Oralová, Iva ; Pavelka, Radim (advisor) ; Venzara, Jan (referee)
Title: Attitudes competitors kickboxing coach to lead the way during the match Objectives: The main objective of this study is to examine how athletes perceive their coach in leadership (coaching) during the match, whether this behavior coincides with their preferences and what attitudes they have. Furthermore, these results are interpreted as a feedback to coach who lead a match to the increase effectiveness of its leadership and also show that perception during the match and athletes's preferences are very individual and different. And therefore coaches should not follow just generalized research into coaching but they should work with each athlete individually and according to theirs specific needs. Methods: In this thesis were used a method of structured questionnaire and semiguided interview. The structured questionnaire was used in the detecting method of coach leading during the match and preferences of participants. The semiguided interview method was used to specify preferences and attitudes towards a method of coach leadership. Results: It was find out It was found that the differences between preferences and the perceived reality occur not only for individual athletes, but also between them. It turned out that there is some consensus, as in the perception of coach's behavior during the...

Milk as potencial source Encephalitozoon cuniculi
TOMANOVÁ, Vendula
The object of this thesis is to find out an incidence and prevalence of Microsporidia infection in uncooked cow´s milk. Samples of milk, faeces and urine from 147 milk cows were screened. One of them was positive for presence of Encephalitozoon cuniculi in sample of milk and stool repeatedly. Theoretic part includes information about Microsporidia, e.g. developmental cycle, transfer and course of disease, treatment and prevention. There is a description of heat treatment of milk as well. A methodical procedure of microsporidia DNA detection from samples of milk, feaces or urine is described in the practical part. The thesis is focused on experimental verification of sterilizing capabilities of various pasteurization methods. Milk was contaminated by Encephalitozoon cuniculi and then heat treatment according to the methods used in practice followed. Contaminated milk was offered to laboratory immunodeficient SCID mice. After three weeks mice were put to death and their organs were examined for the presence of Microsporidia DNA using molecular methods. Finding out of Microsporidia in examined organs was the evidence of insuffient heat treatement.

The development of tax collection from personal income tax in the Czech Republic
Šubová, Lucie ; Láchová, Lenka (advisor) ; Kouba, Tomáš (referee)
This diploma thesis analyses the development of tax collection from personal income tax in the Czech Republic from 2000 to 2012. The first chapter describes the tax reforms that took place in the period in the Czech Republic. Furthermore, it describes the relationship of collection and GDP. In the second chapter I analyse the PIT collection as whole. By regression and correlation analysis I shall identify the relationship and the degree of dependence between collection and different variables such as GDP, the number of taxpayers, unemployment and sub-collections. In the third chapter, the PIT collection is divided into three sub-collections -- levied on tax returns, from employment and withholding. Each of the sub-collection is analysed separately. I describe factors that in a given year or in group of years the most influenced their size. By regression and correlation analysis I shall identify the relationship and the degree of dependence between the sub-collections and GDP. Likewise, I shall identify the relationship and the degree of dependence between the sub-collections each other. In the last chapter the PIT collection is divided by individual financial authorities, ie by region. The development of tax collection in each region is analysed and by regression and correlation analysis is detected the relationship and the degree of dependence between the regional collections and GDP.

Opinions of a woman with a child in prison.
TAMPÍROVÁ, Jana
The work is focused on the prison system - specifically women bringing up their child / children in prison. This specialized department is located in Svetla nad Sazavou and its function is executed since 2002, only in the CR. The department for mothers with children is part of a female prison in Světla nad Sázavou. The aim of this work is to detected the opinion by citizens living in Budějovice on this issue. If they know about this specialist departments in our CR and where are located. Whether citizens agree with this eduaction or not, if they believe a child's stay in prison to be harmful for its further development, whether it should have imprisoned a woman to raise her child in prison for the duration of his sentence, etc. I will use a public inquiry as a research method, and results will be used to evaluation to my hypotheses.

