Studium vlivu půdních kondicionérů na růstové charakteristiky nově vysázených stromů

The theme of this thesis is to study the influence of soil conditioners on growth characteristics of newly planted trees. Thesis is the introduction of the theoretical part focuses on this topic. They included topics related to soil conditioners. The main basis for their use, basic soil properties and sorting improving products commercially available and their comparison, below are described using examples and experiences from scientific and academic experiments. In the first is a set of comprehensive knowledge on this topic. The second part is practical, about research. The survey examined the effect of three products (TerraCottem, Agrosil LR, Hydrogel) on the growth characteristics of newly planted trees over one year. They were evaluated results of this survey. At the conclusion is guided discussion of the theoretical and practical parts of a given conclusion about the use of these products in practice.
Temperature-sensitive hydrogels

Hydrogely jsou měkké látky složené z polymerních sítí nabotnalých ve vodě. Některé z nich při změně teploty reagují skokovou změnou objemu. Tento fázový přechod je označován jako kolaps a odpovídá konformační změně, při které polymerní řetězce přechází z konformace klubka do konformace globule. Vlastnostmi se hydrogely podobají živým tkáním (chrupavkám, šlachám a sval·m), nejen proto jsou perspektivními materiály ohledně aplikací v biotechnologiích a farmacii. Tato práce se zabývá studiem kolapsu v hydrogelech tvořenými jednoduchými a dvojitými sítěmi (speciální případ dvou interpenetrujících polymerních sítí). Připravené vzorky (PNIPAAm, PNNDEAAm, PAMPS, PAAm a odpovídající dvojité sítě s PAAm jako druhou složkou) byly zkoumány pomocí botnacích, DSC a NMR experiment·. Botnací experimenty prokázaly, že dvojité sítě vykazují větší míru nabotnání a také vyšší kritickou teplotu než příslušné sítě jednoduché. Pomocí NMR a DSC experiment· byla určena entalpie kolapsu. Bylo zjištěno, že na signálech NMR se podílí jen část polymerních jednotek teplotně citlivého hydrogelu kv·li velké hustotě sesí'ování. 1

Morphology, swelling and dynamic mechanical properties in broad frequency range of the model gels were tested. The vitrification of gels was found sensitive to microstructure of polymer network and to solvent content and to polymer-solvent interaction, but not to the macroscopic morphology of the gel. The low-frequency mechanical response was found sensitive to both polymer network properties and hydrogel morphology. Lightly crosslinked hydrogels showed relatively high mechanical losses in the rubbery plateau region, due to physical association of chains. Fused-particles type hydrogels revealed a weak secondary relaxation at low frequency, ascribed to motion of the dangling particles aggregates. Modification of hydrogels based on tyramine-modified hyaluronan

This thesis deals with the study of properties and use of TS-NaHY hydrogels. The first part describes the general characteristics of hydrogels and hyaluronic acid, its properties, synthesis, and utilization. The second part is focused on the experimental measurement of the fundamental properties TS-NaHY hydrogels. The aim of this study is to modify the hydrogels addition of surfactant micelles and create them in a domain capable of solubilizing a hydrophobic substance. The selected surfactants were examined for their solubility in water and in PBS solution, then hydrogels containing micelles of selected surfactants were prepared. The Addition of surfactant did not affect the mechanical properties of hydrogels. Furthermore, the release of the hydrophobic dye from the modified hydrogels into the PBS solution was investigated. Dye release is unlikely to occur at the time of study. The surfactant Tween 40 appears to be most suitable, especially for reasons of good aqueous solubility.
Releasing of the solubilized substances from the phase-separated hydrogels

This bachelor thesis is focused on releasing of the solubilized substances from the phase separeted hydrogels. The aim of this work is to determine the solubilization capacity of these hydrogels. Preparation of them were based on interaction between hyaluronan and cationic surfactant carbethopendecinium bromide. Hydrogels were prepared by two possible ways, „wet“ and „dry way“. For solubilization experiments were used fluorescein and acridine orange as fluorescent probes. Primarily UV-VIS spectrophotometry was used to determine if these probes are released form hydrogel or not. Hydrogels were monitored after 24, 48 and 72 hours from their preparation. But this method proved to be inappropriate and inaccurate, because of cloudy supernatants over the hydrogels and also because of very low concentrations of fluorescent probe in this solution. For these reasons instead of this metod was used method, which is based on fluorescence. The instrument is called MicroTime 200. It´s very accurate method and results of this are considered like relevant. It was proved that fluorescein as a negatively charged molecule is released less than acridine orange.
Soil hydrogels based on the principle of controlled release

