SNP polymorphisms of Y chromosome in the population of african fulani people

Markers on the non-recombining region of chromosome Y is a useful tool for study of diversity between populations. SNPs are the most commom polymorphisms in human genome. Mutation rate of SNPs is very low and so they may be used as genetic markers in evolutionary and population studies. We have analyzed 205 unrelated men from 11 Sub-Saharan Fulani's subpopulations. Fulani are an ethnic group of people spread over many countries, mainly in West Africa. Our samples are from Tindangou area, Banfora area (Burkina Faso), Bongor area, Linia area (Chad), Diafarabé area (Mali), Tcheboua area (Cameroon), Banfora area, Diffa area, Zinder area, Ader area and Abalak area (Niger). Using kit Signet Y-SNP Identification Systems and Luminex instrument with LabMAP Luminex Technology we detected particular Y chromosome's SNPs. LabMAP Luminex Technology is universal array platform, which as a probe using fluorescent polystyrene microspheres. We have observed 12 different haplogroups. Haplogroup E, which is typical African haplogroups, is determined with derivated allele in polymorfism M96. We have detected haplogroup E in maximum of 89,3% in the Fulani's subpopulations. Improvement of retroviral vectors for efficient gene transfer

44 CONCLUSIONS Retroviral vectors are transcriptionally unstable in mammalian cells. The ASLV- derived vectors are the most affected by silencing. We modified the vector by insertion of the CpG island inner element (IE) into the vector LTR in three different positions in either sense or antisense orientation. Each vector variant exhibited a certain extent of stabilization. The vector with insertion of a tandem of two IEs between the enhancer and the promoter was the most stable and exhibited almost no silencing after four months of cultivation in rodent and human cells. The IE comprises two high-affinity Sp1 binding sites. The presence of Sp1 binding sites is important for the protective effect of IE, but at least a part of the entire anti-silencing capacity is maintained in IE with mutated Sp1 sites. We identified the Tvc receptor of ASLV. The tvc gene encodes a 488-amino-acid protein most closely related to mammalian butyrophilins, members of the immunoglobulin protein family. To confirm the identification of the Tvc receptor, we disrupted both tvc alleles in normally susceptible DT40 cells. The DT40 tvc-/- clone was resistant to the ASLV(C) infection. Back pain prevention nurses.

The thesis: Backache Prevention in General Nurses, examines whether general nurses practice backache occurrence prevention. The following goals were set for the quantitative research: Goal 1: To find out whether general nurses are sufficiently informed on backache prevention in performance of their job. Goal 2: To find out whether nurses have an opportunity to participate in preventive programmes focused on backache prevention in the organizations they work for. The following hypotheses were set: Hypothesis 1: Backache, as a symptom of overload is a frequent health problem among general nurses. Hypothesis 2: Nurses practice backache prevention. Hypothesis 3: Nurses regularly keep fit. Hypothesis 4: Nurses that attend seminars or training sessions aimed at backache prevention are sufficiently informed on these problems. Hypothesis 5: Organizations where nurses work provide them with some preventive programme aimed at backache prevention. The method of anonymous questionnaire focused on general nurses working in a hospital was used for confirmation of the hypotheses. After assessment of the quantitative research results we discovered, that the first hypothesis in the quantitative research was confirmed. Hypotheses 2 and 3 were not confirmed by the research. Hypothesis 4 was conformed, nevertheless not many nurses participate in seminars or courses aimed at this issue. Hypothesis 5 was confirmed with the comment that an employer should do more for health of nurses. The research has shown that general nurses are actually aware of how to prevent backache occurrence, however not many of them use that. Neither the employer contributes remarkably to prevention of the fact that healthcare is the profession where backache occurs most frequently of all. I am convinced that thorough training of new healthcare staff should be sufficient for remarkable reduction of backache occurrence. A brochure that will be the output of this thesis may serve this purpose, as well as continuous training by a qualified physiotherapist or a physician together with provision of an all-hospital seminar.

