Video analysis for entomology

Analysis of behavioral patterns of Drosophila Melanogaster species is a sub- ject of study of many laboratories. The behavior of individuals, for example in different gene lines, is monitored. In order for this analysis to be possible, it is necessary to have a device that is capable of tracking the fly and providing machine-readable data about them. These data are the output of a trace pro- gram FlyCatcher, which records the positions of the observed objects. Thanks to this, we can determine the location, the mutual position of individuals and other parameters. FlyCatcher is based on the use of the AForge library which handles multimedia files. FlyCatcher lets us process videos made in the lab and provide outputs in the .csv format. This format is further processed by external statistical programs.
Neo-sex chromosomes and their evolutionary significance

Neo-sex chromosomes arise due to rearrangements between ancestral gonosomes and autosomes. Neo-sex chromosomes are valuable systems for the study of sex chromosome evolution. It is possible to use them for analysis of processes driving the formation of differentiated sex chromosomes, especially suppression of recombinations and alosome degeneration. The most important rearrangements forming neo-sex chromosomes are Robertsonian and reciprocal translocations. Speciation events are important consequences of birth of neo-sex chromosomes. The presented study is focused on the evolution of neo-sex chromosomes in drosophilids, muntjacs, and monotremes. Neo-sex chromosomes emerged in many Drosophila species. The genome of D. albomicans carries the youngest known neo-sex chromosome system, which arised only one hundred thousand years ago. Information from research into Drosophila neo-sex chromosomes underlines the importance of Muller's ratchet and background selection at the beginning of sex chromosome differentiation and hitchhiking and deleterious mutations at the end of this process. Genomes of muntjacs carry young and fast evolving neo-sex chromosomes. In several muntjac species, neo-sex chromosomes form a considerable part of the genome. Role of extracellular adenosine in Drosophila

This thesis describes several aspects of the role for extracellular adenosine in Drosophila. Reverse genetic, molecular and microscopic methods together with the most forefront Drosophila research techniques have been applied to elucidate the role of adenosine signaling in the regulation of development, physiology and metabolism of Drosophila larvae. The thesis helps to establish the model for extracellular adenosine as a stress-signal for the release of energy stores. It also describes the elucidation of Drosophila extracellular adenosine production pathway by functional characterization of extracellular ATP/adenosine converting enzymes. Further, this thesis describes the attempt to study adenosine signaling by creation of tissue specific knock-down mutation of Drosophila adenosine receptor. Finally, it provides results of the utilization of in vivo fluorescent reporters for studying of the localization, function and interaction of enzymes implicated in adenosine signaling pathway.
Study of Dco role in \kur{Drosophila melanogaster} hematopoiesis

The main goal of this work was to test, whether an RNA interference (RNAi) construct can be used to knock down the gene Disc overgrown (Dco) in Drosophila melanogaster and what effects this knock down of Dco has on the hematopoiesis, the phenotype and the development of D. melanogaster.
Genetic screening for the dominant suppressors of adgf-a mutant phenotype in Drosophila

Extracellular adenosine is an important signaling molecule in various physiological processes. When not regulated properly it can cause various pathologies such as Severe Combined Immunodeficiency. To study the effects of increased extracellular adenosine level a Drosophila model has been established. This model is based on mutation in the main adenosine deaminase gene in Drosophila (ADGF-A, Adenosine Deaminase-related Growth Factor A). This work is a consequence of large screening for the suppressors of adgf-a mutant phenotype and presents a more detailed study of 3 previously identified regions containing dominant suppressors.
Circadian clock of two insect model species - \kur{Drosophila melanogaster and Tribolium castaneum}

The aim of this study was to determine the specific interactions among clock gene alleles in Drosophila melanogaster and their effect on the function of the circadian clock. The second part of this study deals with the expression pattern (both temporal and spatial) of two core clock factors known from Drosophila, period and timeless, in the central nervous system of the red flour beetle, Tribolium castaneum.
Cold hardiness of larvae of the fruit fly, \kur{Drosophila melanogaster} (Diptera: Drosophilidae)

We assessed survival of larvae of the fruit fly, Drosophila melanogaster at low temperatures (0°C and 5°C) after rearing them under seven different acclimation regimes. Larvae that have developed in a standard diet at 25°C showed 50% mortality after 12,6 min of the exposure to 0°C (Lt50 = 0.21 h). In contrast, larvae that have developed in a diet enriched with glycerol at 15°C, and were cold acclimated at 5°C during last two days of their development, had Lt50 = 38.6 h. It means that it was possible to increase the Lt50 at 0°C more than 180-fold using simple manipulations with rearing temperatures and diet composition. The physiological differences in duration of larval development, fresh mass, dry mass, hydration and total contents of proteins, lipids and glycogen between the larvae belonging to different acclimation variants are described. The samples for future detailed metabolomic analysis were prepared.
