Revealing phosphoproteins playing role in tobacco pollen activated in vitro

5 Abstract Tobacco mature pollen rehydrates in vivo on a stigma tissue, and develops into the rapidly-growing pollen tube. This rehydration process is accompanied by the de-repression of stored mRNA transcripts, resulting in the synthesis of novel proteins. Furthermore, such metabolic switch is also likely to be regulated on the level of post-translational modifications of the already-present proteins, namely via phosphorylation, since it was shown to play a significant regulatory role in numerous cellular processes. Since only a minor part of proteins is phosphorylated in a cell at a time, the employment of various enrichment techniques is usually of key importance. In this diploma project, metal oxide/hydroxide affinity chromatography (MOAC) with aluminium hydroxide matrix was applied in order to enrich phosphoproteins from the mature pollen and the 30-minute in vitro activated pollen crude protein extracts. The enriched fraction was separated by both 2D-GE and gel-free liquid chromatography (LC) approaches with subsequent mass spectrometric analyses. STRATEGY OF CARE WITH DEMENTIAL PROBLEMS AT A SPECIALIZED DEPARTMENT OF CARE UNIT AT KOBYLISY SENIOR HOME

In my bachelor´s thesis I focus on the efficiency of the working methods of the personnel in the integration of activization programs and in the development of predispositions and abilities of individual clients with dementia at the Department of Specialized Care for People with Dementia, hic his a part of the Home for the Elderly, Prague-Kobylisy. In the theoretical part I explain the notion of dementia, describing the types of dementia, its manifestations, stadia and therapeutical methods. Furthermore, I describe the principles of communication with persons stricken by this disease, the evaluation of cognitive and functional abilities of afflicted people, needs of the personnel, activities and therapeutical methods used in the institutional care. The practical part relied on the qualitative research. Techniques applied for data gathering included concerned and unconcerned observation, interview and case studies. The secondary data analysis served as an auxiliary method. The central criteria applied in the selection of CASE was dementia and the terminated adaptation period. For the present research there were selected altogether 20 respondents aged 63 - 97 years afflicted by dementia of various types and in different stages of development. Out of the selected group of 20 respondents 6 persons were chosen for detailed case studies. The central objective of the work was to find out whether this group of clients with dementia can really benefit from the working methods applied at this particular specialized department. The target no. 2 was to find out whether the applied methods contribute to the longest possible preservation of self-sufficiency of clients with dementia. The target no. 3 was to convince family members to maximally collaborate with the personnel. In my view, the targets no. 1 and 3 were fully accomplished. The accomplishment of target no. 2 is relative and differs in individual CASE since the self-sufficiency of respondents gradually diminishes depending on their aging and disease advancement. Persons stricken by dementia are unable to take care of themselves, they have problems in expressing themselves, as if living in their own world, however, they do have their own needs, experiences and joys. These needs can be met by a caring family. However, the strength of the family is limited and very often it happens that the family is totally exhausted and unable to take care of the client. At that point it is necessary to provide institutional care by trained personnel that would ensure clients with dementia a kind and secure atmosphere.

This bachelor thesis deals with the issue of values, particularlywiththeviewof15-30 yearsoldyouthauthority questions. The theoretical part explains concepts such as youth, value, value systems and value orientation. It also focuses on family influence, the impact of school, and peer influence in current youth value systems.Furthermore, the theoretical part examines media and social networks, their influence on current young generation, and the role they play in youth´s lives. The practical part investigates youth´s value systems using a questionnaire, analyzes the sample of respondents, analyzes and compares individual particular results using graphs.

Possibilities of Activation of a Client Set at Liberty from Imprisonment from the Point of View of a Social Worker

As a subject-matter of my bachelor's thesis I have chosen "Actuation possibilities for a person released after serving the imprisonment sentence (SIS) in a social worker's view", because the theme is so actual in respect of the number of the imprisoned and the urgent need to motivate such clients towards active cooperation with social workers in order to succeed in their reintegration to our society. The bachelor's thesis is divided into two parts. Within the theoretical chapter, I am defining key terms and latest approaches which are both the most current in the field of social services and welfare, and essential for the research pursuit. I also deal with the hottest issues of people recently experiencing the SIS, their characteristics, life situation and the imprisonment impact on their personalities. In other chapters, I address to particular activities of social curators, their scope of work and tasks being applied upon treatment of clients released from prison after SIS. As for the elaboration of the research chapter of this bachelor's thesis and the accomplishment of stipulated objectives, a quantitative observation/questioning survey is used. The original and main aim of my bachelor's thesis has been to detect all needs of prospective clients, who are to be released from prison after serving the imprisonment sentence in near future, while residing permanently in the territory of the Pilsen City, to find out more on potential difficulties of a social curator-client cooperation within their mutual continual work and post penitentiary care, and also, in terms of the ascertained needs of such clients including the mentioned collaboration risks, to recommend a practicable actuation process of a client released from prison after serving a sentence. Based on the specified objectives, two hypotheses have been formulated. The research section describes the initial goals of investigation, methodology and techniques chosen for gathering the required information, and I am describing the search sample and the execution of data acquisition, too. Furthermore, I focus on the data interpretation and last but not least the discussion over the interviewing results. The intention of my thesis is fulfilled. The assumption no. 1 cannot be confirmed at all while the second assumption has now been fully proved to be true. This approach could be used as a guideline for other colleagues ? social curators, and/or it might also broaden the awareness horizons of the laics and general public on this tough and complicated situation of the clients released from prison sentence along with a social curator's challenging job.

