Comparing the Price of a Transportation Structure and the Actual Costs Incurred in Various Stages of Construction

This diploma thesis deals with the calculation of the cost of the building work in order to create the price, which at the stage of the contract becomes a selling price, and the costs are fixed as a budget. The subject of this work is to monitor the development of costs in time so that the cost does not exceed the budget and the construction did not get into the negative result. Then set the price by its own methods (item budget, calculation according to budget indicators) and compare the prices and costs thus obtained from different phases of construction between each other and determine the differences. In this work is monitored and compared the cost of a construction contract and its costs in the construction stages. In the practical part,is used the method of direct comparison of cost and price values over time, in the form of different outputs from the controlling program and outputs from compiled item budget and the calculation of the construction. Analyzing the differences between these costs and their evolution over time by more detailed examination of item budgets and cost calculations, they determine the origin of these deviations. In conclusion, is proposed measures for a particular transport structure whitch was handled by the author as"master and co-ordinator of the construction".

Formal Systems Based on Automata and Grammars

Tyto teze navazují na studium gramatických a automatových systémů. Na začátku, práce pojednává o regulárně řízených CD gramatických systémech využívající frázově strukturované gramatiky jako komponenty. Do systémů jsou zavedena tři nová omezení na derivacích a je studován jejich vliv na vyjadřovací sílu těchto systémů. Poté, tato práce definuje dva automatové protějšky ke kanonickým multi-generatiním nonterminálem a pravi\-dlově synchronizovyným gramatickým systemům, generujících vektory řetězců, a ukazuje, že všechny tyto vyšetřované systemy si jsou vzájemně ekvivalentní. Dále táto práce tyto systémy zobecňuje a zakládá fundamentalní hierarchii n-jazyků (množin n-tic řetězců). V~souvislosti se zavedenými systémy tyto teze zavádí automatově-gramatický převodník založený na konečném automatu a bezkontextové gramatice. Tento převodník je pak studovaný a použitý jako nástroj přímého překladu. V~poslední části jsou v této práci zavedené automatové systémy jádrem pársovací metody založené na stromově řízených gramatikách s n omezenými cestami.

Methodology of highly reliable systems design

In the thesis, a methodology alternative to existing methods of digital systems design with increased dependability implemented into FPGA is presented, new features which can be used in the implementation and testing of these systems are demonstrated. The research is based on the use of FPGA partial dynamic reconfiguration for the design of fault tolerant systems. In these applications, the partial dynamic reconfiguration can be used as a mechanism to correct the fault and recover the system after the fault occurrence. First, the general principles of diagnostics, testing and digital systems dependability are presented including a brief description of FPGA components and their architectures. Next, a survey of currently used methods and techniques used for the design and implementation of fault tolerant systems into FPGA is described, especially the methods used for fault detection and localization, their correction, together with the principles of evaluating fault tolerant systems design quality.  The most important part of the thesis is seen in the description of the design methodology, implementation and testing of fault tolerant systems implemented into FPGAs which uses SRAMs as the configuration memory. First, the methodology of developing and automated checker components design for digital systems and communication protocols is presented. Then, a reference architecture of a dependable system implemented into FPGA is demonstrated including several fault tolerant architectures based on the use of partial dynamic reconfiguration as the mechanism of fault correction and the recovery from it. The principles of controlling the reconfiguration process are described together with the description of the test platform which allows to test and verify the design of fault tolerant systems based on the methodology presented in the thesis. The experimental results and the contribution of the thesis are discussed in the conclusions.

Surface modification of glass fibers for polymer composites

The doctoral thesis is aimed at preparation of glass fiber reinforced polymer composites with controlled interphase formed by plasma-polymerized tetravinylsilane and tetravinylsilane/oxygen thin films. The thin polymer films of specific physico-chemical properties and thickness were deposited to improve interfacial adhesion of glass fiber/polyester composites. The fiber surface modification was performed by using plasma enhanced chemical vapor deposition in low-temperature RF plasma operating in an various effective power range and different treatment time. Test results were examined in relation to the interlayer thickness and different treatment conditions. The prepared interlayers were analyzed to evaluate physico-chemical composition and properties (XPS, RBS, ERDA, FTIR and spectroscopic elipsometry). Selected mechanical properties were evaluated by AFM. Mechanical response of plasma interlayers was evaluated by short beam shear test and direct method of testing the interfacial shear strength using microindentation. The interphase shear failure was controlled by the shear strength at the interlayer/fiber interface as follows from experimental and model data.

Optical Measurement of Electromechanical Characteristics of Heart Cells

Dissertation is focused on the application of optical measurement methods using techniques of optical microscopy and fluorescence microscopy in measurements of electromechanical characteristics of isolated cardiac cells and clusters of differentiated cardiomyocytes. The first proposed method uses a practical combination of fluorescence microscopy equipped with fluorescent fast and high-resolution camera and atomic force microscopy for simultaneous measurement of calcium transients and contraction of cardiomyocyte clusters. The signals obtained undergoes filtration, processing and analysis. Result function parameters obtained by analyzing signals after application of caffeine are evaluated by comparison with functional parameters obtained during the control measurement. The second proposed method is applied to the cardiomyocyte clusters for the purpose of cardiomyocyte contraction signals measurement. The signals obtained by optical methods are analyzed and compared with the reference signal obtained using atomic force microscopy. Optical measurement method of cell contractins based on detection of cell ends using adjusting of microscopy images by re-sharpening and fluorescence method for cardiomyocyte contractions measurements were designed to increase realiability in simultaneous measurement of cell contractions simultaneously with calcium transients in isolated cardiomyocytes experiments.

