Biochemická a funkční charakterizace nového transmembránového adaptorového proteinu NVL
Vonková, Ivana ; Černý, Jan (referee) ; Brdička, Tomáš (advisor)
In T cells, signalling events initiated at the immunological synapse by T cell receptor (TCR) have been already thoroughly studied. In contrast, signalling pathways involved in MHCII-mediated signal transduction on the APC side of this structure are still rather poorly understood. Transmembrane adaptor proteins (TRAPs) are a class of proteins with an essential role in signalling triggered by TCR and other immunoreceptors. So far no TRAPs involved in MHCII signalling have been identified. In this work a new TRAP, highly enriched in the immunological synapse in APCs is described and termed Nvl. Nvl is expressed exclusively in professional antigen presenting cells - B cells, monocytes/macrophages and dendritic cells, and the level of its expression positively correlates with the expression of MHCII. Nvl is present in tetraspanin microdomains and together with tetraspanins it is localized to the polarized structures of the cell, including uropod, cleavage furrow, and, most interestingly, the immunological synapse. The presence in the immunological synapse indicates possible role for Nvl in MHCII- mediated signal transduction. Indeed, Nvl becomes tyrosine-phosphorylated after stimulation via MHCII, although strong phosphorylation has also been observed after FcγR stimulation or treatment with...

Renaissance Appearance of Melnik Castle
Čermáková, Lucie ; Zlatohlávek, Martin (advisor) ; Nespěšná Hamsíková, Magdaléna (referee)
Renaissance appearance of Mělník chateau Bakalářská práce - Lucie Čermáková The appearance of Mělník chateau has been developed for centuries. We cannot tell it is a typical Renaissance building structure but the most significant reconstruction took place in a Renaissance era. At that time, its horizontal projection was L-shaped. A person in possession, at those times, Zdislav Berka z Dubé had merit in most extensive treatments of the chateau in mid-1600. He invested considerable amount of money into building-up of the northern arcade wing-wall and a sgraffito decoration of the courtyard. Building treatments got under way in 1542 when Berka acquired the chateau and its finalization is assumed to have come between 1554-1555, also according to extant bills and engraven style found in sgraffito decoration. Sgraffito decoration came into existence in connection with a new build-up of the northern wing-wall and a staircase tower, and it also dates back to the same period. Mělník's sgraffito was considered as the oldest dated sgraffito decoration for a long time. However, we know about older examples of dated sgraffitos nowadays which cannot change the fact that Mělník's arcades, including its decoration, still belong to one of the oldest. The structure of Mělník's arcades are not to be found anywhere...

The Spider Game
Ziegler, Martin ; Plch, Tomáš (advisor) ; Bída, Michal (referee)
We discuss the process of designing and of the following realization of our computer game Spider. We talk about the means that the game design uses to accomplish the goals that we have set for the game. We discuss the game's implementation architecture that uses the OGRE library to render the 3D scene, the Awesomium and NaviLibrary libraries to render the 2D user interface that is described in HTML and the Lua library for scripting purposes. We show the technical solution that we have used to extend the Lua library, so that the scripts can start their execution automatically based on the events in the game world, and so that the scripts can pause their execution on demand.

Comparsion of biochemical profile from plasma in patients with dyslipemia and type 2 diabetes after treatments of MDOC (chloruron).
Vaculíková, Ivana ; Semecký, Vladimír (advisor) ; Martiník, Karel (referee)
The selected set of 28 patients (18 women and 10 men, age 47-79 years, median 65.3) from the database clinic for metabolic diseases and nutritional disorders II. Policlinic , s.r.o. in Hradec Kralove was made the determination of selected biochemical parameters of peripheral blood - BIL, GLU , GHB, liver enzymes ALT, AST, GGT, fat spectrum of values of TC, HDL, TG, VLDL, IA, and RICH proteins CRP and homocysteine. These initial values are important for clinical assessment 2 diabetes type and their values provided above biochemical laboratory equipment. This was compensated for elderly patients suffering from DM 2. typu with dyslipidemia who were treated with lipid-lowering agents containing for long-term HMG-CoA inhibitors (statins). After a period of 3.5 months was administered to patients with the working title nutraceutic Choluron (MDOC) at a daily dose 3 x 2x 500 mg with the intention of a possible effect on lipid and carbohydrate metabolism. After the period mentioned above were examined by biochemical values were recorded and the expected reduction in bilirubin, and so also of TC and VLDL. On the contrary, showed an adverse effect on the HDL and TG. The GLU and GHb were probably influenced by the value of individual lifestyle. The beneficial effect of Choluron was observed also in liver...

A contribution to the history of the Sokol movement in the Ceskomoravska Highlands. The Sokol union in Polna in 1869-1953
Urbánek, Miroslav ; Pokorná, Magdaléna (referee) ; Waic, Marek (referee)
This paper, entitled "A Contribution to the History of the Sokol Movement in the Ceskomoravska Highlands. The Sokol Union in Polna in 1869-1953", describes the history of this association ('Tělocvičná jednota Sokol Polná' in Czech) whose activities often influenced the local historical course of events. The paper is divided into three main chapters. The first chapter gives a general overview of the Sokol movement. The first two parts contain a brief analysis of the circumstances of the foundation of the Sokol movement, the establishment of the COS (the national administration of the Sokol movement) and the subsequent development of the Sokol movement in the Czech lands. The following part briefly explains the history of organisation of the COS, including the establishment of its local and regional bodies, up to the 1950's when the COS ceased to exist. The key events affecting the COS eventually had an impact on all its associations, including TJ Sokol Polna. The next part gives a brief history of the Sokolska zupa Havlickova, the regional association which also included TJ Sokol Polna in 1894-1948. The second chapter as the fundamental component of this work contains, in five parts, the history of the Sokol Union in Polna. The first part is dedicated to "Sokol Polna", established in 1869. The activities of...

