Microcalorimetric study of bacterial degradation of oil and its products

This bachelor thesis deals with a study of the oil degradation as one of the possible products of petroleum using bacterial stems Pseudomonas Putida a Pseudomonas Chlororaphis. The work is motivated by the research of usability of a method which has not been used for those purposes so far – calorimetric analysis. Using IČ spektrometry (ATR), calorimetry and UV-VIS spectrometry was studied a degree and a character of interactions. Calorimetric analysis showed that the Pseudomonas Putida has bigger abbility of the degradability than Pseudomonas Chlororaphis. There was also detected that the optimal concentration for more effective degradation of oil fuel is 5 g/l and 20 g/l. By measurement of the absorbance deppending on time was verified that the bigger is the concentration of the sample, the higher is the resulted absorbance. Using UV-VIS analysis was determined a quantity of biomass. Using IČ spectometry in the ATR mode (reduced reflectance) was detected structure of the sample and also verified that the bacterial cultures Pseudomonas Putida and Chlororaphis degradated entire amount of petroleum.
Study of hyaluronan-albumin systems using light scattering methods and calorimetry

This thesis, which is a continuation of the previous Bachelor thesis, is dedicated to the study of polyelectrolyte complexes between hyaluronan (HA) and bovine serum albumin (BSA). Interactions between HA and BSA and a formation of complexes were studied by SEC-MALLS method, where a molar mass, a root mean square (rms) radius, a hydronamic radius and an intrinsic viscosity of particles of the system were defined. Furthermore the interactions were studied by calorimetric measurement ITC, where thermodynamic character of complex formation was determined. Additionally, dynamic and electrophoretic light scattering methods (DLS and ELS) were applied, where hydrodynamic radius and a value of the zeta potential were defined. Also the effect of higher temperature on the character and complex formation was examined by SEC-MALLS and ITC measurements. The interactions were confirmed at any of the used media, but with different efficiency. It was also proved that complexes become smaller in their radii and viscosity with growing BSA concentration. Furthermore it was proved, that the high ionic strength hinders surface charges of HA and BSA molecules and minimizes their mutual interactions. At the higher ionic strength the radii of the complexes, their intrinsic viscosity and zeta potential are increased. The higher temperature has only minimal effect on the formation of the complexes.
OPTIMIZATION OF TECHNOLOGICAL PROPERTIES OF CEMENT SAND MIXTURES

The work deals with optimization of the technological properties of cement-sand mixtures. The main objective is to find the correct mixture of the individual components which contribute to the rapid solidification of the mixture, hence shorter dismantling. The content is also of a method of measuring and recording during the plasticity of the mixture. Also assessment of the velocity of hydration heat cement mixture in the initial stage of the solidification process in the course of the strength properties of the mixture. To assess these phenomena was used calorimetric method used for measuring hydration heat cement mixture. Further measurements plasticity mixtures and mechanical and physical methods for measuring the compressive strength with the help of a measuring instrument LRU-D.
Mechanical Properties of Microstructural Components of Inorganic Materials

Disertační práce se zabývá studiem strukturních a mechanických vlastností anorganických materiálů. Cílem je nalezení jednotlivých fází ve zkoumaném materiálu a hlavně lokalizace (mechanicky) nejslabšího místa, jeho ovlivnění a následně výroba materiálu o lepších mechanických vlastnostech. Z důvodu velkého množství použitých metod je základní teorie vložena vždy na začátku příslušné kapitoly. Taktéž z důvodu značného množství výsledků jsou na konci kapitol uvedeny dílčí závěry. Práce je rozdělena na tři části, kdy první se zabývá seznámením s možnostmi modelování mikro-mechanických vlastností a provedením experimentů umožňujících posouzení rozsahu platnosti některého modelu. V druhé části je provedeno shrnutí současných možností indentačních zkoušek pro měření mechanických vlastností strukturních složek betonu a praktické zvládnutí metodiky vhodné k užití pro výzkum materiálů zkoumaných domovským pracovištěm. V třetí části je navržena metoda identifikace nejslabších článků struktury anorganických pojiv a její ověření na konkrétním materiálu zkoumaném na domovském pracovišti. V této dizertační práci jsou použity tyto metody: kalorimetrie, ultrazvukové testování, jednoosá pevnost v tlaku, nanoindentace, korelativní mikroskopie a rastrovací elektronová mikroskopie s energiově disperzním spektrometrem. Dílčími výsledky jsou kompletní charakterizace cementových materiálů, upřesnění stávajících poznatků a nalezení optimálního postupu pro charakterizaci. Hlavním výsledkem je inovativní přístup vedoucí k pozitivnímu ovlivnění materiálu.
Development and design of a modular battery box.

