Defibrillator

This bachelor thesis is devided to six thematic areas. At the beginning we deal with the anatomy of the heart, as it is especially important for a good understanding of cardiac muscle activity. In the following part I summarize the heart activity in terms of both biological and of electrical activity, that occurs during its operations. Furthermore, it is necessary to analyze the potentially dangerous conditions occurring during heart aktivity and their solutions with the help of electrical discharge and without it. The most important part of the whole work is a defibrillator itself. First, I summarize the theoretical knowledge and then I focus on the design of an external defibrillator, a device intended for use by professional medical personnel or by the general public. Finally, it is important to note warnings resuting from manipulating with electrical current.

The different extraction techniques using for HPLC evaluation of some drugs

In this study the reversed-phased HPLC method for determination of an atypical antipsychotic drug amisulpride in a rabbit plasma was developed. The amisulpride is a benzamide derivative, which antagonizes human dopamine D2/D3 receptors. The main clinical efficacy of this drug is the therapy of both negative and positive symptoms of schizophrenia and low propensity to produce extrapyramidal sight effects. A Separon SGX C18 (size of particles 7 μm) as a stationary phase, a mixture of an acetonitrile - 0,1M amonium acetate (pH 5,6) 40:60 (v/v) as a mobile phase and an ambroxol as an internal standard were used for the analysis. Before the determination a solid-phase extraction was made. The method is linear in concentrations between 200 and 800 ng/ml (y=0,0065x-0,0695, r2 =0,9930), selective and robust. The analysis time was 11 min. A teoretical part of this study is approached to methods of an extraction, which is a very important step in an analysis of a biological material.

A substantial section of this work in devoted to the research of several selected social care facilities in the South Bohemian town of Strakonice. More precisely two homes for the elderly and nursing home, in which there was carried out a qualitative research and quantitative research, which takes place at the individual facilities on a regular basis. Reality as a system phenomenon

The thesis deals with relations between the way we construct our reality and human existence in terms of its biological, psychic and social system consequences. It is also concerned with the way we construct our reality through communication. The theories discussed in this thesis can be generally described as constructivist or systemic. In the second part of the thesis we applied the prototype approach to a study of understanding of the term "reality" in laymen population. The research was two-phased. In the first phase we acquired statements on what is understood under the term "reality". We then categorized the statements. In the second phase the categories were rated by the subjects on a Likert-type scale. The outcome of our study are some content definitions of the term "reality". Key

Implementation of methods of critical thinking in teaching photosynthesis

This study discuss two phenomenones. The first phenomenon is photosynthesis; this process played a key role in the emergence of life and is necessary to maintain a various life on our Earth. Photosynthesis enables plants to use a bottomless supply of solar energy to produce organic substance and release oxygen. The second phenomenon is critical thinking; the ability to sort and evaluate new information and form "your" own opinion indispensable ability for modern human. In a theoretical part of the study is dealt with term critical thinking, teaching methods of critical thinking and photosynthesis of biological and didactic point of view. In a practical part of the study is dealt with compilation and verification materials for thematic unit of photosynthesis, which would lead to the development of critical thinking of high school pupils (students). Furthermore in the practical part is a questionnaire survey, which is evaluated existing using of methods of critical thinking at biology teachers at high schools and is analyzed their teaching methods to the topic of photosynthesis. In the study is dealt with processing the topic of photosynthesis in selected high school textbooks.

The influence of photoperiod on the egg production of phasmid \kur{Peruphasma schultei}.

The aim is to determine the importance of photoperiod for oviposition of Peruphasma schultei. The theoretical part covers common description of the reproduction and embryonic development of phasmid. Included the climatic factors affecting physiological action and brief description of the biological clock of insects. Experimental individuals were are kept in three different photoperiods with constant temperature of 25 °C. Control and collection of eggs ran at intervals of two hours. It was shown the influence of photoperiod, time and their interaction on the number of eggs and imposed the difference in putting across time.

Education trail along "Blažejovický potok" brook

This thesis deals with the characteristic of the Vysočina region and with a project of founding an educational natural trail "Povodím Blažejovického potoka". The thesis consists two parts. The first theoretic part describes the Vysočina region in light of geomorphological, biological, geological, natur protection and in which chapter is emphasization the area, in which is situated the lane of educational trail. It is compilatory text, which is composion like guide book to the Vysočina region for teachers, who decide for visit with their students not only this education trail, but another places in the Vysočina region too. They can find in the text another interesting places to visit. On the end of theoretic part are two chapters, historical interests and explication the concept of the education trail. The second, practical part include individual information boards with thematic content and their graphic layout. The education trail has 7 km with 12 information boards. It is the combination education trail, it has 1 introductory board, 3 boards with historical informations, 1 geological board, 2 botanical board, 1 zoological board, 2 ecological boards and 1 board about technical memory. The biological importance of CAS SH3 domain tyrosine phosphorylation

Protein CAS is a major tyrosine-phosphorylated protein in cells transformed by v-crk and v-src oncogenes. It is a multidomain adaptor protein, which serves as a scaffold for assembly of signalling complexes which are important for migration and invasiveness of Src-transformed cells. A novel phosphorylation site in N-terminal SH3 domain was identified - tyrosine 12 located on binding surface of CAS SH3 domain. To study biological importance of tyrosine 12 phosphorylation, non-phosphorylable (Y12F) and phosphomimicking ( Y12E) mutant of CAS were prepared. We found that phosphomimicking mutation Y12E leads to decreased interaction of CAS SH domain with kinase FAK a phosphatase PTP-PEST and also reduce tyrosine phosphorylation of FAK. Using GFP-tagged CAS protein, we show that Y12E mutation caused delocalization of CAS from focal adhesion but has no effect on localization of CAS to podosome-type adhesion. Non-phosphorylable mutation Y12F cause hyperphosphorylation of CAS substrate domain and decrease turnover of focal adhesion and associated cell migration of mouse embryonal fibroblasts (MEFs) independent to integrin singalling. Utilization of micropreparative isoelectric focusing in the analysis of biological samples

The aim of this Master Thesis is the optimization and verification of conditions of separation of proteins from biological materials. For the separation a developed technique called micropreparative isoelectric focusing is being used. The theoretical part describes analytical methods of biological materials with a focus on proteins which were used for the optimization of the developing system in general. This part of the Master Thesis also presents this method itself. Furthermore, basic characteristics of other techniques used to control the efficiency of the micropreparative isoelectric focusing are introduced. The experimental part depicts the individual steps of optimization of the micropreparative isoelectric focusing. The ideal separation procedure was firstly found out on the individual standard proteins, secondly on their mixture and lastly the optimised procedure was applied to a real biological sample. The efficiency of the micropreparative isoelectric focusing was verified by SDS-PAGE and MALDI-TOF mass spectrometry.
