The role of evolutionarily conserved proteins BIR-1/Survivin and SKP-1 in the regulation of gene expression

SKIP and BIR/Survivin are evolutionarily conserved proteins. SKIP is a known transcription and splicing cofactor while BIR-1/Survivin regulates cell division, gene expression and development. Loss of function of C. elegans SKIP (SKP-1) and BIR-1 induces overlapping developmental phenotypes. In order to uncover the possible interactions of SKP-1 and BIR-1 on the protein level, we screened the complete C. elegans mRNA library using the yeast two-hybrid system. These experiments identified partially overlapping categories of proteins as SKP-1 and BIR-1 interactors. The interacting proteins included ribosomal proteins, transcription factors, translation factors and cytoskeletal and motor proteins suggesting involvement of the two studied proteins in multiple protein complexes. To visualize the effect of BIR-1 on the proteome of C. elegans we induced a short time pulse BIR-1 overexpression in synchronized L1 larvae. This led to a dramatic alteration of the whole proteome pattern indicating that BIR-1 alone has the capacity to alter the chromatographic profile of many target proteins including proteins found to be interactors in yeast two hybrid screens. The shape of the house is based on the traditional urban block development, respecting the existing arrangements surrounding blocks and visual axes across the territory. The proposal also takes into account the need to link the historic center with the developing territories over Koliště, which is currently barrier in the city. Connection with the planned southern center will be reached by footbridges and through staircases placed in the field to break the border Koliště. The height of the buildings respects the existing buildings, design of the building reaches a height of Morava Palace and the City Hall building, the second multifunctional building is one floor lower.

Analysis of the development of the prices of real estate

The aim of this diploma thesis is to analyze the development of the prices of real estate in the Czech Republic. The thesis is divided into three main parts. The first one deals with the theoretical introduction to valuation of the real estate. Moreover, the thesis presents the current development of the prices of real estate on the Czech market. The last part focuses on co-integration analysis, within which an ADL model is created. This model serves as a base for an error correction model, which describes short-term as well as long-term relations within the time series. The explanatory variables are gross domestic product, consumer price index, the amount of finished apartments, interest rate of mortgage loans, common rate of unemployment, and average gross monthly income. The rising sharing economy and its impact on tourism demand in Prague

This bachelor thesis analyses the impact of the sharing economy (Airbnb) and other selected determinants on tourism demand in Prague. Based on monthly panel data from the period January 2014 to March 2015 the econometric model was estimated. Determinants used in the model for analysis are Airbnb listings of short-term rentals, GDP per capita, relative price, weather temperature, rainfall, marketing and dummy variables summer, autumn and winter. Results from estimations show that Airbnb, the indicator of sharing economy, GDP per capita, which represents the income, relative price and Prague.eu, that represents the marketing component have significantly positive effect on tourism demand.

Applied analytics of Shared service centers

This thesis focuses on the development and implementation of data analysis in an environment of shared service centers. The aim is the selection of analysis and its implementation from raw data to visualization of the results. The work is divided into two parts. The first part describes the shared services centers, their risks and benefits, and the process of data analysis, including a description and examples of visualization and data quality. The second part of the work describes the development of selected analysis. We chose the analysis of vendor master file. We perform anonymization of production raw data using IBM SPSS Modeler. We are completing this process in Microsoft SQL Server Management Studio, where we are continuing with the development of analysis logic and finally assembling dashboards in Tableau visualization program.

Letters of Charles the Elder of Žerotín - patron of education

Annotation This work discourse on literary and agile correspondence life of Charles elder of Žerotín who financially and morally supported lots of gifted aristocrats in their way to the edducation. At the beginning the work contains information about life of Karel starší and about his own edducation. Further, there is a short essay about problems of epistolography and finaly, in the main part of qualifying text there is an analysis of particular letters. For the analysis those papers was chosen which refer to a culture and education in Žerotín´s age, mainly letters to teachers, schoolmasters and also Žerotín´s young relatives and wards. In the letters we can see not only a view of the edducated man (Žerotín), but also attitude towards culture and edducation in noble enviroment of 16th and 17th century. Main source of the work are translated Žerotín´s letters from edition of Noemi Rejchrtová: Z korespondence, Karel starší ze Žerotína. The letters are brought to the context with Charles´ life using the monografy of Tomáš Knoz: Karel st. Ze Žerotína, Don Quijote v labyrintu světa. This monography is the last contribution to the Žerotín´s age.

Design of search engine for modern needs

In this work I argue that field of text search has focused mostly on long text documents, but there is a growing need for efficient short text search, which has different user expectations. Due to this reduced data set size requirements different algorithmic techniques become more computationally affordable. The focus of this work is on approximate and prefix search and purely text based ranking methods, which are needed due to lower precision of text statistics on short text. A basic prototype search engine has been created using the researched techniques. Its capabilities were demonstrated on example search scenarios and the implementation was compared to two other open source systems representing currently recommended approaches for short text search problem. The results show feasibility of the implemented prototype regarding both user expectations and performance. Several options of future direction of the system are proposed.

The Overture to the First Version of the Opera King and Charcoal Burner

The issue of different versions of works of music is one that is discussed in the case of Antonín Dvořák (1841-1904), and in a certain sense it is connected with the theme "old and new", when an older, earlier, or first version becomes the basis for a newer one. This text focuses on a specific example of different versions, that of the overture to the first setting of Dvořák's opera King and Charcoal Burner from the year 1871. An overture which, as an integral part of the opera, was based on the musical material contained in the work, which eventually was never performed. And at the same time an overture that was the only part of this opera to "survive", was later transformed by the composer himself to a concert overture and thus became an independent work, while the opera's libretto was set to new music. This is in a nutshell the rather complicated genesis of the opera King and Charcoal Burner, during which the Concert Overture in F Major took shape as a new composition, created from the musical material of an old one.

Quality analysis of interpretative trails panels in Protected Landscape Area Český Kras

The aim of this thesis is to implement a qualitative analysis on interpretive trails, which are also considered part of local heritage interpretation. This analysis verifies if the authors of panels used recommendations published in two known methodologies during their preparation phase. This research is applied on eighteen interpretive trails in the protected landscape area of Český kras. In the practical part of this thesis, each interpretive trail has been evaluated by applying nine criteria. The outcome of the thesis is an evaluation of visuals and content composed on the interpretive trail panels for the purpose of attractive communication. The most significant deficiencies of the panels of interpretive trails in the protected landscape area of Český kras are quality of graphical design, whether the text communicates with the reader and if the text does not require linear reading. On the other hand, the most important strengths of interpretive trail panels are that the visual elements are always well connected to the content of the text on the panel and the text is easily read without specialised terms. The interpretive trail that has reached the best evaluation in the qualitative analysis is Vodácká NS Berounka. On the other hand, the trails with the lowest scores were NS Památné stromy Karlštejnska and NS Klonk-Suchomasty.
