Analysis of artificial regeneration methods of silver fir (Abies alba Mill.) in the selected forest stands of the Forest district Lužná.

This diploma thesis is dealing with analysis and statistical evaluation of regeneration methods introducing silver fir to forests in the area LS Lužná, forest district Bor. The aim of this thesis was to make the measurements of silver fir plants growing in the existing permanent research plots, to establish new research plots and perform general evaluation of alternative regeneration approaches. The theoretical part deals with the issue of silver fir regeneration, its expansion and areal, ecological requirements, usability and physiological description of this species were also taken into consideration. This part of the thesis was also focused on description of the various alternative ways of reintroducing this species to the forest. It was made a description of the principles of forest tending, natural and artificial regeneration of silver fir stands with respect to its environmental requirements. The biotic and abiotic pests of silver fir and effective defence against these pests were also discussed. It was made the analyses of factors affecting positively and negatively the growth and development of silver fir. Silver fir was described as an ameliorative and stabilizing tree species. In the practical part of this thesis an analysis of local conditions on the ground Bor, LS Lužná; specifically analysis of climate, soil, light and growth conditions was performed. They were recorded and compared to findings on the permanent research plots (established in 2014) and newly established research plots. The results were evaluated by different growth conditions on the research and permanent research plots. The differences in growth conditions in the different regeneration variants were evaluated statistically.

The impact of the Swiss-EU relationships on the Swiss banking secrecy

Switzerland and the European Union share common values and have peaceful and well functioning economic and political agreements. Nevertheless, the Swiss banking secrecy is definitely a thorn in the EU's flesh, especially because of tax fraud or money laundering. Since the existence of the Swiss banking secrecy it has been associated with holocaust money, tax evasion, potentates' money etc. and also has been confronted with espionage attacks by foreign authorities and various other disputes. Recent global events have shed light on the Swiss banking secrecy's wider international agenda. A newly debate about the Swiss banking secrecy started with the global economic crisis and the Euro crisis. Finally, not a day goes by where there is no report or article in the media regarding the banking secrecy, the Swiss banks and its issues concerning the European Union or the United States. This writing focuses on the relationship between Switzerland and the European Union which is affected by Swiss banking secrecy: By explaining the history and importance of the Switzerland as a financial centre, its banking secrecy and the differences to other systems, possible issues and conflicts can be outlined. Biogenesis of Photosystem II in the Model Cyanobacterium Synechocystis sp. PCC 6803 - The Role of Selected Auxiliary Protein Factors and Subcellular Localisation

This thesis explores localisations and roles of three auxiliary protein factors involved in the biogenesis of Photosystem II (PSII) in the cyanobacterium Synechocystis sp. PCC 6803 and contributes to subcellular localisation of the initial steps of PSII biogenesis and repair-related D1 synthesis. The main results consist in i) identification of a functional interaction of the protein factor Psb27 with a lumenal domain of the Photosystem II subunit CP43, ii) discovery of a novel pigment binding complex formed by the Ycf39 protein and high-light-inducible proteins implicated in photoprotection and delivery of recycled chlorophyll to newly synthesized D1 protein during the PSII reaction centre formation, iii) providing evidence that the early steps of PSII assembly and the repair-related D1 synthesis occur in the thylakoid membrane of Synechocystis, and iv) revealing that the cyanobacterial PsbP orthologue, CyanoP, assists in the early phase of PSII biogenesis as an assembly factor facilitating the association of D2 and D1 assembly modules.

HOUSE ON THE EDGE – Valašské Meziříčí, Sokolská Street

Modern and lightweight-looking architectural concept of the town house, which is the counterbalance to the former historical development and highlight of the look-through axe from the Main square. The inner-space functional allocation come through to the exterior look of the building. The former Poláškova street is extended, there is the piazetta with outside seating. The parter is determined by the roof cantilever and the wooden pergola with the outdoor LED illumination. In the ground floor there are two shopping spaces, pharmacy and film café. The first floor contains rehab centre. Its look is maximally lightweighted, the fassade is made only from the structural glassing. This makes the contrast between the ground floor growing up from the ground a the three-storeyed section for living. This part of building is carried by massive frames from reinforced concrete and is designed as a wood framework with pasted frames and gridiron ceiling pannels. It contains 28 appartments. Most of them are low-cost, epecially for young families and singles. In the underground are three storeys of parking. 81 places are the replacement of former public parking. The arrival ramp is situated on the south side by the Sokolská street. The connection point to the busy drive-through road is on the former crossroad with the Králova street, where are newly installed the traffic lights. The access to the shops is situated on the north side by the historical town centre. To cross the parcel is possible next to the building through the small park on the western side, or throught the centre of the bulding. The terrace above the ground floor is walkable and accessible by the stairs from piazetta or by the skew roof of the film café. The skew roof contains also benches and artificial waterfall, which ends in the fountain. The fountain is there for the good feeling of the space and for technical reasins. It is connected with the reservoir of the rain water.

Mathey delineavit. The tombstone of Count Humprecht Jan Černín of Chudenice in light of the written sources (1683–1688)

The tombstone of Count Humprecht Jan Černín in the chapel of St Sigismund in the cathedral of St Vitus is one of the unjustly forgotten sepulchral monument of the 17th century in Bohemia. Based on so-far unpublished correspondence from 1683 until 1688, it is possible not only to document the complicated course of the building of this monument but also to determine the designer of the monument who is the prominent architect active in Bohemia in the 1670s-90s Jean Baptiste Mathey. Some hitherto unknown Mathey´s works are newly published here – the altar retables in Stará Boleslav (Altbunzlau) and in Litvínov (Leutensdorf), also the graphic portrait of the prioress of the Discalced Carmelites in Prague´s Malá Strana the venerable Maria Electa à Jesu.

