Narrative strategies and the themes of Bildungsroman genre in Patrick McCabe's The Butcher Boy, Roddy Doyle's Paddy Clarke Ha Ha Ha, Seamus Deane's Reading in the Dark and Frank McCourt's Angela's Ashes

The approach and use of Bildungsroman in the context of Irish contemporary literature is subject to lively development and invention on the behalf of the writers, thus offering an interesting and wide field of study for the literary criticism. This study of four representative works serves only as an introduction into the subject and does not, by far, cover the whole area. Nevertheless, Patrick McCabe's The Butcher Boy, Roddy Doyle's Paddy Clarke Ha Ha Ha, Seamus Deane's Reading in the Dark and Frank McCourt's Angela's Ashes manage to cover and demonstrate the variety and richness of this genre in Irish literature. Each of the writers tackles the subject with distinctive innovation, each picking a different theme as the centre of their respective novels.

Micropreparative solution isoelectric focusing of peptides and proteins in nonwoven strip

Recently we devised a new instrument for micropreparative analysis [1]. It is based on solution phase isoelectric focusing (sIEF) performed in a narrow channel (approximately 2 – 4 mm wide) with a strip of nonwoven fabric serving as the analysis bed. The strip can be precut before an analysis to make harvesting of fractions easier. Isoelectric focusing is driven by a programmable electrophoretic power supply. Progress of analysis is monitored by addition of colored isoelectric point (pI) markers. Usually sIEF runs are performed overnight. Evaporation of water is one of crucial features of analysis and it forces free liquid to shrink into the nonwoven fabric strip. Moreover, water evaporation increases liquid viscosity by altering ethylene glycol/water ratio. Fractions can be harvested simply by collecting and washing precut segments after the sIEF run. In this paper we used sIEF device for purification of caseinomacropeptide (CMP) from crude whey. Obtained fractions were further analyzed by HPLC. From acquired data a plot with chromatograms of all fractions was constructed to show purification profile of CMP. We proved that the new instrument is capable of quantitative purification of CMP complex from a globulin fraction.

Specifics of Perioperative nursing Care of Seniors from the Point of View an Anaesthesia Nurse

The theoretical part of the thesis is focused on nursing care of seniors during perioperative period from the point of view of an anaesthesia nurse. It is divided into a general and a special part. The general part deals with nursing care approach to a senior, with psychosocial aspects of hospitalization, his/her needs, communication barriers and educational specifics. It also covers the issues of operation in old age, psychosomatic changes and their consequences for anaesthesia care. The theory in the specialized part is divided into preoperative, intraoperative and postoperative care in relation to anaesthesia administration.The first goal of the bachelor thesis was to examine the importance of anaesthesia department from senior?s view. The second goal was to determine the importance of anaesthesia department from the view of an anaesthesia nurse and the third goal was to map nursing care of a senior at an operation theatre in relation to anaesthesia.Qualitative research form, the questioning method was applied to information collection. Non-standardized interview with general nurses and seniors was the data collection technique. The research results show that an anaesthesia department is beneficial from seniors? view.It has also been found that nurses consider the anaesthesia department useful particularly for seniors. The benefit for the nurses themselves has still certain imperfections, particularly in sharing information on nursing care with an anaesthesia nurse. The research in nursing care at the operation theatre shows that anaesthesia nurses participate in the continuity of perioperative care of a senior just partially. They are aware the specifics of care about seniors in intraoperation period and differentiate satisfaction of the needs. The thesis might serve as educational material for nurses involved in perioperative care and nurses in specialization education. Documentation was elaborated from the conclusion of the empirical part upon the found drawbacks, particular ideas of the nurses and in compliance with the procedures recommended by the informed public. A brochure Anaesthesiology Guide was elaborated to make cooperation between a nurse and a senior easier.

Preparation of rat NK cell receptors using HEK293T expression system.

Natural killer cells play a significant role in the immune response against tumor and infected cells. NK cells express a wide variety of surface receptors, including NKRP1, a C-type lectin-like family of both activating and inhibitory receptors. Recently, ligands have been found for some of these previously orphan molecules, some of them lying within the same family. This is also the case of rat Clr-b as a cognitive ligand for rat NKRP1B. It has been shown that in rat, this inhibitory NKRP1B-Clr-b mutual receptor system is subverted by rat cytomegalovirus protein RCTL, a viral version of Clr-b, which serves as a decoy ligand for NK cells. The aim of my diploma thesis was cloning and production of the above mentioned C-type lectin-like proteins based on transient transfection of HEK293T cell line in a suspension culture. This expression system allows us not only to obtain proteins of our interest with a satisfactory yield but also in their native conformation, removing the need for time consuming and often fruitless refolding procedures required in case of using the E. coli expression system. Success was achieved in case of Clr-b and NKRP1B receptors from both WAG and SD strains. Monuments of the Jewish Settlement in Heřmanův Městec

In my thesis I´m dealing with preserved Jewish relics in Heřmanův Městec, the Chrudim region. First of all I just briefly mention the organization of Jewish community in Czech territory, then I examine and search Jewish settlement in Heřmanův Městec from its beginnings until today. Among still standing sights I name especially the synagogue, which was many times destructed. Thanks to the extensive reconstruction it could again come into service, but nowdays it only serves as a cultural tabernacle. Next preserved relic is the building of Jewish school located at close quarters of the synagogue and it´s much newer. Last but not the least I mention the old cemetery, which belongs to the oldest and the largest ones in the Czech Republic. This Jewish dominant feature of Heřmanův Městec is nowdays under the administration of the Jewish Community in Prague.