This bachelor thesis is focused on soil hydrogels, which operate on the principle of controlled release. The aim of the research was on the release of substances, and the practical use of hydrogels in agricultural practice. At work, we focused on the effect of humic substances and the quantities of the substances of the fertilizer NPK of hydrogel. Querying this data will use a variety of methods such as pH-metry, conductometry, spectrophotometry and chromatography and knowledge not only of the analysis but also the macromolecular chemistry. Experiment it was found that the content of sodium lignohumate does not affect the rate of release of potassium and phosphorus into the soil. Hydrogel which is located above the quantity of NPK fertilizer and a certain number of lignohumate released from its structures much larger amounts of ions such as Hydrogel without fertilizer NPK with approximately the same amount of lignohumate
Study of barrier and transport properties of polyelectrolytes using diffusion techniques in hydrogels

This diploma thesis was focused on study of barrier and transport properties of selected polyelectrolytes in hydrogel matrices by using diffusion techniques. The study of these properties was performed in horizontal diffusion cells where is observed the change in diffusion probe concentration over time. Diffusion experiments were performed on an agarose hydrogel with the addition of alginate, hyaluronic acid, polystyrene sulfonate, humic acids and as a model probe rhodamine 6G was used. Important parts of this thesis are also the methods which characterize the substances and hydrogel matrices such as rheology and potentiometric titration. The main aim of this diploma thesis was to investigate the effect of interactions between passing model dye (rhodamine 6G) and the appropriate gel (agarose + polyelectrolyte) on the fundamental diffusion parameters (effective diffusion coefficient, lag time, etc.).
Preparation and characterization of hydrogels based on cationic biopolymer

Presented thesis deals with the preparation and characterization of hydrogels based on positively-charged biopolymer and negatively-charged tenside. As an appropriate representative of positively-charged polymer was chosen chitosan, because of his biocompatibility and biodegrability, and N,N,N-trimethylchitosan, which was synthetised from chitosan in this diploma thesis. The final product of synthesis was characterised by SEC-MALS, elemental analysis, infrared spectroscopy and nuclear magnetic rezonation. Interactions between these polymers and representatives of negatively-charged tensides, which are sodium dodecylsuphate and sodium tetradecylsulphate, were investigated. The main method used in this thesis was rheology, through it were investigated prepared hydrogels based on chitosan and tensides and also prepared microgels based on N,N,N-trimethylchitosan and tensides. The interactions between polymers and tensides have been proved.
Barrier properties of hydrogels

The main objective of this thesis is to judge the barrier properties of hydrogels. It was examined in total six kinds of agarose hydrogels, of which five kinds of gels was enriched with humic acids isolated from various sources. Humic acids were selected from IHSS standards, specifically Leonardite, Elliott Soil, Suwannee River, Pahokee Peat and Nordic Lake. Using UV-VIS spectrometry, copper ion concentrations were used to serve as a diffusion solution. Results of diffusion coefficients were evaluated by several diffusion methods. The method of diffusion in the plane of the board and three possible diffusion solutions of the constant source were used.
Diffusion of organic molecules in the hydrogel environment

This diploma thesis deals with study of hydrogels formed by phase separation of hyaluronan with oppositely charged surfactants cetyltrimethylammonium bromide (CTAB) and Septonex. It follows the bachelor thesis and extends the knowledge about the detailed characterisation of the inner environment of the hydrogel by determining the diffusion behaviour of the fluorescent probes Atto 488 and Nile Red using fluorescence correlation spectroscopy (FCS) technique and its modified version dual-focus fluorescence correlation spectroscopy (2f-FCS). Compared results showed that both methods show similar values and probes specifically interact with CTAB but Atto 488 shows only weak interaction with Septonex compared to Nile Red. Additionally, these interactions were not affected by the molecular weight of hyaluronan. In conclusion, it was recommended to measure this type of hydrogel in a small depth of gel using a conventional method.
