Design of highway bridge

The subject of this bachelo r’s thesis is the design a new supporting bridge construction in Prešovský kraj, district Levoča the territory of Slovak Republic. This object is located on the highway D 26,5/120, at kilometer 13,499 795 and communication of third class – 131-00 bridges. The objective is the design of a perpendicular four-span construction, which is proposed in three variants. The first variant is a chamber girder with inclined walls from post-tensioned concrete height 2,480 m. The second variant is composed of two beamed cross-section and the third variant is composed of one beamed cross-section. For detailed assessment the first variant was chosen with a length of bridging of 166,000 m. The calculation of load effects is done by software Scia Engineer and compared with a manual calculation according to current standards. This bridge is designed according to limit states, construction´s influence on design is taking into consideration. Structural analysis, well arranged drawings and visualization are elaborated in this thesis too.

Stress ribbon bridge across the deep valley

The topic of this thesis is a design of stress ribbon bridge across the deep valley. The construction of the footbridge contains stress ribbon bridge with external cables and final prestressing tendons. The bridge deck is in a parabolic arc and the suspension cables are in two bend down parabolic arcs. The construction is reproduced for evaluation of longitudinal direction in the Ansys 14.0 programme and for transversal direction in the Scia Engineer 15.1 programme. The model was solved in several phases, nonlinerally. The pier was reproduced in the Scia Engineer 15.1 programme for local effect of action. The construction was evaluated for dynamic action. The construction is assessed according to the actual EN standards.

Grinding Box Unit for Vertical Boring and Turning Machine

This diploma thesis solves grinding box unit for vertical boring and turning machine for ČKD Blansko. For fixing is used standard fixing system used in ČKD Blansko. Spindle is drived by engine placed in slide ram. Engine can be also used for milling box unit. Grinding box unit must stand operating load. Using of automatic balancing system is also solved in this project.

BIOFUELS - REASONS AND CONSEQUENCES OF ITS INTRODUCTION

The thesis aims to analyze the economic and environmental consequences of the regulation to mandatorily blend bio-components into the fuel and prove its inefficiency. The theoretical part summarizes in detail the reasons of introducing the regulation, including the legislative framework, and generational evolution of biofuels. The analytical part uses statistical analysis of time series and price and efficiency indices to evaluate the effectiveness of biofuels in relation to reducing greenhouse gas emissions. The impact of the introduction of biofuels on the economy is specified through fuel prices. The impact on engines as well as food prices and social environment is also mentioned.

Deformation behaviour of gellan gum based artificial bone structures under simulated physiological conditions

The paper deals with investigation of deformation behaviour of gellan gum (GG) based\nstructures prepared for regenerative medicine purposes. Investigated material was synthesized as porous spongy-like scaffold reinforced by bioactive glass (BAG) nano-particles in different concentrations. Deformation behavior was obtained employing custom designed experimental setup. This device equipped with bioreactor chamber allows to test the delivered samples under simulated physiological conditions with controlled flow and temperature. Cylindrical samples were subjected to uniaxial quasi-static loading in tension and compression. Material properties of plain scaffold buffered by 50 wt% and 70 wt% BAG were derived from a set of tensile and compression tests. The results are represented in form of stress-strain curves calculated from the acquired force and displacement data.

Analysis of novel members of FAHFA lipid class derived from omega 3-PUFA by LC-MS/MS and LC-MS/MS/MS

White adipose tissue (WAT) is a complex organ with both metabolic and endocrine functions. Dysregulation of all of these functions of WAT, together with low-grade inflammation of the tissue in obesity, contributes to the development of insulin resistance and type 2 diabetes. Omega-3 polyunsaturated fatty acids (PUFA) of marine origin play an important role in resolution of inflammation and exert beneficial metabolic effects. Using LC-MS/MS method, we elucidated the structures of novel members of fatty acid esters of hydroxy fatty acids (FAHFA), which were present in murine and human serum and WAT after omega-3 PUFA supplementation in diet. These compounds contained DHA esterified to 9- and 13-hydroxyoctadecadienoic acid (HLA) termed 9-DHAHLA and 13-DHAHLA, and were synthesized in WAT. Our results document the existence of novel lipid mediators, which are involved in the beneficial anti-inflammatory effects attributed toomega-3 PUFA, in both miceandhumans.

Poly(methylene blue) modified carbon based electrodes for the determination of sulfhydryl-containing compounds

This research presents the study of electrochemical polymerization of phenothiazine derivative, methylene blue (MB). Electrochemical pro perties and analytical application of resulting electrocatalytically active poly(methylene blue) (pMB) have been investigated. Three various carbon-based substrates: basal plane of highly oriented pyro lytic graphite (HOPG), pencil graphite electrode (PGE) and carbon fibre micro electrode (CFE) have been used. Aforementioned electrodes modified with the polymer (HOPG/pMB, PGE/pMB and CFE/pMB) have been prepared by potential cycling in aqueous electrolyte solution containing dissolved monomeric methylene blue. Electrocatalytic activity of three developed electrode systems towards hydrogen sulphide has been compared. Developed electrodes have been further investigated as potential sensors for sulfhydryl-containing compounds.

Stress load in a specific job

The submitted thesis deals with the problematic of stress Fire Brigade, especially workplace emergency number 112, where calls are received and handled and further workplace "Spojař" where are the emergencies handled eventual exit units or passing other rescue units. The thesis also discussed the conditions for the reception of members on these specific staff positions. With regard to the topic of the theoretical part of the thesis will discuss the stress. The concept stress at work is theoretically defined, divided and classified. There is also defined psychological stress at work generally, its origin, species, its causes and consequences. The thesis also presents particular stressors emergency operator and employee workplace signalmen. Is described how operators respond to stress and how to cope with stress. The thesis also mentioned the burnout syndrome, its causes and prevention of this dangerous syndrome. In the empirical part of the research will be described using a questionnaire and the data will be evaluated using appropriate statistical methods.

Voltammetric determination of cancer biomarker 5-hydroxyindole-3-acetic acid at screen-printed carbon electrodes

Tumor biomarker 5-hydroxyindole-3-acetic acid (5-HIAA) is a breakdown product of serotonin; level of this neurotransmitter can predict carcinoid tumors. 5-HIAA has been determined at screen-printed carbpon electrodes in the iptimum medium found: Britton-Robinson buffer (0.04 mol l-1, pH=3.0). Dependence of the peak current on the concentration of 5-HIAA was linear in the concentration region from 0.1 to 100 micromol l-1, with the limit of qiantification of 0.2 micromol l-1.

Design of search engine for modern needs

In this work I argue that field of text search has focused mostly on long text documents, but there is a growing need for efficient short text search, which has different user expectations. Due to this reduced data set size requirements different algorithmic techniques become more computationally affordable. The focus of this work is on approximate and prefix search and purely text based ranking methods, which are needed due to lower precision of text statistics on short text. A basic prototype search engine has been created using the researched techniques. Its capabilities were demonstrated on example search scenarios and the implementation was compared to two other open source systems representing currently recommended approaches for short text search problem. The results show feasibility of the implemented prototype regarding both user expectations and performance. Several options of future direction of the system are proposed.