Ptinciples of semiotic analysis of comics

The first part of the thesis is concerned with the theory of comics. The author considers the concept of graphic space of comics as a basic notion of the presented theory. On the basis of graphic space it is possible to introduce the individual concepts of the comics theory, namely comics sign, comics word, comics image. The author introduces the new concept of "prototypic" comics scheme which serves as the basis for comparative analysis of an old Czech comics Rychlé šípy with modern adult comics. The concept of comics image is viewed by some theoretical approaches of pictorial semiotics (Eco, Goodman, Snesson), and Peirce's general semiotics, and film semiotics. The comics image is defined as rhytmical plural multidimensional comics structure. The thesis is concerned with its various structural dimensions and their units. In the second part, some of the theoretical concepts introduced in the first part are applied in the comparative analysis, which focuses on some devices used in a famous old Czech comics Rychlé šípy by J. Foglar, and in modern adult comics (the corpus of the chosen Czech and worldwide comics is included in the present work). Optimalisation of insulin treatment - continuous subcutaneous insulin infusion

Introduction: To be able to prove the efficiency and benefit of the treatment including its safety on a wide range of patients (as opposed to more or less strictly selected groups form clinical studies), we started to gather data about patients in the Czech register of patients treated with continuous subcutaneous insulin infusion (CSII) - further referred to as Register. The obtained data not only represent an effective feedback on our clinical practice and treatment, but also they serve as an important argument for promotion of the CSII efficacy during the negotiations of reimbursement from the health insurance system (the CSII treatment is significantly more expensive compared to other MDI regimens). Aim of study: The aim of the first study were to present data on current situation and treatment results of CSII on wide unselected population of patients with diabetes mellitus in the Czech Republic. In the second study patients were evaluated to compare treatment indication, efficacy and safety with specific regard to the type of diabetes. Patient and methods: The national register of patients treated with CSII exists in the Czech Republic since 1998. Architectural expression The primary intention is to shape the breaks on a street corner and Koliště horáková and close existing data center housing estates. Two wings of the same width based on boch sides of existing units meet in a breakthrough right at the corner of the intersection. The ground plan is thus to portray one just wide strip folded over the corner area. This philosophy precisely determines the shape of the central mass, which has become a triangle. The center of mass exceeds the side wings and the center at the entrance of the visually deprived of sight at the corner. Two lateral mass height strictly respect the street axis and also high-rise buildings. These materials are broken into large cubic volumes. Each of these volumes contains specific function. Large volumes of each other horizontally poposúvané a team already out of the system suggest the division of functions hidden within. Systémm shifted masses are broken side wings while the middle remains subtle and clearly dominant input into their interior. Cutting out the dice and cubical fragments from different parts of the functional volumes is fed daylight into the interior, which in this way preserve anonymity outside and intimacy within. LAYOUT Different shape volumes are adapted to different functions, which are filled. Different and folding them together are made points of indoor as well as outdoor spaces. Placement areas with the same functions a volume is created by their uncluttered layout. At the same time, this solution provides the possibility of various large bright floor heights for different functions. Galerija spaces in selected volumes divided into levels with greater ground clearance, unlike the administration, Com, workshops and other areas that have a generous amount required. On the ground floor there are areas of trade and lecture hall. Top Galerija spaces are two dimensional objemysituované in the middle of the building. Their prepjenie is provided through the central hall. It serves as a connecting element between them creates the way a visitor from one gallery space to another. All rooms offer light and spatial variability. The second floor is an outdoor exhibition space. Serves mainly for the installation of three-dimensional sculptures and also links, gallery spaces with creative workshops and ateliers. Also serves as a rest area, ktrý visual linking the pedestrian zone alley Captain Jaroša. In the second part of the gallery space is vútorná gallery spanning over two floors. The top floor is a cafe overlooking the city center.

Application of the alternative and augmentative forms of communication by children suffering the Infantil cerebral palsy in České Budějovice

The theoretical part of this work contains the definition of the Infantile cerebral palsy and its associated handicaps, then it contains a short explanation of the term ?communication? as well as a summary of the most widely used and actual methods and instruments for the alternative and augmentative communication. The first aim of this Bachelor work is to get an overview about the methods and systems of the alternative and augmentative communication in the area of České Budějovice used by the communication training and by the communication itself with the children who suffer the Infantil cerebral palsy. The second aim of this work is to find out, what´s the attitude to these methods from the view of these childrens´ parents, who are using such methods by their own children. For gaining the required data, a qualitative research has been carried out, in concrete ?The methods of observation and interview? in the form of a structured interview with opened questions. The interviews were realized both with the concrete parents of these handicaped children, who communicate with their children by using the methods of alternative and augmentative communication ? and also with the specialized staff who work with these handicaped children. The interviews were realized in the ARPIDA, o.s. center and in the Světluška, o.p.s. center in České Budějovice. From this research it has emerged, that from all these alternative and augmentative methods - suitable for the communication training with these handicaped children - the interviewed specified staff from České Budějovice uses mostly the method ?The Picture Exchange Communication System? and as a supporting method ?Communication tables?. The parents, who decided to use the methods of alternative and augmentative communication, find these methods mostly positive. This work could serve as an informational material for parents, specialized staff and for applicants who are interested in systems of alternative and augmentative communication.
