Využití NIR spektroskopie při kontrole falšování kozí syrovátky

The diploma thesis dealt with the theme "The use of NIR spectroscopy in the control of goat whey adulteration". The aim of this thesis was to verify the potential of NIR spectroscopy to check of devaluation goat whey with cow whey. In the theoretical part was briefly described the problems of food adulteration, some of the methods used in the control of food with focusing on the near-infrared spectroscopy technique and its application in the dairy industry. Further was described whey and its composition focusing on goat whey, whey processing and the possibilities of using whey in food industry. In the practical part were measured goat and cow whey and their binary mixtures at various concentrations. For the qualitative evaluated of NIR spectra was used discriminant analysis. The results showed that near-infrared spectroscopy that near-infrared spectroscopy is suitable for the authentication of goat whey.

The thesis deals with the issue of insurance fraud on motor vehicle insurance. Insurance fraud is a serious problem affecting not only insurers, but also the whole society. Insurance fraud is being committed in connection with insurance and participants are both natural and legal persons in order to elicit insurance. Insurers respond to new trends in insurance fraud specialist staff training, creation of specialized teams, investment in new technologies and finding new sources of information for the prevention, detection and disclosure of insurance fraud. In my work to illustrate some cases of insurance fraud, also the tools and options as insurance fraud to detect, prove and make arrangements for the future.
Inhibiční látky v mléce a způsoby jejich detekce

The bachelor work deals with occurrence of inhibitory substatnces in milk and thein detection. They pose a gray concern since they cause problems during the milk processing procedures as well as they may reprepresent possible health risk. There are described individual groups of the inhibitory substances in the milk in this thesis, identified possible sources of contamination and their possible effect. Further on there are discussed possible sources of contamination, including usage of medication, maximal residual limits and prevention of milk contamination in general. Last part deals with detection methods of the inhibitory substances categorized in variol ways.
PCR based assay for detection garden pea lec gene

The method was developed and verified by staff of National reference laboratory for GMO identification and DNA fingerprinting suited for governmental control laboratories and private laboratories that analyze food and feed derived from various plant matrices. Method can be applied for quantification of GM pea in a sample, when reference gene is used as a standard. The method describe internal garden pea specific gene, lektin in this case, detection using PCR and real-time PCR. The general principle of the assay relies on lektin specific sequence presence that represents a species specific gene of garden pea. In many matrices DNA is damaged so amplification of short stretches of DNA is used. After amplification of target exploiting newly developer species specific gene primers PCR products are separated by electrophoresis in agarose gel. Resulting band is visualized by UV light and its position is compare with size standard. Alternatively, real-time PCR and ABI platform in combination with SYBR® Green can be used. Reaction parameters are described and specificity of reaction was verified. LOD as well as LOQ was defined.

Methodics of molecular detection of Barley yellow dwarf virus in vectors by RT-PCR

Barley yellow dwarf is one of the most important viral diseases of cereals in the world. It can cause significant yield losses in most cereal species, including wheat, barley, rice, corn, oat and rye. In this methodology we provide assays for reverse transcription - polymerase chain reaction (RT-PCR) for sensitive detection of barley yellow dwarf virus directly in the vectors, in aphids. This way it is possible to evaluate infectivity of vectors and from that conclude subsequent measures. The methodology is meant to serve the employees of State Phytosanitary Administration.

Method of identification of Fusarium species causing head blight by PCR

1. Multiplex PCR reaction with reliable published primers, enabling an easy detection of selected Fusarium species in one reaction, without time consuming optimalization. 2. Method of micromycetes cultivation using medium covered with cellophane, that is very suitable for following DNA extraction (easier harvesting of mycelia, minimal risk of medium contamination).

Methods of detection of wheat and barley strain of Wheat dwarf virus in vector Psammotettix alienus by PCR-RFLP

Wheat dwarf (causal agent Wheat Dwarf Virus - WDV) is one of the most important cereal diseases in the Czech republic where it can cause huge economic losses in wheat, barley, triticale, rye as well as in oat. Psammotettix alienus is the only known vector of WDV while another way of transmission is unrecognized. This methodology describes a PCR assay for sensitive detection of Wheat Dwarf Virus in Psammotettix alienus, and RFLP assay for barley and wheat strains specific identification.

Technical Means of Drug Control in Traffic.

ABSTRACT The topic of the diploma thesis bears the title Possibilities of using technical means of narcotics checks in road traffic. The thesis endeavours to describe, compare and evaluate searching methods that are used in road checks for habit-forming drugs by public officials. The theoretical part is focused on the description of individual groups of habit-forming drugs and their effects on the human organism. Further, the theoretical part contains an overview of the most frequently used analytic methods. The practical part deals with the questions of detection aids used by the traffic police in the Czech Republic. Based on the methodology used, which included research in domestic and foreign sources and a structured interview, all the information obtained was thoroughly evaluated and it illustrates the substance of the topic. This diploma thesis is structured in such a way that enables its use by police units and other organisations that take part in fighting the use of habit-forming drugs. The thesis can be also a contribution for tuition as study material.
Methodology of detection Penicillium expansum, Monilia frutigena, Botrytis cinerea and Neofabraea alba using multiplex PCR

Multiplex PCR for rapid and reliable detection of species causing rotting apples: Penicillium expansum, Monilia fructigena, Botrytis cinerea and Neofabraea alba is described here. Reaction is optimized for using in a common molecular laboratory. Described method allows both detectino of the pathogens from pure mycological cultrures, and their detection directly from the rot apple tissues. Cultivation of the micromycethes on cellophane surface is also described. References for mycological determination literature are listed, and can be helpful for determination in case of negative results of PCR.