Profile measurements of soil processed.

The aim of the bachelor's thesis is to create an overview of soil profile measuring ways and as a result of its analysis to design and construct a measuring device which would enable to record measured values for further computer processing. Cultivation is one of the most energy-demanding procedures in the crop production. It is possible to choose implements with various energy demand, but at the same way it is necessary to comply with the required agricultural conditions for a particular type of crop. To meet both requirements, it is desirable to ascertain the actual effect of the tool in soil or a real processed profile. The proposed device can detect the depth of cultivation, soil profile and soil looseness. These values serve to determine the working quality of complement, its setting and furthermore they are used for calculating the economic indicators of soil cultivation.

Planning and Analysis swimmers training in the basic stage

Title: Planning and Analysis swimmers training in the basic stage Objectives: The aim of the study was to experimentally applied training plan for swimmers in the basic stage of sports training. At the same time through achieved performance at selected competitions verify the effectiveness and validity of the implemented plan. Methods: In this work we used the method of experimental applications training plan. The method of indirect observation of the diagnostic techniques in swimming flume. The method of direct observation techniques for the implementation of training. Furthermore, for the diagnosis of swimming techniques was designed and used tables for qualitative analysis of swimming styles. Comparison method were compared to selected indicators of general training and the achieved performance swimmers. And lastly, was also used the method Conconi test for the detection of physiological parameters in stress tests. Results: We found that the training program was effective. Has improved the technical level swimmers and achieved performance improvement in swimming competitions. The test subjects were also monitored parameters VO2 max. Comprehensive investigation of penetration enhancers with complementary analytical techniques.

Transdermal drug delivery system is in the centre of attention in recent years. For efficient dermal drug delivery the drug has to overcome the barrier of the outermost layer of the skin, the stratum corneum. For facilitating dermal drug transport, the barrier properties of the stratum corneum can be varied by applying chemical penetration enhancers. The aim of this work was to characterize various penetration enhancers and investigate their mechanism of action. We combined well established techniques like differential scanning calorimetry (DSC) and infrared spectroscopy (IR) with confocal Raman microscopy (CRM) as an upcoming technique in skin research. CRM offers the possibility of label-free and non- destructive, chemically selective analysis of stratum corneum lipids and proteins. We used isolated human stratum corneum for incubation with the penetration enhancers. As a novel approach, the samples of treated stratum corneum were freeze dried to avoid any discrepancies which might come up with differences in the hydratation state of stratum corneum (SC). Furthermore, the structure of lipids and proteins in the stratum corneum was analyzed. Joint Learning of Syntax and Semantics

This master thesis addresses the problem of learning varying levels of abstraction of linguistic knowledge. The main focus is on learning latent semantic information as represented by frame classes, verb classes and role fillers by exploring recent successes in Bayesian modeling with hidden variables. Furthermore, close coupling of syntax and semantics is captured in the joint model which also incorporates valuable lexical information. The result is a language-independent, feature-less model of semantic information with a performance comparable to the current state of the art.

Modulations in PLC systems

The bachelor thesis describes principles and methods of Powerline Communications, practical usage possibilities both in industrial applications but mainly in broadband systems that may represent promising competition to conventional broadband access, e. g. xDSL connections without the need of new wiring, WIFI etc. The thesis also focuses on problems that can occur in the communication channel from the view of electromagnetic compatibility and assorted interferences. The 50 Hz frequency electric network (60 Hz in the USA) was not designed for data signal transmission, problems may arise mainly at higher frequencies and broadband communication. It is necessary to implement methods which enable transfer of information signal through a communication channel with minimalization of problems such as interference, cable attenuation, electromagnetic compatibility, bit error rate (BER), etc. Furthermore, currently used modulation principles and their characteristics are described, such as spread spectrum modulation DSSS, FHSS, traditional OFDM, alternative modulations FMT and DWMT with filter banks. The last chapter of the bachelor thesis features models of particular modulations created in the Matlab-Simulink program and the conclusion presents advantages and disadvantages of their implementation in PLC systems.

The technique of the throwing disciplines in the heptathlon of 14 to 16 years female athletes

Title: The technique of throwing disciplines in the heptathlon of 14 to 16 years female athletes Objectives: The aim of the research is to observe changes in locomotory tasks - discovering changes in the technique of shot put and javelin throw and revealing changes of performance in both disciplines of young heptathletes within four months (March-June 2010). Methods: The research consists of a detailed video image analysis of the locomotory tasks within an interval of four months. Comparing the technique of shot put and javelin throw and also concerning the preparatory training, the training was intended to help to improve technical expression and performance. The girls were measured and pre-controlled in tests in running (50m of the high start-up item), jumping (long jump from the place) and throwing (medicine ball over the head weighting 1kg and 2 kg). Furthermore, height and weight of the observed girls were recorded, too. Data were collected by non-standard methods of valuation and comparison. Results: I found out that training exercises are necessary for practising the technique and that they have indeed a relevant influence on the performance.
