Lifelong learning programme of educators in kindegartens within the Znojmo region.

The subject of the thesis is to realize on which conditions the further education of pedagogical staff is based in Znojmo region. Theoretical part is focused to the current legal framework ruling the professional readiness of pre-school teachers and i have outlined some of the possible options in further education of pedagogical staff. Second part is focused on unique factors and issues of Znojmo region, numbers and sizes of kindergartens and transportation situation in the region.In the third, practical part of my work i describe and analyze opinions of the teachers and director of respective kindergartens.

Rozdělení velkých genomů čeledi Triticeae tříděných chromozomů

Nuclear genomes of some Triticeae species are characterized by large size and prevalence of repetitive DNA sequences. These features hamper their physical mapping and gene cloning. Purification of individual chromosomes by flow cytometry can simplify these tasks by providing small and defined genome fractions. This lecture reviews the development of the methodology and its potential for genome mapping in barley, rye and wheat Due to small differences in relative DNA content, only one chromosome type can be discriminated and sorted in each of the three species. Cytogenetic stocks facilitate separation of other parts of the genomes as individual chromosomes, translocation chromosomes and chromosome arms. Chromosome analysis by flow cytometry permits quantitative detection of structural and numerical chromosome changes. Chromosomes sorted onto microscopic slides have been used for discovery of rare structural changes and for high-resolution cytogenetic mapping using FISH. The use of sorted chromosomes for HAPPY mapping, targeted isolation of low copy "genie" sequences, and high-throughput physical mapping of ESTs on microarrays are attractive options. As millions of chromosomes with intact DNA may be sorted, construction of BAC libraries is possible. Subgenomic, chromosome-specific and chromosome arm-specific BAC libraries have already been produced in wheat and represent unique resources for genomics of cereals.

Automatic Tool Changer

The thesis is focus on design solutions of automatic tool changer for CNC vertical milling machine SLV 300 in cooperation with the design company IMC/Engineering s. r. o. The main goal of theses is the creation of several design variants based on the critical research of similar machinery and the subsequent choice of the most appropriate solution. The assessment of the various options is done with amount of the changes caused on the existing design of the machine. It is essential the suitability of the solution from the perspective of the customer's company SolidVision s. r. o. Design solution is being designed for the possibility of a presentation device with unique features in its class, which is meant to be a simple maintenance free design with the possibility of easy service access For the chosen solution of the automatic tool changer are by calculation verified by the most exposed part of the device. In the thesis is also design documentation, for a better understand of the design solution.

Business Plan

This diploma thesis deals with the development of the business plan that will serve the company's management as a basis for implement the introduction of an entirely unique service. The theoretical part deals with the problems of small and medium businesses, subjects of the national economy and the demands of the business plan. The practical part deals with the identification and analysis of the selected company, judging the possible options of implementation of the business plan, including a tipping point.

Design of unique bicycle

The aim of this bachelor work is design and construction of chopper bicycle designed for promotional purposes. The first part is defining chopper bike and it's suitability for promotional purposes. The second part deals with the concept of solution options and selecting solutions for construction of bicycle. The final draft of construction and economic evaluation addresses the third part.

Pension income insurance position in the concept of pension reform

Pension income insurance is in the Czech market since 1994 and throughout its operation, the number of its clients increase. Its unique position is due to tax attractive and state contributions. Currently, on the market oprate 10 pension funds, which provides its clients with pension plans for 5 years and then can be saved as a single draw funds or annuity, according to the pension plan. The entire pension system is composed of two pillars PAYG and fading based , which are mutually complementary. Due to adverse demographic changes and financial sustainability of the system, it is necessary to reform the pension system. On the basis of economic expertise have been drawn up various options for changes in the voluntary pillar. Due to the necessary legislative changes, there was also a significant change in the pension income insurance scheme and its position was substantially changed. The voluntary nature of pension obligations have been replaced and now have the ability to save clients money into funds with different investment strategies. "Old" pension scheme will operate in parallelly with the new system, but access to it is no longer possible, it is possible that the pension insurance with state contribution in the longer term expires. The implementation of the government will decide next month.

