Potentionalities and advantages of nonprofit organisations while finding solutions for emergency events

We meet different kinds of exposures with extraordinary crisis situations in today world. It is both from nature (earthquake, overflooding) and from people themselves ( escape of dangerous chemical substance, fire and so on). Even if we try to prevent these situations, we cannot exclude they will never happen again. In these situations there should be some state apparatus which represents the crisis management, Integrated rescue system- Rescue service, Fire department, and Police of the Czech Republic. Also the public service and another physical subjects concerned with this problems. It is up to all of us not to be reckless and to try to help. Because all of us can help not only as a person but also in some organization which we can use in these situations. There are many different public organization in the Czech Republic which are prepared to help people in crisis situations. But there is a big difference between them, mostly because of their externalization. What is common for all of them is that they are all for helping people. Some of them help people in crisis situations, some help the alcoholics, drug addicts, seniors, children. Each of us think about something different when we say crisis. The person is very various kind, who is adaptable even in very difficult situations. The question is, when he can manage all the problems alone and when all his energy is gone and he looks for help from another subjects, also from the organization. The extraordinary situations have happened in South Bohemia for ages. The effectual and quality help should be all-round and therefore it is necessary the help should be offered in cooperation of all local recources.

Sports - regulation centers and their significance for tourism

The main aim of the thesis is to analyse the operation of sport facilities and their influence on tourist trade in Ceske Budejovice. Situational analyses of South Bohemia Region, the Ceske Budejovice Region and the Ceske Budejovice Municipality are introduced in the thesis. In addition, there is a situational analysis of Budvar Arena, a selected sport facility. On the basis a field research questionnaire, spectators´ satisfaction with available services and attendance of the Arena were detected. Next to it, suggestions for improvement of current situation of Budvar Arena leading to a higher visit rate were laid down. The concluding statement of fact is that Budvar Arena has a positive influence on tourist trade development in Ceske Budejovice but there is still a possibility to attract more tourists into the area.

Burial-ground of Bylany Culture from Lovosice

The work is concerned with the archaeological finds from Lovosice, Ustecky kraj, CR obtained in 2002. The major part consists of description and drawing documentation of 15 well-equipped from the Early Iron Age. Movable artifacts are analyzed typologically and chronologically. The funeral rite is evaluated in the frame of spatially-functional relations between human relicts and burnt offerings. Immovable finds are analyzed with respect to the parameters of holes, construction and things. In the same way the graves are categorized into the finding groups. The funeral site is evaluated with regard to the form, structure, total range and chronological trend. The main goal consists in a definition of evolutionary horizons of the funeral site and their comparison between Czech and Central European chronological systems. These horizons, in the future, can make one of the main pillars for evaluating grave and settlement finds of Hallstatt age in NW and C Bohemia. The area of Lovosice necropolis is interpreted as a site of central relevancy and the dead as elite with significant influence over the cultural and economic development of the region. Nowadays, Lovosice represents one of the richest, most widely researched and therefore most important funeral site of Ha C1 - Ha D1 in Bohemia. Keywords Hallstatt...

Program education of citizens in the field of population, with special emphasis on disabled persóna

The aim of this thesis was to analyse the current state of upbringing and education in the field of human protection in emergency situations, with particular emphasis on persons with disabilities. For this purpose, there was implemented a questionnaire survey and there were created two questionnaires a questionnaire for teaching staff at special schools and a questionnaire for their directors in the South Bohemia region. The questionnaire for teaching staff comprised 37 questions and was organized into two thematic sections. The questionnaire was a combination of open and closed questions. The first seven questions were used to obtain the information needed to characterize the investigation file. The first block was focused on respondents' knowledge; questions were focused on the issues of the integrated rescue system, civil protection, first aid and questions of selected emergencies (floods, radiation accident, biological agents), then the knowledge of the recommended ways of behaviour in the development of selected incidents. The aim of the first block of the questionnaire was to find out the awareness of teachers. The second block was aimed at self-education in this field and the assessment of the importance of teaching these issues in schools. The replies of teachers of this thematic bloc served to complement the views, experiences and observations of respondents related to this issue. The questionnaire for directors contained 19 questions. The questionnaire was a combination of open and closed questions. The first 6 questions were used to obtain the information needed to characterize the investigation file. The second part of the questionnaire focused on issues that dealt with school directors' own opinions in the South Bohemia region. The results of the questionnaire were analysed using Excel from Microsoft Office software package. The main objective of this thesis was to determine the awareness of teachers. For good measure of awareness I set the limit of 75% correct answers. Results can be assessed as very startling. I learned that most of the teaching staff haven´t basic knowledge in the issues of personal safety in emergencies. For this reason, they would like to educate themselves in the form of organized trainings. Directors, who also think that their schools have enough teaching aids for both normal and disabled students, also showed this interest. Although these answers were disproved by the teaching staff, who claim the opposite. As a result, it can be understood that the present state of education in the field of human protection in emergency is very individual and depends only on the approach of teaching staff. There are no special teaching materials and uniform system that would unite all schools and provide them with the necessary tools. The thesis provides insight on teaching human protection in emergencies and the teaching staff's own opinions about it. My results can be used as a basis for further research investigation, because I think it would be appropriate to elaborate on some passages. Performance of work at special schools for the disabled entails a number mentally, but also physically demanding situations. These situations teachers and directors manage very well. In this area there are many schools, specifically aimed at one disability, but also those that work with various disabilities. School education programs and this teaching the topic of human protection in emergencies need to be adapted to the needs of children and students with special educational needs so as to achieve consistency between educational requirements and actual abilities of these pupils. Currently, schools use to teach a variety of assistive aids to address special educational needs of the pupils, but there are no didactical tools, special textbooks (study materials in Braille, sign language recordings) that would through its form and content reflect the needs of disabled persons.