Renovation of municipality office

This thesis deals with the reconstruction of the Adamov municipality,based on a solution of barrier-free building use, overall improvement of the energy performance of the building and creation of new office spaces in the form of a loft. The object is located on a building site in the town Adamov. The object is currently connected to a III. road, parking space and utilities. It is a building with three above-ground floors and one basement floor. The ground plan of the municipality is rectangle-shaped, the maximum outline dimensions of the rectangle are 28.38 x 16.70m. Existing vertical supporting constructions are mason made of fired bricks. The roof frame structure is made of wood, construction framework stagnant stool, fired tiles are used as roofing. Floor surfaces are formed by ceramic tiles or vinyl. The entrance part is covered with a flat roof. The entrance area will be redesigned as a barrier-free, with a connection to the elevator that goes through the entire object. The sanitary facilities in the first and in the second floor will be renovated. Newly created attic floor will be – except for the lift - connected to the interior of the building by a new steel staircase and the walls on this floor will be of a light type. Holes fillings will be changed in the perimeter walls. A thermal insulation of the entire object by a contact heating system will take place. This thesis consists of preparatory work and studies, implementing documentation, text reports, calculations of the thermal technical assessment and fire safety solutions. Project documentation is made in accordance with currently valid laws, regulations and standards.

Lodging house for cyclists

The subject of this thesis is a design of a newly built guesthouse on a bicycle path in the village Spálené Poříčí. The building is designed in a gently sloping terrain towards the west. The main entrance faces the south. It is an object without a basement and with two above ground floors. The structure of the building is a transverse wall system made of ceramic fittings. Other walls are also made of ceramic fittings. The entire building is based upon the stripe foundations and footings. Pension is covered in the part of the accommodation and in the restaurant part with a gable roof. For the restaurant, a terrace is designed on the west side of the building. In this thesis, static and building equipments are solved. The building is also evaluated in the terms of lighting, thermal, acoustic requirements and the fire safety of building is also solved.

Visual Art in the Housing Estates of Brno (1945–1989)

With special attention being paid to the development in the city of Brno, the dissertation focuses on the issue of applying artworks in Czechoslovak architecture after World War II. The main research questions – Why were in Czechoslovakia after 1945 artworks again integrated in architecture and what strategies were applied in the case of the newly built housing estates in the city of Brno? – are answered in two successive sections. First, the institutional background for application of art in architecture in the light of the events in Brno (1945–1993) is examined; the closing part is devoted to three case studies that illustrate local development in the period of socialist realism, creative release of the1960s and political normalization. The aim is to highlight the complexity of the examined subject and refute a popularly handed down conviction that integration of artworks into architecture in the period of socialism was motivated exclusively by the communist propaganda, and to show that the attempts to create a synthesis of art and architecture were also based both on earlier theoretical considerations about the educational function of art and its effect on humans and the need to improve poor economic situation of artists after World War II.

Hidden diversity and evolutionary trends in malacosporean parasites (Cnidaria: Myxozoa) identified using molecular phylogenetics

Malacosporeans represent only a minor part of the huge myxozoan diversity, accounting for more than 2000 species. In contrast to the remainder of the Myxozoa, malacosporeans cycle between vertebrate (fish) and invertebrate (bryozoans) hosts and are restricted to freshwater habitats. The present study applies light microscopy and molecular methods to scrutinize the morphology, prevalence, diversity, distribution as well as host and habitat preference of malacosporeans in fish hosts. Comprehensive phylogenetic analyses based on newly obtained malacosporean SSU rDNA sequences and those available in GenBank reveal important evolutionary trends in this group. The significant increase in malacosporean species richness unveiled in the present study points to a hidden biodiversity. Reasons for this cryptic species diversity, the potential existence of malacosporean life cycles in marine environment as well as the evolution of worm- and sac-like morphology in bryozoan host are discussed.

Study of Interspecies Differences in Plasma Protein Binding of New Radiopharmaceuticals

Study of interspecies differences in plasma protein binding of new radiopharmaceuticals Abstract This paper engages in a study on the plasma protein binding of chosen radiopharmaceuticals from the perspective of interspecies comparison. An equlibrium dialysis at 37řC was employed for the plasma protein binding determination and experiments were implemented on human, bovine, rabbit and rat plasma. Studies on the plasma protein binding were carried out with three new radiolabelled chelates and clinically routinely used bifunctional chelate DTPA radiolabelled with 99mTc (99mTc-DTPA). Binding was determined for the newly bifunctional chelate 90Y-DTPA-oxn and for the two potencional peptide based radiopharmaceuticals: 177Lu-DOTA-TATE a 177Lu-DOTA-MG-1. The results received were evaluated in light of interspecies comparison and compared with the results of the plasma protein binding of a clinically routinely used 99mTc-DTPA. Measuring data were analyzed using statistic data processing. The results of this analysis showed that statistically significant difference is for the complex 99mTc-DTPA between human and bovine and also between human and rabit plasma at the level of significance alfa 0,05.