Do Consumers Really Follow a Rule of Thumb? Three Thousand Estimates from 130 Studies Say “Probably Not”

We show that three factors combine to explain the mean excess sensitivity reported in studies estimating consumption Euler equations: the use of macro data, publication bias, and liquidity constraints. When micro data are used, publication bias is corrected for, and the households under examination do not face liquidity constraints, the literature implies no evidence for the excess sensitivity of consumption to income. Hence little remains for pure rule-of-thumb behavior. The results hold when we control for 45 additional variables reflecting the methods employed by researchers and use Bayesian model averaging to account for model uncertainty. The estimates of excess sensitivity are also systematically affected by the order of approximation of the Euler equation, the treatment of non-separability between consumption and leisure, and the choice of proxy for consumption.

Inhibice regenerace prýtů na explantátech petunie (Petunia x hybrida) in vitro způsobená fluoranthenem

The aim of thesis was to study the effect of fluoranthene (FLT) on growth and biochemical processes of plant Petunia (Petunia hybrida cv. Lady blue) in in vitro conditions. In this experiment were used segments of the middle parts of the leaves, shoots from organogenesis in vitro and apical meristem of stem with four to five leaves from long-term in vitro culture. It was observed if fluoranthene influence organogenesis of shoots in vitro, contents of chlorophyll a and b, content of cytokinins and rating of shoots (from organogenesis in vitro) and apical meristem of stem with four to five leaves. It was used culture MS medium containing benzylaminopurin (1.5 mg/l), 1-naphtylacetic acid (0.18 mg/l) and fluoranthene (1 mg/l) or without fluoranthene (control). In the case of organogenesis in vitro, the experiment was performed at 4 different variations of media with subculturing to other media in triplicate. Variations of the media were chosen to allow period of cultivation with fluoranthene, which affects growth and biochemical processes. MS medium containing indol-3-butyric acid (0.5 mg/l) and FLT in different concentrations (0.1, 1, 3 a 5 mg/l) was used for monitoring rooting. The results showed that fluoranthen is able to affect organogenesis of shoots formed on the segments of leaves. There was lower frequency compared to the control organogenesis. It was found that fluoranthene more affects beginning of cultivation compared to later exposure of fluoranthene during cultivation. It was found the highest uptake of benzyladenine and its riboside from the culture medium. Fluoranthene affected significantly content of dihydrozeatin. Zeatin and its riboside were recorded in a minimal amount. Fluoranthene caused lower content of chlorophylls a and b in explantats. In the case of chlorophyll a were no significant differences between influence by fluoranthene at the beginning of the cultivation or during of cultivation. It was noted that fluoranthene influence the morphology of the roots subcultured shoots from organogenesis in vitro. Plant roots exhibited less branching and roots were shorter in culture medium containing higher content of fluoranthene (3 and 5 mg/l).

ME AND HER (OR CREATING AN ACTING CHARACTER )

The thesis focuses mainly on getting to know myself as an actress. I describe my way of work on creating a character (Kačenka, Guests by Oliver Bukowski) using my four years experience on the theatre faculty. I focus mainly on the work with a text, director and acting partners. I am trying to express the moments which influenced my developement as an actor and determine how they affected my own creations. In regards with this I touch on the problematics of self-control. This thesis is mainly a self-reflection which arose from the need of working on myself in the field of acting as well as directing-dramaturgy which I currently study

Project management in selected company

This work deals with project management in chosen company. It has two parts. The first part is theoretical, which is focused to theoretical treatment of the subject. It explains what is the project, management of projects, organizations of projects, processes of project management, what is target of project, communication, control, changes and risks and more. In the practical part I focused to concrete company and project management and than I analyzed concrete project. Name of the mentioned company is OHL ŽS, a.s. It is company from Spain, which acts all over the world. In the Czech Republic has the base in Brno. This company has several divisions and every of them is focused to different professional architecture. I have chosen division of structural engineering. This division builds all kinds of buildings. It could be construction of new buildings or reconstruction. To analysis of chosen project I used various business documents, contracts with investor or suppliers, document which include billing, timetable. I especially used my experience with projects of this company, and I was part of this project. Name of this project is Reconstruction of cladding including replacement windows and insulation. During the analysis of project I found a couple major gaps. If we could avoid these gaps, we could increase profit. These graphs is especially in management. At the beginning there were appointed two temporary construction managers. They doesn't care about result project, because they knew that they will not be to the end of the project. These gaps are because there is missing motivation system. Project manager can´t affect results of employees. It doesn´t depend on performance of employees, they always has the same wages. For elimination of these gaps I proposed several suggestions for improvement. The first suggestion is modification and extension of motivation system. New motivation system include personal evaluation. This evaluation depends on performance of employee. The second part is bonus from over profit. The next suggestion is make a new working position. This position would take care about controlling and project documentation. During the building project, especially at the end is construction manager very busy. It depend of decision of construction 63 manager what kind of activity will have prefer. Project documentation and administration or controlling of building works. If they are doing everything correct and technology are respect. Suggested position would take care about invoices, real work, which is done, project documentation, controlling of budget. The third suggestion is purchase of small machinery such as chipping hammers and drills. At this time they rent all devices. Rent is higher than purchase price. The last suggestion is use credit to restoration of fleet. At this time they have obsolete fleet, because the bought all cars by cash. It is not conveniently.