Spliceosome assembly

Pre-mRNA splicing is a process in which introns are removed from eukaryotic transcripts and exons are ligated together. Splicing is catalyzed by spliceosome, a large ribonucleoprotein complex composed of five small nuclear RNAs and more than 100 additional proteins, which recognizes 5' splice site, branch point site and 3' splice site and performs two transesterification reactions to produce mRNA molecules. 5' splice site is recognized by U1 snRNP and U2 auxiliary factor (U2AF) is involved in branch point and 3' splice site recognition in the early splicing complex. There is some evidence of splice sites cooperation during intron recognition in vitro but little is known about the situation in vivo. Using Fluorescence resonance energy transfer (FRET) and RNA immunoprecipitation (RIP) methods, we have investigated the early stages of spliceosome assembly. We have employed splicing reporters based on -globin gene and MS2 stem loops to detect interactions of proteins on RNA molecule directly in the cell nucleus. Results of FRET indicate that intact 5' splice site is required for U2AF35 interaction with 3' splice site and that U1C recruitment to 5' splice site is partially limited upon 3' splice site mutation. Introduction of an Early Intervention System in Prague

The System of Timely Intervention (SVI) is new system of multidisciplinary cooperation of the social and legal protection of children and youth and crime prevention. SVI is a tool for faster and more effectively deal with these cases, allowing better communication, prompt, adequate and effective response to concerned authorities (police, Department of social-legal protection of children, courts, prosecution, Probation and Mediation Service, schools, health facilities and NGOs ). In my work I describe The System of Timely Intervention and implementation options in the Capital City of Prague. Present organizational structure of the Capital City of Prague (as a region), the main actors involved in the system, the principles of mutual cooperation, legislation and related concepts and basic pillars of the project SVI. On the basis of seven specific case studies, I attempted to describe the current cooperation between institutions in the socio-legal protection of children and potential risks of its introduction and throughout the region (The Capital City of Prague). This work summarizes the current situation in the care of vulnerable children and offer possible recommendations for more effective work within the Timely Intervention System.

Stuccoed ornamentation by Santino Cereghetti at Svatá Hora

The bachelor work deals with the work of Santino Cereghetti (about 1640 - 1791), stucco worker of Italian origin, in Svatá Hora. The first part deals with the technique concerning the stuccoed decoration. The second part concerns the Italian artists who came from the area surrounded by the lakes at the today's border between Italy and Switzerland and who worked as architects and stucco workers in the whole Europe. The third part of the bachelor work follows the changes in the stuccoed decoration and its form in Bohemia in the baroque time. The core of the bachelor work is in the fourth part in which the stuccoed work by Santino Cereghetti in Svatá Hora is described. The determination of the stuccoed decoration made by Cereghetti results from the contracts which prove his work and from the similarity of elements present in his form of the stuccoed decoration. Cereghetti's stuccoed decoration is embedded in the early baroque architecture. Changes in his stucco work are obvious. His first stuccoed decoration is of early baroque character. Foundations of Marie Margaret Countess of Waldstein

RESUMÉ MARIE MARKETA OF WALDSTEIN'S FOUNDATIONS This bachelor's work represents Marie Marketa, Countess of Waldstein, as the founder of sacral objects in her estates, and presents an overview of buildings with an explanation of their origin in the given locality. Bachelor's thesis is part of research on Waldstejn's patronage in northern Bohemia prepared for the exhibition of this important aristocratic family in the Museum of the Bohemian Paradise in Turnov. During my research, I did mapping of individual sites and I described them. From my point of view the work in the field was important, this means acquisition of photographs of individual churches and gathering additional information on the creation of buildings. Most reference period was the years 1724-1729, when the Countess of Waldstein was truly generous in promoting the arts, when the favorable financial situation help her. Thanks to this situation newly built or restored religious buildings on Waldstein's estates were built. Such as decanal church of St. James in Mnichovo Hradiště, St. Nicholas in Turnov, and St. Vojtěch in Kruh near Doksy, St. Wenceslas in Boseň and the chapel of St. John of Nepomuk at Waldstein, St. Stapin in Klokočka. The Pupil with Autism

The main subject for this diploma thesis is questions of integrating children with autism in mainstream basic schools. The aim of diploma thesis is to analyse current situation of integrated education of pupils with autism in mainstream schools in Central Bohemia Region. The analysis goes from the view of region coordinator for autism as a new institution responsible for coordination of counselling and educational services focused on clients with autism in region, and as a significant instrument for realization of the integration process. Partial aim of diploma thesis is focused on detection of factual experience of parents and assistant educators of integrated children, especially on disclosure of fundamental problems they often deal with in integration process. Diploma thesis would like to contribute to pedagogic practice in the field of integrating pupils with autism in mainstream schools by means of definition current problems to be solved in the future.
