Methylene-bis(phosphonate) derivatives for modification of biomolecules

Thirty compounds (from which 29 are new) were prepared with moderate or good yields. All compounds were fully characterized by NMR and MS. Four new crystal structures were obtained by RTG monocrystal analysis. Derivative containing 4-nitrobenzyl group in the side chain was used for study of bis(phosphonates) adsorption parametres to hydroxyapatite (with quantification by UV-Vis spectroscopy). Several derivatives containing characteristic functi- onal groups (-NH2, -COOH, -NCS) were prepared. These compound are suitable for further derivatization, mainly for conjugation with more complex structures through amide bond or thiourea bridge. Conjugates of building blocks with fluorescent probe (fluorescein and dansyl) were prepared for potencial optical imaging. Inhibiting properties of the inhibitory protein TbIF1 to \kur{T. brucei} F1-ATPase.

To map and define the minimal inhibiting sequence of the inhibitory peptide TbIF1 and to improve its inhibiting property, a series of the N- and C- terminal truncations of TbIF1 was created. The mutated recombinant TbIF1 his-tagged proteins were over-expressed and purified from the E.coli cells and their binding and inhibiting properties were tested using purified F1-ATPase complex.

The Analysis of the VAT gap

The dissertation thesis describes the methods of calculation of the VAT gap. VAT gap is the difference between the theoretical VAT liability in the whole economy and the accrued VAT receipts. It is expressed in relative terms to the theoretical VAT liability and it approximates the amount of the VAT evasion. The VAT gap in the Czech Republic in the years 2002 to 2010 ranges in average from 16 to 17 %. It is steadily increasing from 11 % to 26 % during the last 5 years of the relevant period. Furthermore, the thesis also analyses the determinants of the VAT gap in the EU member states in the years 2000 to 2006. The share of VAT on total tax revenues and the Tax quota were identified as main tax factors that reduce the VAT gap. On the other hand, the complexity of the VAT system, expressed by the number of VAT rates, the difference between them and the number of preliminary questions to the European Court of Justice, increases the VAT gap. Further significant factor influencing the VAT gap in the negative direction is the Corruption perception Index.

Functional analysis of invertebrate (Branchiostoma floridae) promoters in heterologous systems

Understanding the mechanisms of transcriptional regulation and the constraints that operate in gene promoter sequences is the key step in understanding the evolutionary conservation of transcriptional regulation. It is well known that regulatory regions with the same expression outputs do not have to share the sequence similarity. The most important elements in regulatory sequences are transcription factor binding sites and their position relocation does not usually influence the expression output. The least complex transcriptional regulation is characteristic for housekeeping genes. For their expression they require only basal core promoter elements (sometimes only CpG islands are sufficient) and general transcription factors, so they can be transcribed easily and immediately whenever they are needed. In this study we focused on transcriptional regulation of invertebrate amphioxus (Branchiostoma floridae) housekeeping genes in vertebrate systems. We prepared a set of constructs with amphioxus regulatory regions for testing their activity in different mammalian cell lines and a set of constructs with the same amphioxus regulatory regions for observing their spatial recognition in developing medaka fish embryo. Erotica in a puppet animated film

Eroticism and pornography in animated puppet film is a niche genre. Nevertheless, it deserves attention for its unique means of expression. The subject matter of this thesis is a reflection on the relationship between corporeality and the puppet in film. It also includes selected auteur independent films and their analysis, in which sexual behavior is the prevalent topic or which contain explicit erotic scenes. The puppet functions as a representation of human body; specifically, within the context of this thesis, of the body in erotic situations. The body of the puppet becomes a representation of our own body. With the emergence of new technologies, we can become a representation of the puppet, which in turn becomes a puppeteer, an animator. The principle of identification with the main protagonist is essential for interpretation of the role and function of the puppet in film. One of the key instruments of interpretation of the role of the puppet in film and understanding its function in film. The chosen theme is accompanied by definitions of key terms and a brief overview of legislation concerning pornographic animated film.

