Reasons of discarding blood donors at transfusion department of Hospital České Budějovice a.s.

The topic of thesis are reasons for discarding blood donors. The aim of this thesis is to evaluace the amount of discarded blood donors and the reasons for elimination in 2015 and 2016, further implementation of pre-sampling and post-sampling tests in the Blood Bank Department of the Hospital ČB, Ltd. In the theoretical part I attend to important events in the history of transfusion medicine. Next I deal with criteria for receiving blood donors, evaluation of the blood donors and ways, amounts and frequency of blood collections. The important part of my thesis are the reasons for discarding blood donors, which are divided into temporary and permanent. There are registers, in which these donors are registered. In the methodological part I attend to pre-sampling and post-sampling tests, which are performed in the Blood Bank Department of the Hospital ČB, Ltd. At pre-sampling laboratory measurement of hemoglobin amount on HemoCue 201+ is performed. For new blood donors the blood group orientation is examined on the slide. Donors plasma, platelets or in case of chylozity blood at the last sample hematokrit is examined. Further examination post-sampling test, which are include tests of infectious markers and imunohematology. For examination infectious markers are of service analyzer Architect, tests make on this analyzer are: Ab anti HCV, Ag and Ab HIV, Ab against Treponema pallidum and HBsAg. Immunohematology tests are examination with a dispenser Qasar and imagine analyzer Duet Reader. These two devices are jointly involved in the testing of AB0 blood group and RhD, screening of irregular anti-erythrocyte antibodies and examination of a group of Rh Kell. In 2015 there were 15 048 blood donors and 360 women and 306 men were discarded. In 2016 there were 15 788 blood donors and 286 women and 211 men were discarded. The most blood donors were discarded because of unsufficient amount of hemoglobin.

Petr Fabián, one of the most significant architects dealing with revitalizations and reconstructions of historical buildings in the Czech Republic, mainly in the Olomouc region, notably in the towns of Olomouc, Šumperk and Zábřeh. Judgement of most of the population on the architecture of buildings is mostly based on the fact whether they like it or no. For those who understand this branch more, the works of Petr Fabián provide the unique example of high architectural quality. His works reflect his persistence, refined style and theoretical knowledge gained during his studies at the Faculty of Architecture of VUT in Brno, under the supervision of prof. Helena Zemánková. Architect Jiří Voženílek in Zlín

Jiří Voženílek was born on August 14th, 1909 in Holešov. He attended high school in Prague, where he also studied architecture at the ČVUT and he became a university professor at this institution in the 1960’s. Later, he became the main architect of The Capital of Prague. He started to work for the Baťa Company on April 20th, 1937. His first task was to assist the architect Vladimír Karfík on the Roman Catholic Church project in Otrokovice-Baťov. Then he became the leader of the collective 276, Construction Department no. 8 and the head architect as well. He cooperated with architect Jaroslav Fragner for the Baťa Company on the creation of the urban concept called The Industrial City of Kolín in 1940-41. He created an interesting industrial project, The Rendering Plant in Otrokovice–Baťov on his own. Several of his other architectural works can be found in the former Baťa factory in Zlín, particularly the factory buildings no. 15 (built in 1946-47) and no. 14 (built in 1947-48). However, the most famous work of Jiří Voženílek is the Collective House in Zlín. He is the author of many famous urban projects in his homeland and around the world as well. He became the head of the group of architects who created the Regulation Plan of Zlín in 1946-47, which is valid even today.
The Creative Centre Brno-Cejl

The main purpose of this project is reconstruction former penitentiary for a Creative Centre. The lokation is in Brno, between Cejl streer, Bratislavská street and Soudní street. The main idea of this project is „make the place with incubators for young architects, creativ persons and designers. The building program is adapted to the historical essence and construction and technical situacion of this building. There are designed spaces for their creations, exhibions, distributions and auctions of their artistic products. The idea of concept contains too the space for lectures, workshops, seminars, courses and untraditional educational projects, visual and acustic and different productions. There is too the possibility of short-term accommondation in attic. The part of this object is reserved for dancing lessons. All these spaces are designed for a rent. There are designed too new entrance spaces, parking places, technical equipment and facilities. The inseparable part of this project are inner atriums, entrance spaces and contunuity on public street spaces and communications. The complex of Creative centre is without mobile roofing and without covered passages. The main idea of this complex is to preserve the main square mass with the central wing, and is to clean up the less historical insignia extensions of the Cejl street and of the Bratislavská street. From the constructional poit of view consists this buiding two aboveground floors with used attic. The attic is coved with gabled roof made of interlocking tiles. The complex is based on stone basis with bearing wall made from full of burnt brick. The ceilings are made of wooden beams with arches of full of burnt brick. At the present the complex is without permanent using. The part of this complex was sold in personal property. Investigation for infectious markers of blood donors in Blood Establishment Hospital Č.B. a.s.

