Transformation of Sacrifice in the Scripture. Theology of Sacrifice.

Diachronic comparison of three political and religious systems of three independent geographical areas (ancient Egypt, Mesopotamia, and Syria Palestine) with the faith of ancient Israel shows certain aspects important for the kerygmatic theological message of Tanach as well as the New Testament. Significance of the king, priest, prophet, and religious ceremonies in the temple is derived from their specific understanding of the divine. However, their religious systems completely lack (with the exception of Israel) eschatology. The message about the transcendent/immanent God Lord (Jesus' Father) is impossible to reconstruct from the available sources through diachronic methodology. Furthermore it cannot be understood as a mere result of religious synchronic redefinition of the Scriptures. The New Testament transforms the Old Testament religious system to narrative kerygma about the sacrifice of both Father and Son. It speaks about the Risen Lord, who is able to reconcile God with his adherent. Such a message is unheard of (unexpected) in the Second Temple Judaism. Even the Church Fathers do not interpret Christ's conscious ebed-like sacrifice along the lines of the ritual forgiveness of the OT. Based on the scholarly analyses from the proponents of the so called Prague school, as well as those coming from theologians stressing the paradigm of biblical interpretation through the event of resurrection, this dissertation concluded in the subject matter of soteriological transformation of the sacrifice the following: Agnus Dei, with his crucifixion being a ritual slaughter, founds the new community through his own Pasch, which has also the futuristic (eschatological) aspect both in the NT and in the faith of the early Church. The meaning of this sacrifice will be actualized in 'drinking of the chalice' by the Lord Jesus with the resurrected Church in the coming new Creation. Thus, the Eucharist becomes a unique 'cultic' homage (Temple ceremony) through existential acceptance of the risen Lord. The same concept can be found at the heart of the Patristic thinking (inclusive paradigm of Christ's sacrifice, that is Christian obedience even unto death) . The Fathers understood the sacrifice as means of overcoming alienation from God by metamorphosis of one's life based on Christ's sacrifice not to God, but to people. Such a sacrifice is life-giving activity of the Lord God himself and is able to renew the life of God's people and make them heirs to the eternal life.

The effect of new taxol-binding peptides on proliferation of cancer cells

Charles University in Prague Faculcy of Pharmacy in Hradec Králové Department of Biochemical science Candidate: Hana Vávrová Supervisor: Prof. RNDr. Lenka Skálová, Ph.D. Title of diploma thesis: The effect of new-binding peptides on proliferation of cancer cells Paclitaxel (PTX), alkaloid from the bark of Taxus brevifolia, is used in treatment of ovary, breast and some form of lungs cancer. In Institute of experimental botany in Prague were preparated new conjugates with peptide agonist of receptors for gonadotropin-releasing hormon (GnRH). The previous studies showed that this conjugate have higher or same antiproliferative effect. This antiproliferative effect was compared with PTX on the cell line from breast cancer which have receptors for GnRH. The aim of this study was to compare the anti-proliferativ effect of PTX and its conjugates. For the experiment was chosen to two cell systemes - HCT 8, which is derivate from colorectal adenocarcinoma and primary culture from liver cell from sewer-rat. Both cell lines have not receptors for GnRH. The results were evaluated by three different methods (MTT, NRU and x-Celligence). The conjugates MP 264, MP 211 and MP 260 showed lower antiproliferative effect on the cell line HCT 8 and the conjugate MP 264 and MP 265 showed lower cytotoxic effect than PTX...

Use of bioproducts in public catering systems in Czech Republoc and Austria

The paper studied the structure of meals in selected restaurants in the Czech Republic and compared with selected Austrian catering facilities. The analysis shows that Austria is against the Czech Republic ahead of not only the quantity used in organic food, but also in the knowledge that is related to organic food. And not just in public catering facilities, but also in households. In the questionnaire survey was to determine knowledge about organic foods, the principles under which they were prepared, where you can buy at what price range varies. About the quality of organic food, people's interest in organic food, organic food as often used in homes both Czech and Austrian. Were also detected in the possibilities of school catering facilities, the reasons why organic food is often used and more. All of this was examined children's nursery school, primary school pupils and college students and executives interviewed kitchen. The result is that in the Czech Republic are used almost exclusively organic foods at home and rarely. While in the kitchens of school facilities for them is great interest either from the kitchen diners and employees. By contrast, in Austria, organic food is commonly used both in households and in public catering facilities and thein use is considered an asset. SWOT analysis derived from several sources provided by the Ministry of Agriculture, shows the appropriateness of inclusion of organic food to offer menus. Understanding the concept of organic food is gradually increasing, from our survey suggests that this concept students know 72% and 75% of staff kitchen. In Austria, this question knew the answer 100% of the respondents. 13% of respondents would welcome the offer of meals of organic foods in their cafeterias, dining halls in the Austrian would appreciate higher proportion of organic food. 60% of students in the questionnaire said that they can byu organic food in supermarkets, while in Austria prevails buy organic food directly on the farm. Most in the Czech Republic to sign purchase organic fruits and vegetables, in second place as milk and dairy products. In contrast to Austria, where most purchases organic milk and dairy products, fruit in second place and third place is vegetables.