Tato práce se zabývá problematikou battery packů. V první části popisuje návrh battery packu z hlediska volby baterií, jejich tvaru, velikosti, typu a konfigurace zapojení. Dále uvádí vliv těchto charakteristik na výkonnost battery packu a jeho spolehlivost. Zabývá se problematikou termálního managementu battery packů a jeho vlivu na životní cyklus baterií. V druhé části se zabývá praktickým návrhem konceptu battery packu pro Lithium-polymerové baterie. Kalorimetricky je zde zjištěno množství produkovaného tepla při vybíjení, které je poté využito v CFD simulaci chlazení článku.
Higher Heating Value of Sewage Sludge

The thesis is focused on problems of thermal treatment of sludge from wastewater treatment plants. The main objective is to establish a formula for calculating higher heating value (HHV) and lower heating value (LHV) of sludge based on its chemical analysis. The work is divided into two main parts: The theoretical part deals with the characteristics of sludge, thermal treatment of sludge, it isn´t omitted higher heating value and lower heating value. The main topic of a practical part is formulation of eguations for HHV and LHV calculation. Chemical analysis of sludge samples were supplied by TÜV NORD Czech s.r.o., together with the values of HHV and LHV. The calculations were verified by the program MINITAB.
Synthesis, properties and product of hydration of strontium aluminate cement

This diploma thesis deals with non-traditional binder, which is strontium aluminate, with his preparation and hydration. The behaviour of binary and ternary mixtures of strontium-calcium-barium aluminates was also studied. These aluminates were prepared by firing an equimolar mixture of aluminum oxide and the appropriate carbonate. Samples prepared this way, including their mixtures, were characterized by analytical methods such as X-ray diffraction analysis (XRD), thermal analysis with evolved gas analysis (TG-DTA and EGA), scanning electron microscopy (SEM), infrared spectroscopy (IR) and calorimetry, with the greatest importance for the study of the hydration process.
Hyaluronan hydrogels for medical applications

This work studies properties of hydrogels prepared by the interaction of polyelectrolyte hyaluronate with cationic surfactant cetyltrimethylammonium bromide (CTAB) in a model physiological solution of 0,15 M NaCl. Effects of different pH and final pH of different samples of hydrogels were studied. Furthermore, stability of hydrogels was observed by fluorescence spectroscopy, where pyrene was used as a fluorescence probe, and isothermal microcalorimetry. It was found that low temperature disrupts gel integrity and it is not recommended to expose hydrogels to other than a neutral pH.
The effect of boron and its compounds on setting time of portland cement

The aim of this work is to study the influence of boron and its compounds, boric acid and borax, on hydratation of portland cement. The first part describes chemical composition of portland cement and reactivity of its components. The second part deals with hydratation of portland cement. The teoretical part ends with description of retardation of portland cement using gypsum or other sulphate options. The experimental part describes preparation of samples – cement paste – modified by addition of boron, boric acid and borax in diferent concentrations. Results are obtained by evaluation of calorimetric curves, mechanical properties and phase composition. The goal is to describe kinetics of targeted slowed down process of hydratation and to find such addition which would suspend setting of boron modified system entirely. Comparison of effects of each boron-containing compound is based on its concentration and on its kind.