The influence of nursing documentation audit on nursing care quality

Abstrakt The keeping of nursing documentation is an inseparable part of the work of a nurse. It is an instrument of professional practice, which is intended to support the process of care. The quality of the keeping of the nursing documentation is a reflection of the level of professional practice. The nursing documentation is an invaluable manner of supporting communication within the framework of the health care team and between the health workers and their patients/clients. Well kept nursing documentation is thus the product of good teamwork and at the same time an important instrument for increasing the quality of care. An audit is one of the parts of the management process and its final aim is to incease the quality of the provided care. An audit can play an important role during the delivery of the quality of the provided care and this is also true for the process of the keeping of the nursing documentation. During an audit of the kept documentation areas are identified where it is intended to ensure the improvement and education of the staff. The theoretical part the work also includes chapters related to the given problems that is the nursing aim, the nursing process, the nursing documentation, the quality of nursing care and the nursing audit. In the research part I deal with the influence of the audit of the nursing documentation on the quality of the provided nursing care. The aim of my research is to ascertain the influence of the standard of the nursing documentation on the quality of nursing care and to ascertain whether the results of the audit of the nursing documentation help the nurses in the wards to improve the quality of the provided nursing care. During the research my first hypothesis that the quality of the nursing care is increased by the carrying out of a standard ``Unified keeping of nursing documentation`` was confirmed and 26 of the questionned nurses (57%) agreed with it. The second hypothesis, that the quality of the nursing care is increased by the introduction of an audit of the nursing documentation into practice, was not confirmed. Only 18 of the interviewed nurses (39%) think that the introduced audit of the nursing documentation into practice is an increase in the quality of the nursing care. In order to obtain the required data to accomplish the aims and answer the hypothesis of quantitative research I used a method of auditing and an enquiry method using questionnaires. The research sample made up 50 (100%) of the examination documentation from five selected wards of Pelhřimov Hospital. The body of research in the case of the questionnaires was made by the nurses, working in the wards, where the audit of the nursing documentation was carried out. In total 46 of the nurses (100%) from all the assigned clinics took part in the survey. The results of the research were placed at the disposal of the administration of Pelhřimov Hospital, as a pilot study for the creation of their own complex standard and audit of the nursing documentation.

The main aim of graduation theses was today{\crq}s loom of species distribution, which were in former times surely ample in integral area Blatensko. On the basis of it to rewiev the diversity. With village{\crq}s alternation the diversity changed, too. The 22 villages scattered in the whole area phytogeografically subregion Blatensko and the 59 plant species from archeofyts, apofyts and neofyts were selected. My objective was to note, if these species are still occurred in Blatensko region or not. The number of monitored plant species decreases and every year are more and more abundant.

My bachelor thesis deals with theme inventories. Contains of their limiting, kinds, means of accounting. Of course the evaluation of supplies could not be left out. That is why one part is dedicated to the means of evaluation, decrease of inventories and their advantages or disadvantages. My work is also focused on special accounting of inventories as claim, deficiency, excess, goods in way, activation, discounts.

Characterization of fungal community in spruce (Picea abies) litter using cultivation and T-RFLP

Fungi have a key role in the decomposition of coniferous litter and affect nutrient cycling in forest ecosystems. Therefore, great emphasis is placed on exploring the diversity of these organisms. The aim of this thesis was to describe fungal diversity in spruce litter and revealed temporal development of this community in a forest regenerating after bark beetle outbreak. Another objective was to compare sites with different length of bark beetle damage. The study area was located in the Bohemian Forest mountain range. Litter bags with spruce needles were placed on the forest soil and several samplings were performed in the course of three years. Diversity of fungi and changes in the fungal community were assessed by two methods: cultivation of needles on 2ř MEA and fingerprinting method T-RFLP. In total 71 fungal species were obtained from needles during a three-year succession by cultivation approach. Using T-RFLP 122 different fragments were generally recorded. The dominant species were Scleroconidioma sphagnicola, Thysanophora penicillioides, Hormonema dematiodes, Ceuthospora pinastri, species of genus Chalara, Trichoderma polysporum, Mycena galopus and unknown species Helotiales sp. 1. SEPA and its implementation

The bachelor thesis describes the European cross-border integration, with emphasing on non-cash transactions and SEPA. It defines the basic characteristics and goals of SEPA, its organizational structure, provisions in the form of PSD, the basic objectives and the sectors involved in the operation of this project. Further on the thesis analyzes the progress of the project, its historical development up to the present time, discusses the actual payment instruments in detail, describes the migration phases and the actual course of the implementation of project in the Eurozone. The final part is dedicated to the implementation of SEPA in the in the Slovak Republic and describes the organizational structure, division of powers, the process itself and the state of implementation.

The main of the diploma work was to study the internal situations in individual states of Latin America so as to find out, how these countries were perceived in the Czech media. As a study material a group of articles was selected between the period of 2000-2003 from two renowned national newspapers, "Mlada fronta Dnes and Prava". Generally 278 articles were analyzed. Although the European Union is one of the richest parts of the world, there are striking internal disparities of income and opportunity between its regions. The entry of 10 new member countries in May 2004, whose incomes are well below the EU average, has widened these gaps. Regional policy transfers resources from affluent to poorer regions. It is both an instrument of financial solidarity and a powerful force for economic integration. The European Union allocates more than a third of the budget to the reduction of the gaps in development among the regions and disparities among the citizens in terms of well-being. One of the current priorities is to bring living standards in the new member states closer to the EU average as quickly as possible. The increasing budget of financial instruments for regional help is connected with appreciation of European regions and its role in the politics field. The last years have showed the tendency of strengthening the position of particular regions. Today regions are stronger partners than national states for lot of institutions. It is likely that this tendency is still intensifying after the enlargement in the year 2004.
