Characterization of bacterial genes encoding DbtC-like extradiol dioxygenase with bioremediation potential for aromatic compounds in locality Hradčany
Šnajdrová, Renata ; Bořek Dohalská, Lucie (advisor) ; Nešvera, Jan (referee)
Aromatic pollutants pose a serious environmental problem. Petroleum and its derivates belong to the most abundant contaminants in the Czech Republic and their sanation is a priority objective for improving the life quality of the population. Bioremediation is a technology taking advantage of the natural capacity of soil and water microbial community to degrade environmental pollutants. Deeper understanding and detailed knowledge on specialized bacterial species, pathways and genes is required for selection, optimization and application of targeted bioremediation approach and for monitoring of its results. Recent analysis of a metagenomic library constructed from highly contaminated soil of the former military air-base Hradčany has identified a novel group of catabolic genes encoding extradiol dioxygenase similar to DbtC of Burkholderia sp. DBT1. The DbtC-like enzymes are among the three priority groups of extradiol dioxygenases with biodegradation relevance for the locality. The present study of soil bacterial isolates and metagenomic fosmid clones harboring the genes of interest gained evidence about the dbtC-like genes as a part of highly mobile gene cassette. Transposon insertion mutagenesis identified the genes joined with the expression of the extradiol dioxygenase activity. The dbtC-like genes were...

Corporate Performance Measurement
Pavlová, Petra ; Novotný, Ota (advisor) ; Krejčí, Jiří (referee)
This thesis deals with the application of Business Intelligence (BI) to support the corporate performance management in ISS Europe, spol. s r. o. This company provides licences and implements original software products as well as third-party software products. First, an analysis is conducted in the given company, which then serves as basis for the implementation of the BI solution that should be interconnected with the company strategies. The main goal is the implementation of a pilot BI solution to aid the monitoring and optimisation of corporate performance. Among secondary goals are the analysis of related concepts, business strategy analysis, strategic goals and systems identification and the proposition and implementation of a pilot BI solution. In its theoretical part, this thesis focuses on the analysis of concepts related to corporate performance and BI implementations and shortly describes the company together with its business strategy. The following practical part is based on the theoretical findings. An analysis of the company is carried out using the Balanced Scorecard (BSC) methodology, the result of which is depicted in a strategic map. This methodology is then supplemented by the Activity Based Costing (ABC) analytical method, which divides expenses according to assets. The results are informational data about which expenses are linked to handling individual developmental, implementational and operational demands for particular contracts. This is followed by an original proposition and the implementation of a BI solution which includes the creation of a Data Warehouse (DWH), designing Extract Transform and Load (ETL) and Online Analytical Processing (OLAP) systems and generating sample reports. The main contribution of this thesis is in providing the company management with an analysis of company data using a multidimensional perspective which can be used as basis for prompt and correct decision-making, realistic planning and performance and product optimisation.

Grísa, Tomáš ; Druckmüller, Miloslav (referee) ; Martišek, Dalibor (advisor)
The airborne hyperspectral remote sensing is used as an approach to monitor and analyse actual state of environmental components. This thesis deals with hyperspectral image data, especially it is focused on normalization with respect to scanning angle. The thesis proposes specific algorithm, which is based on the statistical analysis of spectral lines across the scan line and on a physical models describing the process of spectral reflectance. An important part of this thesis is software implementation of proposed algorithm, that allows to calculate required normalization for real datasets.

SMV-2014-22: Development of magnetic-resonance methodology for research into Parkinson’s disease in an animal model
Starčuk jr., Zenon
Collaborative development of an animal model of Parkinson’s disease, design and optimization of a suitable set of MR measurements, measurement of sample animals provided (40 mice, anatomy+diffusion), data adaptation for hand-over. The work was focused to the verification of a transgenic mouse model of Parkinson’s disease and the development of an optimal protocol for anatomic MRI of the mouse brain, with an emphasis on the regions of substantia nigra, hippocampus, striatum and on the comparison between the diagnostic values of DTI and DKI measurements in these regions. Experimental data were individually analyzed for each mouse and statistically evaluated in groups of test and control animals. Based on pathophysiology assessment further MR procedures were suggested for patophysiology research and the development of early diagnostics and therapy monitoring.

The Ethics of the Clinical research from the perspective of a Clinical research associate
Norková, Olga ; Haškovcová, Helena (advisor) ; Rohanová, Eva (referee)
Monitoring is one of the most important methods of overseeing the pursuit of the clinical research. A monitoring person (Clinical research associate) is responsible for verifying correctness of the monitoring process in the trial site and checks that the respect for dignity and rights of individual subjects exposed to the clinical research are being abided by. The theoretical part of the MA thesis examines the issue of the clinical research of medicinal products and explains the basic terminology pertaining to the performance of the clinical research. The thesis further analyses the position of Clinical research associate and describes the monitoring process. Concurrently, the ethical problems, which Clinical research associate encounters in practice, are traced. The empirical section studies the outcomes of an investigation conducted via a questionnaire, which focused on how Clinical research associate perceive the ethical problems. As a result, the study identifies the most frequently observed ethical issues. Respondents of the questionnaire view the ethical issues as less significant. Clinical research associates are not united in tagging the most frequent occurrence of ethical problems (in the field of medicine and the catchment area). In the same section, the MA thesis rebuts a hypothesis...

Concept of the revitalization of Nature Reserve Kapličky peat bog and continual monitoring of water regime and vegetation
PAVELCOVÁ, Lenka
The "Rašeliniště Kapličky" Nature Reserve is situated in the eastern part of the Šumava Mts. The reserve contains different peatland vegetation types - ombrogenous and transition bog parts. The area was partly drained due to extensive mining, pasturing and forestry in the past There are variously old drainage canals now. My thesis contains (i) the evaluation of groundwater table fluctuation and its relationship to vegetation composition and the drainage intensity and (ii) the evaluation of plant species composition in differently drained parts of the peatbog. There was strong relationship between actual groundwater table and precipitation, especially in drained areas, the diversity and the occurrence of expansive species were also related to groundwater table. A map of dranaige network inside the peatbog (1 : 3 500) and vegetation map (1 : 4 000) in the ArcMap programme for GIS environment were constructed. In the most drained part of the area the revitalisation measure was proposed {--} installation of wooden dams for blocking of drainage system.
