NADP-dehydrogenases of medicinal plants.

Medicinal plants usually contain metabolites formed by secondary biosynthetic pathways for example phenylpropanoid and shikimate pathway, which depend on NAPDH availability. In this work total content of phenolic compounds and flavonoids was determined in 10 medicinal plants common for Czech Republic. Activity of enzymes providing NADPH was determined as well. These enzymes were glucose-6-phosphate dehydrogenase (G6PDH, EC 1.1.1.49), NADP-isocitrate dehydrogenase (NADP-ICDH, EC 1.1.1.42), NADP-malic enzyme (oxalacetate decarboxylating) (NADP-ME, EC 1.1.1.40). The activity of shikimate dehydrogenase (SDH, EC 1.1.1.25), the main enzyme of shikimate pathway, was also determined. Extraction methods were optimized for each assessment. Applicable source for NADP-dependent enzymes are black radish (Raphanus sativus L. var. niger), garlic (Allium sativum L.) and mint (Mentha piperita L). The highest content of phenolic compounds and flavonoids was determined in sage (Salvia officinalis), lemon balm (Melissa officinalis L.) and Mentha citrata. Extraction with methanol was more effective than extraction with hot water. Possibilities of affecting secondary metabolites production in "in vitro" cultures of medicinal plants

1 ABSTRACT Possibilities of affecting secondary metabolites production in in vitro cultures of medicinal plants Elicitation is one of the methods used for increasing the production of secondary metabolites.The aim of this work was to affect the flavonoid production in callus and suspension cultures of St. John's Wort (Hypericum perforatum L.). Abiotic elicitor selendioxide in various concentrations (9.012.10-3 mol/l; 9.012.10-4 mol/l; 9.012.10-5 mol/l) was added in in vitro culture. The content of flavonoids was determinated by high performance liquid chromatography (HPLC). The samples were taken after 6, 12, 24, 48, 72, 168 hours after elicitor treatment. Selendioxide raised the production of hyperoside and quercitrin. The highest content of hyperoside (0.2 %) was detected after selendioxide (in concentration of 9.012.10-3 mol/l) treatment and 168 hours sampling in suspension culture. The highest production of quercitrin (0.37 %) was reached after selendioxide treatment (in concentration of 9.012.10-3 mol/l) and 6 hours sampling also in suspension culture. The thesis devotes onself to usage of medicinal plants at a specific part of a cadastral community. The thesis is focused on usage of stinging nettle (Urtica dioica L.) and black elder (Sambucus nigra L.). There was made a collection of these plants at experimental areas to specify the shrinking ratio. The averange value of the black elder was 1 : 5,54 and 1: 5,1 of the stinging nettle. Next was specified a productible supply of stinging nettles (dried tops) 954 kg and productible supplies of black elders (dried flowers) 57,55 kg.

The aim of the thesis is to elaborate the possibilities of utilization of medicinal plants in forest stands Šutra in the cadastral area of Rasošky village. In the thesis, the occurrence of medicinal plants was find out in the forest stands Šutra in the cadastral area of the village Rasošky. An inventory of species was identified with the determination of coverage, the possibilities of nettles and elderberry collecting, drying and utilization were studied on plots. The resulting average moisture loss ratio of Urtica dioica L. was 5,6 : 1 and 5,3 : 1 for Sambucus nigra L. and their stock on the surveyed area was 1 970 kg/ha for Urtica dioica L. and 950 kg/ha for Sambucus nigra L.

The final work is focused on medicinal and poisonous plants and gardens appearing in history from the beginnings to the Middle Ages. It deals with the design of a poisonous plant bed and organizes seminars, courses and workshops to raise awareness among the general public.

The thesis describes 5 species of their botanical description, content substances, use in cosmetics. food industry, ornamental horticulture. The task was also to organize an overview of companies in the Czech Republic and Slovakia dealing with the multiplication and sale of the examined assortment. The main topic of the work was to deal with vegetative propagation by its description, characteristics of the type of propagation and their results in individual species studied

This thesis focuses on medicinal plants cultivated or imported, collected and used in Middle Ages. It examines the processing of those plants in history as well as in today s traditional medicine, phytotherapy, and in pharmacology. There is mentioned more than 20 historically most important botanical species in 12 families in the literary part of this thesis. With every species we learn about its area of origin, brief botanical description, collected parts of the plant, which part of the plant is used as a drug and what effects it has on human organism. The basic charts contain an overview of 379 taxons described in Middle Ages. The charts containing the seeds and seedlings are based on the range of goods of four important companies which offer these days the same range of medicinal plants that were available in Middle Ages. These charts contain 95 species of medicinal plants.

Great attention is paid to the basic characterictics of those kinds, which can be planted in Czech Republic. For selected kinds history, origin and expantion, botanical description, medicinal usage and the conditions of planting in Czech Republic were processed. The diploma thesis concentrates on medicinal plants, their use and effects in the treatment of diseases. The thesis summarizes some effects of medicinal substances contained in plants and herbs. They are suitable for use in selected diseases. The research part of diploma thesis deals with the mapping of methods of treatment and production of herbal products among university students. The thesis also examines whether students are inclined to use more herbal therapy over conventional pharmaceutical drugs. It investigated whether information on herbs in schools sufficient. The aim of the thesis is to evaluate the prevalence of dietary suplement use by patients before surgery. Another aim is to point to potential danger which could endanger the patient while irrational using of the dietary supplement before the medical interventions (interactions of dietary supplement, side effects of dietary supplement). Methodology: The questionnaire research was conducted in order to obtain the data. It was counducted between 9th May 2017 till 30th June 2018 at 9 departments of the University Hospital in Hradec Králové. In total 332 patients in the time before surgery were asked and 257 completed questionnaires were analysed. Results: From the results of the study we can see the prevalence of the dietary supplement use by the patients in the time before surgery, which amounts to 42 %.