Dust-UV interaction
Nouzák, Libor ; Pavlů, Jiří (advisor) ; Žilavý, Peter (referee)
Title: Dust - UV interaction Author: Libor Nouzak Department: Department of Surface and Plasma Science Supervisor: RNDr. Jiri Pavlu, Ph.D. Supervisor's e-mail address: jiri.pavlu@mff.cuni.cz Abstract: Micrometer objects (dust grains) are an integral part of the universe. As other objects in the plasma, these dust grains charge to potencial close to the potencial of the plasmas (in this case, interplanetary plasma). In the universe, the photoelectric charging dust grain dominantes all other charging processes. In general, the resulting charge of dust grain is given by a balance of all processes, which haven't been mostly jet theoretically described. In our laboratory, we are simulating space conditions and measure resulting charge and his changes on a single separated dust grain. This work is partly focused to UV source application and to finishing its electronics, and partly on test measurements and model calculations, connected with newly built experiment (e.g., to estimate effects of backgound currents, surfaces, and the geometry of the dust trap electrodes). The work is finished by the first measurements of glass grain charge under electron bombardment with provisional detection optics. Obtained results are compared with previous measurements on the same type of dust grains. Key words: dust, dusty plasma,...

Proteomic analysis of lysozyme and lysozyme-like proteins of synanthropic mites
Chum, Tomáš ; Erban, Tomáš (advisor) ; Mikeš, Libor (referee)
This diploma thesis was focused on the study of lysozyme and lysozyme-like proteins, either of similar function (antibacterial) or molecular weight (14 - 17 kDa), of synanthropic acaroid mites. In general, animals utilize lysozymes for defensive (antimicrobial) or digestive purposes but also as a digestive enzyme. Some chitinases or other enzymes that act similarly to lysozyme can be utilized for similar purposes. Chitinases belong to house dust mite allergens. One of major mite are historically named lysozyme-like proteins which name relates to their size similar to lysozyme. Bacteriolytic activity has also 14.5 kDa (UniprotKB Q8MWR6) protein. The species selected for the study were domestic mites Dermatophagoides farinae, D. pteronyssinus and Lepidoglyphus destructor. Presence of lysozyme was detected by direct detection with polyclonal antibody using immunohistochemistry and dot blots. Immunohistochemistry proved presence of lysozyme epitopes in the feces of D. farinae, D pteronyssinus a L. destructor. Dot blot analysis demonstrated the presence of imunoreactivity of antibody in spent growth medium extracts (SGME) of all three species. This implies that lysozyme is synthesized in the midgut. The presence of lysozyme and lysozyme-like proteins was proved using 2D electrophoresis and MALDI TOF/TOF...

Krajíček, Jan ; Svobodová, Eva (referee) ; Bosáková, Zuzana (advisor)
Pterines belong to an important group of compounds, acting as pigments in many species. Some of them are probably responsible for characteristic coloration of the insect group (Hereroptera). This coloration is considered to be a visual warning signal for optically orientating predators (birds, lizards etc.). In this work, pterines in the species of Graphosoma semipunctatum have been investigated by high performance liquid chromatography. To develope an appropriate separation method, the reverse-phase separation mode with C18 stationary phase (Spherisorb ODS 2) and binary mobile phase (organic modifier/buffer or water) was used. The effect of type and content of organic modifiers (methanol, ethanol, tetrahydrofuran) and concentration of phosphate buffer pH 3.0 (10 - 30 mM) in the mobile phases on retention and separation behavior of the studied pterines (leukopterin, biopterin, xanthopterin, isoxanthopterin and erythropterin) was studied. Under the optimized separation conditions (5/95 (v/v) methanol/20 mM phosphate buffer, pH 3.0, flow rate 0.7 ml.min-1 , UV detection at 290 nm), the extract from the integument of Graphosoma semipunctatum was analyzed.

Schmied, Marten ; Štěrbová, Ludmila (advisor) ; Mevenkamp, Nils (referee)
Public healthcare provision is under increasing economic restraints making an efficient and sustainable managerial planning a necessity in the hospital sector. Business Intelligence is the extraction of business relevant knowledge in order to adjust and refine executive operations. On the free market industries have applied the according methods and software dedicated to the generation of Business Intelligence is offered by variety of companies. Data Mining, furthermore, describes the facilitation of algorithms in order to train programs to detect unseen patterns from huge amounts of data. Therefore mining techniques are suitable for adding to the business relevant knowledge, particularly as they can produce more accurate predictions. The thesis examined the status of Information Technology normally utilized in Austrian hospitals and simultaneously identified studies that apply Data Mining to a Hospital Information System to gain Business Intelligence. While the general level of Austrian Hospital Information Systems is well sophisticated, common challenges are present in a separation between clinical and administrative systems and their interfaces. For the Data Mining a majority of studies aims at medical improvements. Some applications were found to have good business relevant prospects but their feasible introduction into the practice needs additional fostering.
