Creation and termination of employment

The thesis deals with the stipulation and termination of employment and other legal alternatives, known to the Czech Labour law. It explains in its theoretical part present legal form of employment. The wider attention is granted to the term of dependent work and fixed time employment. The practical part contains specifically formulated procedures of entering into contract of employment and its discharge. The thesis also prospects the shortcomings of current legal regulations and submits suggestions to changes on the basis of reflections de lege ferenda.
Species variability of bentic organisms in Dračice river classicifed by PERLA system

Examination ecological size selected part of river (from village Františkov to border with Austria) on the basis of field watching. Observed results are analysed by system PERLA. Method is based on analysing biodiversity makrozoobenthos sample from fordables running water. Sampling can be apply in running water, where water column not exceed 1 m and speed of water stream to 1 m/s. Taking is made by normal or low water stage.
Possibility of utilization of molecular methods for study of population genetics of noble crayfish Astacus astacus

The noble crayfish (Astacus astacus) is one of two native species of crayfish living in this country who are all over our country strictly protecte, because they are identified as critically endangered species. In Europe it is one of five native species of crayfish, reported by IUCN as an endangered species that needs protection management. Its population was dramatically reduced due to crayfish plagues, which carry non-native crayfish species from North America, who were introduced in the past in Europe and later to the Czech Republic. In the past, have been isolated and described microsatellite markers for crayfish (Koiv et al., 2008a). As part of this work was tested using the eight microsatellite markers on samples obtained from the czech population of crayfish. Testing was performed on 53 samples from six populations of crayfish. Test samples were subjected to isolation of DNA from tissues of the third walking legs using DNA Lego Kit. Then test isolate DNA electrophoresis on agarose gel. Testing the temperature cycle of PCR amplification based on the original publication Koiv et al., (2008a), followed by PCR cycles were adjusted according to the quality of PCR products obtained in our laboratory. Primer annealing temperature 60°C was chosen as the best for six tested loci i.e. Aas 3666, Aas 3115, Aas 790, Aas 1198, Aas 3950 and Aas 766, for two other loci Aas 2489 and Aas 3040 was chosen 55°C annealing temperature. The resulting PCR products were tested on agarose gel and subsequently fragment analysis on an automatic sequencer Beckman Coulter CEQ8000 determining the lenght of PCR products in multiplex consisting of several loci. In the individual loci were in our 53 samples found from 1 to 13 alleles ? one of the loci was monomorphic in all samples analyzed. The moravian population in the Boskovice tank showed the greatest variability, the average number of alleles per locus 3.86, then the north bohemian population from Jaroměř 3.43 alleles per locus. Zelenohorská population of 3 alleles at a locus and Světlohorská population of 2.57 alleles per locus. The lowest average number of alleles per locus had a population from Landštejn 2.43. The aim of this bachelor thesis was to develop a literature search of methods use in molecular biology studies of crayfish and also validate the use of eight microsatellite markers have described for the noble crayfish. In laboratory conditions were succesfully optimized using seven microsatellite markers from the eight described. These microsatellite markers should be used for population studies, or for determining parentage (paternity) in breeding experiments. The verification of the applicability of microsatellite markers have been evaluate and some fundamental characteristics of the population, usability testing and optimization of microsatellite markers, however, was done only on a very small number of samples, because these characteristics have only a very limited explanatory power.
Toxicity of selected substances for spiny-cheek crayfish

The thesis examined the toxicity of selected compounds for spiny-cheek crayfish (Orconectes limosus) in laboratory conditions. Tested the acute toxicity of the insecticide Diazinon 60 EC and herbicide Roundup Biaktiv. Tests were chosen to juveniles spiny-cheek crayfish (Orconectes limosus), and juvenile and adult specimens of signal crayfish (Pacifastacus leniusculus).These species of crayfish have been selected as an easily accessible model organism. Both species of crayfish spreading and in Europe they are unoriginal. Work focused on the observed toxicity of these substances on spiny-cheek crayfish and signal crayfish and the demands on the quality of the environment.
The potential use of PIT telemetry for identifying and tracking Spinycheek crayfish in their natural environment.

Main purpose of this diploma work was to find out possibility using PIT telemetry in monitoring strange crayfish species Spinycheek crayfish Orconectes Limosus. Research of the theme consisted of three steps: two laboratory researches and one research in the natural environment. The first laboratory study concerned tagging passive integrated transponder (PIT) to various part of crayfish body. PIT tags were internally implanted to cephalothorax, abdomen and claw. The highest mortality was in tagging to claw. The second laboratory research consisted of comparing different tagging methods to PIT telemetry. Crayfish were tagging by PIT telemetry, soldering, visible implant elastomers and visible implant Alpha tags. Among tagging groups there were not to find out different in mortality. In the third research the crayfish were tagging in their natural environment. There were using three methods: PIT telemetry, soldering and VI Alpha tags. This experiment wasn´t successful. During recapturing crayfish there were not to find any tagging crayfish.

Noble crayfish (Astacus astacus L.) is the night animal. Spiny-cheek crayfish (Orconectes limosus Raf.) is the most agresive species with more activity during the day, whose is of itself ecologic and reproductions characteristic typical r{--}strategist. And over than above it is a vector of crayfish plague. Objective of this study was the observe the daily and noctural of both species, the influence of fish predators and murmurs of agresivity. The bulk of this work was in ethologic laboratory with using videotechnic in control conditions and directed lightregimen. The findings see the big diference between species, that the spiny-cheek crayfish is more activated that the noble crayfish (51,4%{$\pm$}4,33 of the 24 hour activity) with slight diference between the day and night. Someones of the subjects don´t look for the cover during the experiment, it were mainly the females, whose activity were better than the male. Noble crayfish embodied low aktivity (15,1%{$\pm$}8,22 of the 24 hour activity) with more highly diference between the day and night and with slight diference between the sex. The inherence of the fish predator have had slight influence on the both species, rather sometimes take on the crayfish attention. The agresivity of the both is at least comparable, so it isn´t clear, that the spiny-cheek crayfish would be more successful in proximate competetion without the infection of crayfish plague.

The diploma thesis observed the indicators of growth in individual developmental stages of our native crayfish (Astacus astacus, Austropotamobius torrentium, Astacus leptodactylus) and non-native crayfish (Pacifastacus leniusculus, Orconectes limosus) under experimental conditions. The next aim of work was to describe the time of hatching of these species, the moulting interval, the frequency of moulting and increase after individual moulting during the first vegetation season in two different temperatures (15{$\rm^o$}C a 21{$\rm^o$}C).

Because the product is services, which are intouchable, impossible to test, it is more about good marketing.Communication with customer and continual contact with him is essential. In the thesis there was realised and assignemet of the marketing communication of the Image Lab s.r.o. company. From the results it is clear, that it is necesarry to monitor customers satisfaction and to find out their needs and wishes.

The subject of this bachelor work is homeworking, distantworking and virtual teams. The work defines these terms in their mutual context and shows their present usage. Weaknesses and gains of these constructs are analyzed to bring an objective view of modern work-methods. The work also presents the basic technologies, that relate to working from remote places. In addition are also included various specific methods of home-earning in the theme.
