Influence of aminoacid side-chain ionization on protein structure

Protein folding is governed by many different types of interactions. Aside from the formation of hydrophobic cores and hydrogen bonding, Coulombic forces are believed to play an important role in this process. Their importance arises mainly in partially unfolded states and pH sensitive regions where ionization of amino acid side chains may trigger conformational changes. Better understanding of the role of ionizable residues in folding thereby provides us with new opportunities in protein design and property-enhancing protein modifications.
Mass Spectrometry

Hmotnostní spektrometrie je metoda analytické chemie používaná k analýze organických i anorganických sloučenin. O tomto tématu se běžně dozvídáme z prací týkajících se chemie, protože tato metoda může být využita k identifikaci mnoha druhů sloučenin. Vlastní fungování přístrojů používaných v hmotnostní spektrometrii ale musíme hledat ve specializovaných knihách zaměřených na tuto metodu a tyto informace jsou obvykle popsané velmi komplikovaně. Proto jsem se rozhodla napsat práci, která uvádí základní informace o hmotnostní spektrometrii z pohledu elektrotechniky, srozumitelně popisuje jednotlivé fáze této metody a která nebude zaměřena na výstup analýzy, ale na technický proces. V druhé části práce se věnuji praktickému použití hmotnostních spektrometrů v průmyslu při testování těsnosti produktů a typům testování těsnosti, vysvětluji důležité termíny a detailně popisuji spektrometr LDS3000. Mým záměrem je ukázat, že tato metoda je skutečně zajímavá a poutavá a chci ji proto představit ostatním.
Design and development of the electrostatic precipitator

This bachelor thesis deals with the design of an electrostatic precipitator and familiarization with the problems of separation on these separators. The aim of this work is to construct a functional model of an electrostatic precipitator. The principle of operation and design will be described. In addition, the design of a separator is elaborated, which was subsequently realized and experimentally verified in the CVVOZE Power Laboratories Research Infrastructure. The first part deals with the description of the legislation on air protection, the most frequent input material of separators and descriptions of the basic types of separators. In addition, the electric separator, its area of use, construction and individual components, as well as the undesirable operating state of the separator, are further elaborated. In the second part are presented the equations and calculations for the design of the separator. In the next part, the separator designer and his laboratory tests are described. The separator was subjected to basic voltage tests from the viewpoint of its operating voltage. Its functionality fits the realized experiments up to 30 kV. On the basis of this work and the production of the equipment, it is possible in the future to test the properties of flue gases and other gases.
Acid-base equilibria and tautomerism of nucleobases

Nucleic acids and their components belong to the most important molecules essential for life. Hydrogen atom migration across the skeleton of purine or pyrimidine bases of nucleic acids gives rise to their various tautomeric forms. Furthermore, depending on pH of the medium, these bases might become protonated or deprotonated. Protonation/deprotonation of nucleic acid bases is involved in many biochemical processes. These include mainly regulation of replication and gene expression by means of specific secondary structure stabilization or acid- base catalysis of phosphodiester backbone cleavage. Additionally, secondary structures that include ionized bases are also associated with various genetic diseases. However, protonation and deprotonation are very fast dynamic processes and their experimental detection is difficult. Therefore, this work focuses on development of reliable method for relatively fast detection of pyrimidine base derivates primary protonation site. At first, pyrimidine bases with protonation site determined by structure were analyzed in both neutral and ionized state using NMR spectroscopy. Design of Portable Air Purifier

The topic of my Bachelor's project is focused on the design of a air purifier. Specifically product with a content of up to 500 m3 / h and a power consumption of 65 W. I must use materials from the FSI Science fund 2016. The aim of the design is to design a functional model with respect to technical, graphic, ergonomic and especially material requirements.
Membraneless speaker

The bachelor thesis consists of theoretic basics and describes the design and construction of one-channel membraneless speaker. Absence of mechanical components gives the membraneless speaker no frequency limitation. The design consists of power source, control electronics, which modulates the signal, convertor and output transformer. The power source is realized by transformer, which lowers the voltage and stabilizators. Modulation is provided by integrated circuit TL 594, its duty is adjusted by potentiometer P1 and frequency by potentiometer P2. Power conversion provides dual forward converter. As a output transformer was used transformer from color television.
Application of plasma arc in technical practice

ZAJÍC, Jiří: Application of plasma arc in technical practice The thesis is focused on the description of technologies using plasma, for its high temperatures, dynamic effects and ionization. The first part of the thesis is focused on the description of the process of ionization of gases and individual factors influencing the parameters of the resulting plasma arc. The following part is a description of the developed technologies that use gas ionization for their function, such as welding, cutting, thermal spraying and heat treatment. In a final phase of thesis is disposed practical experiment containing comparing welds made by plasma technology and welds made by TIG technology. These welds are compared in terms of hardness, heat input and the plasma welds in dilution of weld metal.
Architektonické a technické prostředky tvorby elektroiontového mikroklimatu budov

Mikroklima budovy je nezbytnou podmínkou k tomu, aby stavba splnila účel, pro který byla navržena. Škála požadavků na vnitřní mikroklima je tak široká, jak široká je škála způsobů užívání. A stejně široké jsou i možnosti architektů a inženýrů při hledání cest k zajištění co nejlepšího prostředí v budovách. Cílem článku je vymezení oblasti problematiky mikroklimatu budov, která bude řešena v disertační práci na téma Reálnost tvorby mikroklimatu budov architektonickými a technickými prostředky.
Air ion measurement

This thesis focuses on the basic problems of ions and the ionization itself, what influence ions have to the organism. Further it examines compensation of small negative ions in the residential environment. Introduction to measurement of ion concentrations. First measurements with the aspiration capacitor which is now the most commonly used measurement of ion fields. Measurement is focused on the concentration of air ions in the building, VUT FEKT T-10, then the concentration changes depending on the daily cycle, ventilation and concentration before teaching and after teaching.