International terrorism and human rights
Dutkevičová, Lenka ; Šturma, Pavel (advisor) ; Bílková, Veronika (referee)
145 International Terrorism and Human Rights Terrorism has posed a threat to humans, and their lives and rights for centuries; however, it started gaining attention in the 19th century, as it became quite a widespread means of reaching political goals first of anarchists and later other groups. Every generation has claimed 'their terrorism' to be a completely new sort of terrorism never seen before, but all this has been more about perception rather than real change of the situation. One such new era allegedly started with the terrorist attacks on the WTA and the Pentagon in the USA on 11 September 2001. It is arguable whether that terrorism is a new one, but the response was definitely harsher than before, because this time reaction to one event engulfed the whole world. The attack was certainly bigger than usual, that much must be admitted, and it targeted symbols of a nation that had been endangered on its soil in such a way just once before, a superpower whose vulnerability was suddenly revealed. The people all around the world got scared and politicians reached to strong means of fighting the threat and protecting their people, sometimes restoring to controversial or blatantly unlawful ways, occasionally clearly exploiting the situation. International law - general, humanitarian, criminal and mainly...

The analysis of influence of reimbursement regulatory fee in regional hospitals on the length of stay.
Junga, Přemysl ; Bolcha, Peter (advisor) ; Bartoň, Petr (referee)
The thesis analyses the influence of reimbursement regulatory fee for hospitalization which was introduced in regional hospitals in 2009 in Czech Republic. The difference in difference analysis was used to research the possible relationship between reimbursement of the fee and length of hospitalization in acute care hospitals and in after-care facilities. In acute care the influence was 0,5-1 % of the length and in after-care facilities between 8-12 %. This relationship may be biased because of introduction of DRG system which may decrease the length of stay and may be differently distributed between treatment and control group.

Process of forming Satchō Alliance and its influence on the fall of Tokugawa shogunate
Hrůzek, Vilém ; Labus, David (advisor) ; Sýkora, Jan (referee)
In my work, I am dealing with the process of forming the Saccho military alliance and its role in overthrowing the Tokugawa reign. I present an overview of major events in the 1860s that led to the formation of the alliance, and also give insight into the political scene of these years. Furthermore I deal with important political movements that influenced the last days of the Tokugawa shogunate. In addition, I focuse my attention on the important military events that led to the negotiation on the alliance. In the process of forming the Saccho alliance itself I briefly describe the key personalities who have contributed to the successful conclusion in the formation of the alliance between Sacuma and Choshu.

Possibilities of logopedic intersention in geriatric medical institution
Baborová, Eva ; Vondrášková, Dana (referee) ; Housarová, Blanka (advisor)
The topic of the thesis deals with the issue of the people after cerebrovascular events and their subsequent rehabilitation. Introduction of the theoretical part is devoted to the definition of age in terms of physiology and psychology. The starting point for a theoretical treatment of the subject is the concept of comprehensive care for people after cerebrovascular events. On the basis of theoretical texts, this part focuses on the most common types of communication disorders in people with neurological disease, especially on aphasia, dysarthria, dysphagia, and cognitive communication disorders. I focused the practical part on the case histories of two patiens at Geriatric and rehabilitation center in Kladno, where I had watched their three-month stay. I gave the greatest attention primarily on speech therapy, but I also focused on other provided care. This section also includes a rating scale of the availability and level of speech intervention. The results of this work is evaluation of speech therapy, physiotherapy and ergotherapeutic activities in a rehabilitation facility. The information should help to consider the need for high quality and comprehensive care for people after cerebrovascular events.

Nursing care of patient with prostate cancer
Taitlová, Eliška ; Zvoníčková, Marie (advisor) ; Otava, Zdeněk (referee)
For a long time I am interested in the issue of patients terminally ill and dying, especially in the context of palliative and hospice care. It is for this reason I chose a case study of a patient with advanced disease for whom curative medicine has exhausted all its possibilities, and for which the ideal has become a palliative care provided in a hospice inpatient facility. In this work, first I present the anatomy and physiology of the prostate gland / prostate cancer, the pathophysiological basis of prostate cancer, diagnosis and treatment options, and basic patient data, all in the clinical part. The nursing section deals with the idea of palliative medicine and hospice care specifics, according to which the patient was maintained throughout the nursing process. In drawing up the plan of care, I focused on searching for current and potential patient problems and their effective solutions. I lined nursing problems / diagnoses according to priorities. For each diagnosis, I identified the target, which should realistically be achieved and accordingly determine the appropriate nursing interventions. In conclusion, I assess the process and outcome enrolled. Nursing section concludes psychological evaluation and patient education. In the end, work is a list of reference books and other sources, abbreviations,...
