Expression and function of cellular prion protein in blood cells

The cellular prion protein (PrPc) is essential for pathogenesis of fatal neurodegenerative prion diseases. Recently reported four cases of vCJD transmission by blood transfusion raise concerns about the safety of blood products. Proper understanding of PrPc in blood is necessary for development of currently unavailable blood screening tests for prion diseases. Flow cytometry is an attractive method for prion detection, however, the reports on the quantity of PrPc on human blood cells are contradictory. We showed that the majority of PrPc in resting platelets is present in the intracellular pool and is localized in α-granules. We demostrated that both, human platelets and red blood cells (RBC) express significant amount of PrPc and thus may play an important role in the transmission of prions by blood transfusion. Our results suggest a unique modification of PrPc on human RBC. Such modification of pathological prion protein could distort the results of blood screening tests for prions. Further we showed that the storage of blood prior to analysis and the choice of anti-prion antibody greatly affect the detection of PrPc by flow cytometry and we identified platelet satellitism as a factor contributing to the heterogeneity of PrPc detection in blood cells. Link between inflammation and cancer

Chronic inflammation caused by many initiators can lead to a development of a tumor disease. Among these initiators, we found chronic infections as well as other biological, chemical or physical factors which have endogenous and exogenous origins as for example tobacco smoke, alcohol, radiation, obesity and others. The inflammatory response is orchestrated by immune system cells which contribute to a tumorigenesis by producing reactive oxygen and nitrogen species which harm cell structures, and by releasing cytokines - important mediators of inflammation - which increase cell proliferation and angiogenesis. But apart from higher risk of tumourigenesis due to chronic inflammation, the immune system cells also participate in tumor microenvironment formation. The main contributors are tumor associated macrophages, dendritic cells and T-cells. Besides other things, the complex tumor microenvironment is characterized by the presence of many inflammation mediators which assist in malignant cell proliferation, tumour progression and metastasis and angiogenesis. Changes in expression of immune-related genes after induction of inflammation in birds

Inflammation is one of the most important immunological mechanisms that protect animals from diseases. In my thesis I summarized information on changes in gene expression in tissues in the course of the inflammatory immune response in birds. I tried to describe the main features of the immune response in birds and highlight the differences to other vertebrates, especially mammals. I focused mainly on the surface molecules of various types of immune cells, as well as on signalling by soluble factors called cytokines. Although in general features are the inflammatory reactions in birds and mammals similar, in my thesis I have shown that a large portion of mediators and signalling molecules work very differently in these two vertebrate groups (e. g. the TNF SF-mediated pro-inflammatory signalling) and that substantial variation many be found even between different species of birds. Impact of Toll-like receptor 4 polymorphism on pro-inflammatory responsiveness in great tit (Parus major)

Toll-like receptor 4 (TLR4) belongs among chief bacteria-sensing Pattern recognition receptors. Endotoxin (lipopolysaccharide, LPS) recognition by TLR4 triggers signalling leading to release of cytokines that direct leukocyte infiltration into the inflammatory site and cause swelling. Effector mechanisms that ensure pathogen elimination include phagocytosis and oxidative burst. It has been repeatedly reported that the polymorphism in TLR4 may affect host resistance to various diseases. TLR4 may be, therefore, an important molecule in host-parasite co-evolution. Herein, I focused on TLR4 amino acid substitution Q549R which is associated with ornamentation in great tits. In tits I describe immune responsiveness to LPS stimulation on morphological and molecular level and examine effects of the Q549R substitution on inflammation and general body condition. In LPS- treated individuals I found decrease in heterophil-lymphocyte ratio (H/L) that might be caused by attraction of the blood-borne cells into the inflamed tissue. This is in striking contrast with increase in H/L in PBS-treated animals resulting from the stress response. Analysis of selected physiological parameters of athletes during exercise

Regular sport activity is considered as neccessary part of our lifes. Sport is reccomended for preventing various diseases, enhancing immunity system or for controlling weight and for enhancing conditions. However, how severaly is sport activity benefit for human health and when it becomes harmful for it? This work deals with possible negative effects of strenous excercise on human organism. In our case it is ultra-maraton and mountbiking. In following parts of this work it is tried to explaine metabolic changes and influance of strenous excercise on immunological and biochemical parametres. The parameters immunoglobulin A, M, leukocytes, creatinine, sodium and potassium ions and enzymes such as lactatedehydrogenase, creatinekinase and alanineaminotransferase were studied. The values of these parameters were obtained from the blood samples taken before and after the race. A 23 samples from a 24-hour cycling race and 24 run-of-the-run race samples per 100 km were examined. A matlab tool was created for statistical analysis of data. A huge increase in the number of leukocytes was found in both types of races. In the MTB, it is an increase from 5.75 ± 1.29 x 109 / l to 11.80 ± 3.08 x 109 / l, in the race from 4.99 ± 1.84 x 109 / l to 13, 20 ± 5.36 x 109 / l. IgA did not significantly decrease, on the contrary slightly increased. IgM values have not changed significantly in runners. Cyclists experienced a statistically significant reduction in the original amount of 60.23 ± 6.54 mg / dl to 52.60 ± 10.43 mg / dl. There were statistically significant changes in CK and LDH levels in the race. From the original CK values of 4.25 ± 2.74 kat / l, the activity of the enzyme increased by 50.09 ± 60.08 kat / l due to the race. LDH is not significantly different in the cycling race as well as in the ALT enzyme. In this case, there was no statistically significant change to either type of race. Concentration of potassium and sodium ions decreased significantly. In potassium, its concentration dropped from 5.61 ± 0.59 mmol / l to 4.57 ± 0.39 mmol / l. Sodium ions dropped from 138.22 ± 1.17 mmol / l to 137.00 ± 1, 75 mmol / l.
Verification of practical possibilities to strengthen the immune system in women

The topic of this final bachelor work is immune system which comprehensively describes its function and factors that influence it. The theoretical part deals with function of the immune system, introduces its cells and organs which are split synoptically. Further on, the theoretical part describes factors with influence on the function of the immune system. These are factors which reinforce but also weaken immune system. Every factor in this work is explained with its influence on immune system and it also describes the signs of its absence or surplus in human's health. Practical part of the bachelor work deals with immune training of Hermann Geesing (2008). The immune system is applied on eight women for the time of eight months. They were noting the activities of immune training into tables during two periods May to August and September to December. The results are evaluated by two graphs. One of them shows percentage of the set activities that were fulfilled. The second one describes subjective and complex condition of the women who participated in the training. The results are rated after every period and separately on every woman. At the end, the work describes the benefit of the immune training for every woman.
Application of Haematological Methods in Zoological Studies

Haematological methods are widely used in zoological researches. Unfortunately, especially in ecological studies there is often much imperfection in techniques adopted, and misinterpretation of results is also common. This is particularly the case of methods serving to assessment of the peripheral blood cellular composition. In this thesis I therefore decided to describe basic haematological methods, their mechanisms, advantages and disadvantages. I also highlighted possible application potential in several less widely utilised haematological techniques. In the practical part of this thesis I have shown that the examination of immature erythrocyte differential count may serve as a meaningful indicator of health state in birds.
