Project of nuclear power unit secure emergency power supply for design basis accident as well as for extension conditions

The present Master´s thesis aims at designing an evolutionary scheme of secured power supply at nuclear power plants with VVER 440 reactors during design basis accidents as well as design extension conditions. In the first part of this thesis, concepts relating to the defence in depth of nuclear reactors, operating modes of the blocks as well as types and possibilities of electrical power supply and electric circuits of the block are defined. Although the present thesis deals with PWR 440 in general, special emphasis is put on the Czech NPP in Dukovany, where there are four PWR 440 reactors in operation, and on the possibilities for enhancing the defence in depth in this area. The second part of the thesis deals with the difference in station blackout definitions before and after Fukushima; not only the differences in situation evaluation are dealt with, but a solution is also proposed to make the system of secured power supply system during design basis accidents as well as design extension conditions more robust. This option has been selected out of a number of possibilities based on the evaluation of reliability, availability and cost-effectiveness of the proposal.

Assessment influence of the various aspects on the QoE in 3DTV: Subjective tests

This thesis studies aspects influencing image quality 3D TV systems. Thesis is focused on comparing TV´s of the same brand with different diagonals and compares the change in perception of the evaluators by this parameter. The basic evaluated parameters are image depth, contrast, color rendition, image crossover and overall impression. Describes the creation of testing sequences for subjective tests, testing and evaluation of testing.

Expression of epistemic modality in Spanish news texts in comparison with Czech texts.

The core of the present master's thesis is a comparative analysis of means of expressing epistemic modality on the background of Spanish and Czech journalistic texts. Initially, the discourse analysis and the concept of theory of enunciation are presented. Secondly, the author defines the term modality and its concept in terms of modal logic and linguistics. She concentrates on different typologies of modality presented by foreign and Czech authors as well. Further, the works focuses on the category of evidentiality and means of expression (especially lexical) of epistemic modality. The proper research is based on qualitative and quantitative analysis. The aim of this investigation is to compare the frequency of occurrence of the epistemic markers between the aforementioned languages.

Europe Direct České Budějovice and Eurocentrum České Budějovice as Two Main Sources of Information Concerning EU in the South Bohemia: a Comparative Analysis

The main theme of the presented bachelor thesis is a comparative analysis of two main European Union information sources in South-Bohemian Region, the Europe Direct České Budějovice and the Eurocentrum České Budějovice. The first part is a theoretical excursus into problems of information science which plays a lead role in present world. The views of the South-Bohemian Region´s citizens were assessed by the questionnaire survey carried out on 100 subjects. Another part concentrates on the in-depth analysis of the mentioned centers. Analyzed data are related to the legal basis of the centers, history and description of the centers, method of funding, objectives and services provided by the centers, as well as their cooperation with other institutions. Considering the previous chapters, the last part is focused on comparison of the Europe Direct České Budějovice and the Eurocentrum České Budějovice. The conclusion summarizes the main coincident and different points of both compared centers, as well as the views of the South-Bohemian Region´s citizens on the level of acquaintance.

Transformation of the values of the individual by Friedrich Wilhelm Nietsche and Søren Aabye Kierkegaard.

Goal of this work is to show that the values in European and a traditional context could be transformed. I chose two philosophers F.Nietzsche and S.Kierkegaard where I can document this change. Just the two of them proved to be very important for the 20th century .In the main part of my work, I am describing and comparing the evolution of the axiological views of both philosophers. My study on existential philosophers includes topics such as freedom, sense of life, base of christianity, perception of human being. Nietzsche and Kierkegaard are philosophers who have a lot in common, but they would not get on well with each other. The things that they share are the same time what they deal with in a difááé=ferent way. They treat topics such as existence, futility, strength of faith. However Kierkegaard studies in depth existentialism, immersion in irrationality, desolation. In Nietzsche we can find the will of power, the nihilist opinions and the criticism of christianity.

Opinions of students and their parents on the prestige of various types of education in secondary schools in Písek

This dissertation pays attention to opinions of parents and students on the prestige of various types of education on secondary schools in Písek. The dissertation presents conclusions of several Czech as well as international studies, dealing with the matter of secondary school education. Further on it describes some circumstances influencing secondary school education. It also introduces historical development and the present of Czech secondary school education and the system of secondary school education in some of other European states. It characterises various secondary schools in Písek and also looks at a secondary school student from the psycho-sociological point of view. The data obtained from a personal qualitative research are further on interpreted an evaluated and in the following discussion compared with the findings of the initial research. The most prestigious type of secondary education amongst students as well as their parents is general secondary school education followed by specialized secondary school education and finally vocational education. It is evident that the parents' evaluation differs and is also influenced by the importance for further professional life. Cultural capital also plays a very important role. Parents prefer the type of specialized secondary school education followed by a graduation exam. The prestige of all types of secondary school education has recently been decreasing especially due to the absence of entrance examinations and badly established education financing system and a high number of private secondary schools.