The theme of the bachelor thesis is ?Infectious Disease Marker Testing of Blood Donors?. According legal rules, donated blood is tested for infectious agents of AIDS, syphilis and HBV, HCV. The risk of transmission of such diseases has been decreased in recent decades nevertheless it is still a threat to transfusion medicine. At the Transfusion Department of the České Budějovice Hospital, Inc., the samples of blood donors are tested in the Blood Donors Analytics Laboratory. At present, all the tests are performed on a single instrument Abbott Architect. Before testing on the Architect analyzer, viral markers were tested on the Abbott AxSYM instrument, while syphilis was tested via TP-PA method using a diagnostic kit by Serodia. The objective of the bachelor thesis is to compare the current testing systems with the previous ones, describe individual testing methods, count the numbers of reactive results and compare results of its confirmation tests, find out the frequency of reactive and positive results in new blood donors, verify results of the second confirmation of control samples. The thesis informs about basic criteria to select blood donors, individual infectious markers which cover the infectious diseases, in which a prolonged presence of the agents in blood occurs, and thus the transmission of the infection is possible. Further, the principles of methods used by the AxSYM analyzer, the Architect analyzer and Serodia TP-PA diagnostic kit are described. The samples showing the reactive results are sent to the National Reference Laboratory to be retested. In the period from 1st January 2011 to 31st December 2011 17,262 blood donors were tested using the AxSYM analyzer to detect HIV, HBsAg and HCV infections. The same number of the tests was performed also by TP-PA method to diagnose syphilis. In the period from 1st February 2012 to 31st January 2013, 18,346 of blood donor samples were tested to detect HIV, HBV, HCV and syphilis. 57 samples from the total number of tests performed on the AxSYM analyzer and TP-PA diagnostic kit were reactive in the year 2011. With the installation of the Architect analyzer the numbers of reactive samples increased more than twice, namely to 121 reactive samples. The largest increase in reactivity is evident in case of syphilis diagnostics. It was the similar with anti-HCV method. In both cases, the methods for antibodies detection are involved, and so it can be taken into account the interference with other non-specific antibodies. In the period of testing by TP-PA method on the AxSYM system, 20 reactive control samples, 10 reactive first time blood donors samples and 27 reactive regular donors samples were detected. After the changeover to Architect system, the numbers of reactive samples were increased unexpectedly in regular donors the most, namely up to 88 samples within the monitored period unlike 8 reactive samples in first time donors or 25 reactive control samples. In most cases, the confirmation test results of control samples are negative regardless the type of analyzer used for screening tests showing reactive results. The largest number of unclear results from the National Reference Laboratory (NRL) was observed with anti-HCV method. The NRL confirmed altogether 5 positive results for the monitored infection diseases, from which 4 were in new donors. This finding confirms the assumption that the first time donors are a risk group while with the regular blood donation the safety of blood transfusion is increasing. Based on all observed parameters the both analyzers can be regard as the safe systems for blood donors testing showing the sufficient specificity. The AxSYM analyzer was replaced by the newer Architect analyzer because of upgrading the machine equipment primarily. The chemiluminiscent test technology is preferred because of its high sensitivity, simplicity and reliability.
New fountains on the Republic Square in Pilsen

I have chosen three public fountains called the Angel, the Camel and the Grey-hound bitch like a topic for my Bachelor work. These fountain were emplaced on the Republic Square in Pilsen in July 2010. The author of this work is Mga Ondřej Císler, whose fountains were whose fountains were honour for honorable mention of honorable mention in prestigious competition of architects called Grand Prix. It was in Architectonic design cathegory. Throught writing of my thesis I tried to bent on these object like author made himself. His work helps the koncept of monumental medieval square to grow in nowadays work which exemplifies scale and material permeation of the space.1 My main source-books were Cisler´s personal materials for his project. Second one was literature deals with sign and literature charting History of Pilsen. The topics of the fountains are inpirated by heraldic sign of the town. Other sourses come from publications bent on modern architecture. My bachelor thesis contains mention of previous fountains staying on the square before.
A picture of building activity of a noble family in the age of historicism.
KUČERA, Petr
This graduation theses deals with the conditions of reconstruction of the Krivoklat castle by the noble family of the Fürstenbergs. Their reconstruction activities are viewed according to the temporal way of thinking of the historical noble families and common attitude to architecture and rising movement of care of historical monuments in the 19th century. The castle plays the role of the Fürstenbergs possession and the other society changed according to the raising of nationalistic moods and the great personalities of the community. Another important part of this work is a survey of the architects who changed the appearance of the castle, particularly J. B. Urban who was the chief of Fürstenberg´s building section. His memories serves as a worth source of information.