The Pardubice Crematorium

Main objective of this thesis is the building of Pardubice crematorium, which is an example of national architect style, so-called Rondocubism, created by the architect Pavel Janák. First part is dedicated to the theoretical fundaments derived from the former pre-war style of kubismus. In the second part these fundaments are explained on the submitted drawing proposals for the structure of Pardubice crematorium dating from the year 1919. Then the analysis of these proposals from Janak's first drafts till the final appearance of Pardubice crematorium can be seen as the main contribution of this work. Furthermore author adds both the chapter documenting the opinion changes relating to this building and the short outlook on the topic of cremation which gives reader a better understanding of period social context. Preparation of chicken antibodies against phosphoserine residues of phosphoproteins

New type of immunogen for the preparation of chicken antibodies specific for O- phosphorylated amino acid residues of phosphoproteins was prepared: the α-casein phosphopeptide mixture was coupled to maleinylated derivative of bovine serum albumin. Affinity chromatography on immobilized Fe(III) ions (Fe(III)-IDA-Sepharose) was used for the separation of the α-casein phosphopeptide fraction from the α-casein peptide mixture obtained by the proteolytic digestion using trypsin. The presence of coupled phosphopeptide was shown by means of MALDI-TOF MS analysis. After chicken immunisaion with the prepared immunogen the immunoglobulin fraction was isolated from egg yolks that was further purified using affinity chromatography on α-casein-Sepharose and bovine serum albumin immobilized on Sepharose (BSA-Sepharose). The specificity of obtaine immunoglobulin fractions was tested by means of ELISA tests. The obtained results showed, that the prepared chicken antibodies and their fractions obtained by affinity chromatography on α-casein-Sepharose do not recognize the O-fosforyl-L-serine residues of phosphoproteins (α-casein, phosvitin, ovalbumin). Using the prepared different antigens it has been shown, that prepared chicken antibodies interact with maleinylated chain coupled to carrier proteins. .

In experimental cardiovascular research, enzymatically dispersed neonatal or adult cardiomyocytes from various animal species are considered as a "gold standard" for such experiments. However, besides isolated primary cultures, permanent cell lines are becoming extremely popular among cardiovascular scientists. H9c2(2-1) is the first permanent cardiomyocyte-derived cell line. Although these cells are known since the mid-seventies, there is not much information about their phenotype. This study followed the results of the shotgun proteomic analysis, which identified about 1500 proteins. Professional Chaplain Service in Czech Prison

This thesis is about prison chaplains in the Czech Republic. It focuses on the connection of their work with the civil society, motivation for their work, methods they use and difficulties they face. In additional it also deals with the role of churchmanship. The theoretical part of this work describes how the activity of prison chaplains is embedded in the legislature of the Czech Republic and their connection to the civil society through the non-governmental organization Vězeňská duchovenská péče (Prison spiritual ministry). In this part motivation from the psychological point of view is also described. From this part the specific analysis and observation of the motivation for social and pastoral activities, over confessional and ecumenical characteristic of the NGO Vězeňská duchovenská péče derives. The practical part of the thesis describes the research carried out with 17 prison chaplains from 11 prisons and its results. The motivation of prison chaplains relates to their goals. The major finding is that there is their intent to offer help and support to people that find themselves in very difficult situations: human help (closeness, giving an ear and encouragement), and spiritual help (sacraments and prayers). Author's name: Vendula Stará Year: 2009 Abstract: Photodynamic therapy (PDT) is a new significant treatment modality for several diseases, most notably cancer. Light, oxygen and photosensitizing drug (PS) are combined to produce a selective therapeutic effect. An important breakthrough in PDT is searching for a new PS, modification of their photophysical and optical properties. In this my diploma thesis I synthesized butyl 2,3-dicyanoquinoxaline-6-karboxylate acid by esterification of carboxyl group through its chloride. I used this butylester to synthesize the symmetric and asymmetric AzaPc. The synthesis of the asymmetric AzaPc with one carboxy group in the structure was not possible, because the separation of A3B congener was strongly adsorbed on silica. Esterification of the carboxy group did not lead to better results. Further, I synthesized symmetric AzaPc with butyloxycarbonyl group on the periphery of makrocycle. I prepared zinc (II) dye from its metal-free derivative which was prepared from magnesium (II) analog.

Ordinal numbers in contemporary Czech

The thesis "Ordinal Numbers in Contemporary Czech" characterizes and defines Czech ordinal numbers on the basis of theoretical conceptions of Czech grammar books and linguistic handbooks, and using data from the Czech National Corpus database it examines variant and problematic phenomena in this category. It also deals with ordinal numbers in an adverbial form, with words derived from ordinal numbers and with idioms, part of which the ordinal numbers are. It also analyses the issue of forming, writing and orthography of variant ordinals.

The Concept of Science According to Paul Karl Feyerabend

The approach to science to Paul KarI Feyerabend follows this approximate model: The pragmatic goal is to find the solution of a problem. Based on a concrete situation, we elaborate on a random hypothesis which can solve. the problem, and because of that, we expand our knowledge. The approach to the choice of hypotheses or theories cannot be restricted by the scientific point of view, nor by scientific method or scientific demarcation. We are left to elaborate, and let alternative hypotheses compete with each other. Feyerabend provided evidence that this is free approach to the extetion of knowledge more effectively defends against rigid exclusivity, and indoctrination of ideology. Reality is more uncomprehensively rich and methodology binds. In this work a view of the world in which we are now living is presented. It is an approach to the world derived from history. It is about Relativism. This view is presented by the life work of the Austrian professor of philosophy Paul KarI Feyerabend.