Further Determinants of Private Equity

This thesis examines determinants of two variables important in evaluating private equity investments. First of them is the price of companies acquired by private equity funds expressed as a multiple of a profit indicator and the impact of primary and secondary buyouts. The second variable researched is the internal rate of return, a popular tool to measure profitability of private equity funds. The internal rate of return is studied for different groups of private equity funds and is reflected in a post-crisis perspective. The first conclusion stemming from the research is that the profit multiples in secondary buyouts are estimated to be higher compared to primary buyouts. The second conclusion proves the underperformance of real estate funds and the outperformance of distressed debt, turnaround and secondaries funds.

Religous-political Aspects of Culture of Martyrdom in the Islamic Republic of Iran

The presented doctoral thesis examines character and significance of the culture of martyrdom (farhang-e shahadat) in contemporary Iranian society with a particular emphasis on delimitation religious-political dimension of this phenomenon and determination of causes and implications of relatively recent reinterpretation which has radically shifted this culture from the religious-spiritual sphere to the political-ideological and even revolutionary. On the one hand, the thesis deals with a complex view of mechanism and manners of the utilization of the Karbala paradigm as the instrument for formulation of politically (and otherwise) motivated aims by different spectres of power structures of the ruling establishment and diverse oppositional social movements and on the other hand it reflects its apolitical standpoint associated with a spiritual experience of believers, which proceeds from the popular religion. These aspects of the political and religious boundary of the culture of martyrdom are examined in detail and given in the context of current socio-political reality. Nurse cooperation in care for child with Down syndrome

Abstract We most often see a family as a social group characterised by interaction within itself. A family is linked through numerous relations and it is also affected by a child. This applies to any other family even a family with a disabled child. In the world, there are about five million people who are affected by Down syndrome. It is the most widespread of all forms of mental retardation. In the Czech Republic, approximately seventy children with Down syndrome are born each year. In this paper, I worked on ``Collaboration of nurses caring of children with Down syndrome{\crqq}. The diploma paper was to give comprehensive information on specialized care of children with Down syndrome, including information on diseases and problems that might occur in the phylogenesis and life of such a child. The purpose of the paper is also proper understanding these issues, linkages with nursing care, therapeutic and diagnostic procedures as well as but finding the right approach to a family, education of families, and communication with parents. As a result, the paper should provide information about the collaboration of nurses and parents in influencing health and development of a child with Down syndrome. The objectives of the paper to map the problem of collaboration of nurses and parents raising a child with Down syndrome and to map the impact of nurses{\crq} collaboration with parents on complex care of children with Down syndrome have been met. Quantitative research was conducted using a questionnaire for nurses in surgeries of paediatricians and general practitioners for adolescents in the South Bohemian and Pilsen the Region. The hypothesis that nurses educate the parents of children with Down syndrome in the field of nursing care has been refuted on the basis of evaluated questionnaires. For qualitative research the following question was specified: ``What is the role of nurses in complex nursing care of children of children with Down syndrome.{\crqq} The answer was assessed based on interviews with parents bringing up a child with Down syndrome. The role of a nurse is one of a care provider, educator, adviser and protector. The findings of this diploma paper could serve as feedback for self-reflection of nurses in paediatric surgeries and to improve collaboration between parents and nurses in complex care of children Down syndrome.

First part of the thesis compares available approaches on business planning, namely the traditional and newly emerged and proposes a Business Planning Approach Selection Framework that should help (newbie) entrepreneurs with orientation in the chaotic business planning world and choosing an approach suiting their specific needs. Conclusion of the theoretical part is that both the traditional and modern business plans have their role in todays business environment. While the old are great for external presentation and funds securing, the new enable to create more complex process for planning successful business from the original idea to and beyond their launch, building on customer feedback and gradual iterations. The practical part then builds on the theoretical research and aims to check feasibility of an actual business project, a multisided platform mediating sales of automotive financial products. It describes the model refinement and product creation process as well as eventually proves viability and feasibility of the whole business.
