPs under the identical separation conditions. The evaluation of the enantioselective performance of two commercially available teicoplanin (T) CSPs with different chiral selector (CS) coverage, i.e., Chirobiotic T and T2 was carried out for three groups of structurally diverse chiral compounds. Changes of rainfall-runoff regime in the Stropnice river-basin

This paper deals with the rainfall-runoff regime in the Stropnice river-basin and its development. The aim of this paper is to answer the question of‚were there any changes in the runoff regime during the period 1945 - 2009, and if so, were these changes conditioned by climate or were they caused by human interventions' (e. g. river network modification, changes of the land cover, drainage systems development etc.). The single-mass and double- mass curves were used as the primary methodology. The homogeneity of the time series of mean discharge, precipitation and air temperature were statistically tested using Mann-Whitney-Pettit test and Mann-Whitney test. The Mann-Whitney-Pettit test was performed using the programme AnClim (v5.012), which is freely available online, the Mann-Whitney test was performed using SPSS 15.0. Non-parametric Mann-Kendall test was used for the trend detection, performed in MULTMK/PARTMK by C. Libiseller and A. Grimvall, also freely available online. Some changes in the runoff regime were uncovered in the early 80s', when the runoff decreased, which also happened after 1988. During these periods almost no annual flood occured. Social services for people with HIV/AIDS

Worldwide statistics show an increasing number of people infected with HIV. Unfortunately, number of people with HIV is also increasing in the Czech Republic (CR). That's why I was in my thesis entitled Social services for persons with HIV / AIDS deeply devoted to care, which is in the Czech Republic provided to persons affected by this insidious disease. I focused on social services, which device provides them, how users are satisfied and whether they have constant contact with the family. The bachelor thesis is divided in two parts, namely the theoretical and practical part. The theoretical part is thematically divided into nine sub-chapters, which are further divided into sections. The first subsection deals with the current situation, therefore the definition of HIV / AIDS, the second chapter explains the entire history of HIV and in the third chapter I write about the transmission of HIV. In this section there are described different ways of transmission of the virus. In this part I try to describe to the reader what that HIV / AIDS is. In the fourth and fifth subchapter is referred clinical picture of the disease, how the disease manifests itself, which are the stages of the disease and especially epidemiology, when to readers are approximated data and statistics from the Czech Republic and also from abroad. In the following three sub-sections, which are: treatment, testing and prevention, is briefly described treating HIV / AIDS, what are the methods of testing and how you can defend against HIV. The final ninth chapter is devoted to the principal topic of my thesis, and that?s social services (pursuant to Act 108/2006 Sb.) that are provided to people with HIV / AIDS, where are they provided, how are they financed, etc. Last but not least is mentioned where can a person who feels threatened by the virus, reverse. The practical part of this work was conducted through qualitative research, secondary data analysis and content analysis data. The first objective was to monitor the overall range of social services in the Czech Republic. The second goal was also monitoring, but in this case it was monitoring the satisfaction of users of social services DS with services provided by that social center. Since DS is the only facility providing the service in the Czech Republic, the research was conducted exclusively in this device. The third aim of my thesis was to compare data and statistics HIV / AIDS in the Czech Republic and other European countries. For comparison, I chose autonomous Switzerland, which seemed the most appropriate for comparison. For the purposes of research, I have identified three research questions: What is the incidence and prevalence of HIV / AIDS in the Czech Republic 31 12th 2012? What is the situation of HIV / AIDS in Switzerland compared to the Czech Republic? Is satisfaction of respondents with social services sufficient or D9+-S users have concerns about social services? Even in 2011 didn?t the Czech Republic belong among risk countries with a high incidence and prevalence. In the past year (2012) increased prevalence and incidence and the Czech Republic got on the list of countries at risk. It is paradoxical that while in the Czech Republic, is only one device for people with HIV / AIDS, which provides social services and the number of infected increases every year, there is no need to expand the provision of social services. DS coverers enough the needs of people who need this service and is always trying to find the best and most optimal solution for both sides. The work could help to raise the profile of this serious health, economic and social problem and at the same time could serve as study material not only for students but also for the general public. Furthermore, it could be extended to the framework of the thesis.

Doxorubicin cytotoxicity and metabolism in MCF7 cell line

RNDr. Lenka Skálová, Ph.D. Title of a diploma work: Doxorubicin cytotoxicity and metabolism in MCF-7 cell line. Doxorubicin (DOX) is ranked among anthracycline chemoterapeutics, significantly participant in the treatment of solid tumors, inclusive breast carcinoma and haematologic malignancies. DOX is metabolized to 13-OH-doxorubicinol (DOX-OL) produced by carbonyl reductase 1 (CBR1). DOX-OL is lower antineoplastic, but more cardiotoxic compared to the parent drug. Aim of this work was research of reduction DOX and testing possible increasing cytotoxicity of DOX through inhibition his reductase. For experiments has been selected human breast cancer cell line MCF-7. Initially has been elicit optimal composition of cultural medium and optimalized process on sample preparation for HPLC. In the study of DOX metabolism in cytosol the MCF-7 cells act oracin (potentional chemoterapeutic) as significantly kompetition's inhibitor of DOX reductases. In cytotoxicity tests acted DOX more toxic in cells with bovine serum in medium, oracin was again more toxic in cells without bovine serum. Combination DOX with oracin...

Generating data using the LM Reverse-Miner

In past years, great attention has been paid to evolutionary algorithms and they have been utilized in wide range of industries including data mining field, which nowadays presents a highly demanded product for many commercial institutions. Both mentioned topics are combined in this work. Main thesis subject is testing of new Reverse-Miner module, which can generate data with hidden properties using evolutionary algorithms while using also other modules of LISp-Miner system, commonly used for the purposes of data mining. Main goal lies in generation of two databases by the module in such way so they would meet explicitly set requirements. Other goals are also set within the thesis in the form of understanding the domain necessary for subsequent modeling. The result of the practical part of the thesis is represented not only by two successfully generated databases, but also by description of steps, methods and techniques used. The common recommendations for data preparation by module Reverse-Miner are later summarized, based on experience with modeling. Previous thesis outputs are furthermore contemplating the conclusion of analysis of technical means used for generation and they also provide several suggestions for possible future extensions.

Dynamic web pages

Serving dynamic web pages raises higher load of web servers and associated technologies. This can to some extent eliminate setting up reverse proxy with cache in front of the web server. The primary goal of this thesis is to implement this technique via presently most popular web server -- Apache. These Apache's proxy features were at first well tested and described, later practically applied in real LAMP software bundle enviroment (Linux, Apache, PHP, MySQL).