Mercury speciation in forest soils

We compared distribution of Hg in soil profiles with different level of Hg content in forest floor humus. Detailed study of soil profiles revealed that at the locality with lower Hg content in humus was higher Hg content in mineral horizons in comparison to locality with higher content in organic horizons. Most of the Hg in forest soils was bound to organic matter.

Family business

This thesis focuses on family business companies and above all on their problem of succession planning. For the purposes of this work, I have chosen a family business company that is owned by two shareholders. Both shareholders are going to leave the company within next 5 to 10 years. The thesis deals with the succession plan of both shareholders and this concerning the rate of preparedness as well as the rate of coordination of both plans. Prior to the research, two hypotheses were fixed. The first one says that succession in family is the best strategy of succession for the chosen company. This strategy has been confirmed. The second strategy saying that the shareholders as well as the generation of successors have different ideas about the company handover was not confirmed. The shareholders and their chosen successors accord in basic points with the company handover. The STEP analysis of macro-relationships as well as the Stakeholder analysis of micro-relationships was done as a part of the research. The evaluation of influence of different interest groups in the scope of the Stakeholder analysis was performed by the shareholders. The primary research was completed with three interviews: the first one, analysing interview with one of the company shareholders; the second one, structured depth interview with both shareholders and with their sons ? the chosen successors and the third one, evaluating interview. On the basis of the above mentioned analyses and interviews, a SWOT analysis of internal milieu (of its strenghts and weaknesses) as well as of external milieu of the company (of its threats and opportunities) was performed and a strategy focused on the succession planning was proposed. This strategy was presented to all interested participants during the mentioned third, evaluating interview and the objectives including deadlines and responsibilities were fixed. The proposed strategy compounds of three parts. The first and second part focuses on elimination of weaknesses of the company. The company thus has to weaken the risk resulting from the splitting of company shares between two shareholders with equal parts and further, the company has to eliminate the risk of its bankruptcy during the handover to the generation of successors. Third part of strategy focuses on exploitation of the company´s strenghts in order to weaken the external risk and to valorise the opportunities. The company will thus try to exploit its competitive advantages at the market threatened with the present depression. Detected competitive advantages are the exploitation of modern methods and technologies in company management and extension of its services through an engineer's office.

Analysis of recombinant clones of apoptotic nucleases in "leaf factory" system upon coinfiltration with modifying genes

TBN1 is a nuclease with antitumor activity. The main goal of this work was to estimate how TBN1 and its modificated variants are stable in the ?leaf factory? system used for its production and whether it can be enhanced or influenced by chosen potential ?modificators? i. e. silencing supressors, transcription factors, glycosyltransferases and kinases. Nicotiana benthamina plants were infiltrated with the mixture of Agrobacterium tumefaciens strains bearing the nuclease plant expression vectors and co-infiltrated with the ?modifying? vectors. The nuclease and protein analyses revealed that nuclease TBN1 wt and its modificated variants are stable in the used ?leaf factory? system as to their molecular mass, only quantitative changes were detected. Expreximents showed that activity and production of the nucleases increased upon coinfiltration with silencing supressor and decreased upon coexpression with chosen transcription factor. Glycosyltransferases and kinases influenced activity and production only insignificantly. The experiments also revealed that modificated variants of TBN1 have different molecular weight suggesting that different N-glycosylation domains have different length of sugar chain and influence on nuclear activity. Our data show that this expression in planta seems to be suitable for production for study of antitumor activity of these nucleases.

Crustal Structure of the Bohemian Massif Based on Seismic Refraction Data

The deep structure of the Bohemian Massif, the largest stable outcrop of the Variscan rocks in central Europe, was studied using the data of the international seismic refraction and wide- angle reflection experiments CELEBRATION 2000, ALP 2002 and SUDETES 2003. The data were interpreted by seismic tomographic inversion and by 2-D trial-and-error forward modelling of the P and S waves. Above, additional constraints on the crustal structure were imposed by reflectivity or gravity modelling, and by receiver function interpretation. Knowledge of the crustal velocity structure in the Bohemian Massif was complemented by its azimuthal variation. Though consolidated, the Bohemian Massif can be subdivided into several tectonic units separated by faults, shear zones, or thrusts reflecting varying influence of the crust forming processes. The resultant velocity models determined different types of the crust-mantle transition reflecting variable crustal thickness and delimiting contacts of these tectonic units at depth.
