TITLE: Analysis of graphics editors ABSTRACT: The aim of the diploma work is to compare and evaluate mainly free raster and vector editors and viewers (photo file managers) which could be used at the environment of primary and secondary schools. In the opening part of the work there are briefly mentioned real possibilities of using graphics applications at schools. Separate analyses of all programmes are described in following parts. The analyses deal with features of the programmes, hardware requirements and usability. The comparisons should offer enough general information for choosing the correct software. The important part of the work is the research and it's three questionnaire investigations. Their results demonstrate real usage of graphic software among informants from all of the Czech Republic. Possibilities of Social Workers at Schools

This thesis is about the possibilities whether to employ social workers in primary schools. The aim my work was find out, whether pedagogical staff in primary schools consider the presence of a social worker as very useful and important thing in primary schools. Furthermore I compared views of the pedagogical staff of primary school in villages consisting of up to10 000 inhabitants and above 10 000 inhabitants their views concerned the utility of social workers in primary school. In theoretic parts I explained the important areas related to a specific subject like: social - work, a caseworker, school social - work, consultancy at schools and relationship between the family and the school. In practical parts are described aims of work, conjectures, methods research and rating experimental set. These are all present above the record quantitative research. I chose this asking method through the questionnaire techniques. The research went ahead via direct and indirect (electronic by post) contacting form. The pedagogues were the experimental set of the selected primary school in towns consisting of up to 10 000 inhabitant and above 10 000 inhabitant. Records showed that the pedagogues assume that the introduction of this position for school caseworker at primary school would be in Czech Republic beneficial. To employ the caseworker at primary school were The views about employing a caseworker at primary school were both the same (pedagogues of primary school in municipality up to 10 000 inhabitants and above 10 000 inhabitants). Curtail school activities for the caseworker would be social consultancy, social therapeutic activity, activity in the area of prevention and social rehabilitation. According to pedagogues the biggest problem in introduction of school social - work is the lack of finances in school resort. There is not mention a lot about school social - work currently in the Czech republic, therefore I would like this thesis to become the base for discussions upon this subject and for others experimental activity.

Study of transferrin as a marker of congenital disorders of glycosylation

Congenital disorders of glycosylation (CDG) represent a heterogeneous group of mul- tisystemic metabolic disorders which are caused by defects in biosynthetic pathways of glycoproteins. The screening test for N-glycosylation disorders is the analyses of sialylated isoforms of serum transferrin (Tf) by means of isoelectric focusing (IEF). Two distinct pathological IEF patterns of Tf are observed. A type I pattern is cha- racterized by a decrease of tetra- and an increase of di- and asialotransferrin, whereas a type II pattern shows in addition an increase of tri- and monosialotransferrin. The aims of diploma thesis were: 1) to evaluate reference range for spectrum of sialylated forms of Tf separated by IEF and 2) to perform biochemical and molecular analyses in three patients (P1-P3) with clinical suspicion for CDG. Serum and genomic DNA from three patients with clinical suspicion for CDG and family members of P1 were analysed. Sera from 99 healthy volunteers within the age range of 2-42 years served as a control group. Tf was analysed by IEF with direct immunofixation, SDS-PAGE and Western blot using specific antibody against human Tf (Dako). Profiles of Tf were quantified by AlphaEaseFC software (Alpha Innotech). The posibility of inorganic sulphate determination in patients with severe metabolic disease

Inorganic Sulphate play important role in various metabolic pathways. The aim of the work was to assess the posibility of inorganic sulphate determination in patients with severe kidney diseses undergoing hemodialysis. Serum sulphate levels were determined in 68 patients (31 men, mean age 67, range 31 - 86 and 37 women mean age 71, range 44 - 86) before and after hemodialysis. Two turbidimetric methods Quantichrom TM Sulfate Assay Kit and BGR gelatine method were used for determination. Both methods were performed separately in group of 34 patients. Results were as follows: Mean serum sulphate levels in patients before hemodialysis are significantly elevated than the levels after hemodialysis (p<0.0001, non parametric t-test, alpha = 0.05). BGR gelatine method show less significant differences in sulphate levels before and after hemodialysis than the Quantichrome Assay method. (p = 0.037 vs. p < 0.001, non parametric t-test, alpha = 0.05 ). This fact could be explained by the significant role of interferences in BGR gelatine method. We confirm the relevance of both metod in inorganic sulphate determination. BGR gelatine method is more affected by interference, than the QuantiChrome Assay method. Powered by TCPDF (www.tcpdf.org)

Analysis of aesculin, aesculetin and phenylephrine in eye drops by micellar electrokinetic chromatography

Title of Diploma Thesis: Analysis of aesculin, aesculetin and phenylephrine in eye drops by micellar electrokinetic chromatography A new method of micellar electrokinetic chromatography with UV detection for the simultaneous determination of aesculin, aesculetin and phenylephrine was devised and validated. The separation was optimized by examining a number of experimental conditions, such as type, concentration and pH of the background electrolyte, the addition of a surface-active agent (sodium dodecyl sulfate, SDS) and methanol and effect of applied voltage and operating temperature. The separation was carried out in a fused-silica capillary (internal diameter 50 µm, total length 64.5 cm and effective length 8.5 cm) with UV detection at 210 nm, using hydrodynamic injection under the pressure of −50 mbar for 6 seconds. Optimal conditions for the separation and determination of the drugs were: background electrolyte 20mM boric acid of pH 8.6 (adjusted with 100mM NaOH) containing 60 mM SDS and 5 % (V/V) of methanol. Application of metamaterial structures in terahertz spectral range

The thesis is devoted to a research on metamaterials for terahertz spectral range based on deeply etched silicon. The aim of the work is a theoretical conception and experimental realization of wave plates for selected terahertz frequencies. These wave plates are made of silicon substrates with an etched two-dimensional periodic microstructure where the dimensions of an elementary cell are below the considered wavelength. Theoretical proposal is optimized using the transfer matrix formalism. We designed and experimentally characterized quarter-wave plates for frequencies 0.5 THz and 1 THz and a half-wave plate for frequency 1 THz.

HPLC analysis of drug candidates from the group of aroylhydrazones II.

1. ABSTRACT High performance liquid chromatography (HPLC) is one of the most frequently used analytical techniques for the analysis of drugs. Although iron is a vital element, excessive amounts in the body are highly toxic. The search for highly selective and effective iron chelating agents has been mainly inspired by the need to mobilize iron from tissues that are chronically overloaded with iron. However, recent investigations focused on the possibility to use iron chelators for the treatment of many other pathologies. Salicylaldehyde isonicotinoyl hydrazone (SIH), a biocompatible iron chelator derived from aroylhydrazone, is under extensive investigation as a promising drug candidate. Besides ability to bind iron, it shows interesting pharmacological effects: antioxidative, antiproliferative, cardioprotective, antimalarical and antimicrobic. The aim of this study was to develop optimal HPLC conditions for the separation of SIH and its potential metabolites (isoniazide, acetylisoniazide, salicylaldehyde) and to apply the method to the study focused on the isolation of analytes from rabbit urine using SPE.