Drawing expression of autistic child

The presented thesis deals with drawing of children with autism. It is divided into two parts, the theoretical and the empirical. In the theoretical parts there are four chapters. Three chapters deal with problem of autism spectrum disorders, types of autism, options of diagnostics, frequency and possible causes of autism. The fourth chapter deals with basic information about children's drawing, unique manifestation of the autistic disablement in the children's drawing. In the empirical part there are analysed 12 autistic children's drawings. Results are gained on the basis of observation drawing and evaluation by predetermined criterias. The goal of the bachelor thesis is to find out how the typical characteristics of autism displays itself in the pictures.

Environmental Scanning Electron Microscopy and its Application Possibilites in ISI ASCR

The first commercially available environmental scanning electron microscope (ESEM) was introduced in 1988 by Dr. G.D. Danilat and his company Electro Scan. Prof. Autrata and Doc. Jirák of the Institute of Scientific Instruments of the Academy of Science of the Czech Republic, public research institution, and the Brno University of Technology launched a laboratory operation with the first purely Czech ESEM AQUASEM in 1995. The Team of Environmental Electron Microscopy (EEM), headed by Dr. Vilém Neděla, a former student of Professor Autrata, has continued the nearly twenty-year tradition of ESEM progress in the Czech Republic. The team has studied interactions of electrons with high-pressure gas environments, designed, developed and simulated detection systems for SEM and ESEM and performed simulations of gas flows in ESEM. In interdisciplinary cooperation with various partners the team has developed and tested methods of observation of sensitive, native or live specimen studied under conditions of dynamic in-situ experiments using the today already obsolete ESEM AQUASEM II with directly heated tungsten cathode converted by Dr. Neděla at the Institute of Scientific Instruments of the Academy of Science of the Czech Republic still in his student years. In near future the institute plans purchase of a new ESEM with high resolution and a unique configuration of accessory analytical and other equipment. Thus a new laboratory of environmental electron microscopy with state-of-the-art equipment will be established at the ISI ASCR in Brno. The new laboratory will allow for specimen study with electron beam in combination with optional micro handling, dynamic in-situ experiments with specimen temperature variation from -25°C to 1000°C, or local gas and liquid injecting directly onto the sample.

Identification and characterization of transcription facotrs affecting late stages of male gametophyte development in Arabidopsis thaliana

Conclusions My thesis was devoted to the investigation of a complex regulatory network of transcription factors in the Arabidopsis thaliana male gametophyte. In addition to the abovementioned objective, the second aim of the thesis was the creation of a web-based transcriptome database and data mining tool characterized by an innovative concept of presentation and evaluation of the data from DNA chips. The results of my thesis can be summarized as follows: 1. We created a database called Arabidopsis Gene Family Profiler (arabidopsisGFP) (http://agfp.ueb.cas.cz/) which was made available on-line. This database was developed in order to provide a new tool for presentation of gene expression data (transcriptome), allowing the user to display the data in a quick and intuitive way using the innovative and when launched also unique concept of a virtual plant. The database employs a progressive idea "from simple to complex", but also provides the user with more traditional data presenting options. The aGFP database can serve as a reference manual for the expression of individual genes and gene families in Arabidopsis thaliana under physiological conditions at different phases of the life cycle and in individual organs and tissues or cell types. 2. Taxation of income of nonprofit organizations in the Czech Republic

The subject of this thesis is alternative approaches to taxation of nonprofit organizations. Interpretation of the Law on Income Tax is the taxation of nonprofit organization unique, and therefore my goal is to highlight the differences in tax liability in the case of different interpretations of this law, but also by using different methods or shift keying costs by activity among the main activities, if this option statutes authorize. All data used come from internal documents and accounting books of Prague's civic association, but because of the anonymization is not named. Evaluating all the alternatives can be considered that the ambiguous interpretation of the law may lead to different tax obligations and the law on income tax would be in this area was more specific.
