Nanoscopy, spectroscopy and modication of individual nanoobjects in liquid environment

In this diploma thesis we will study the luminescence properties of nanocrystals. We will summarize the basic division according to size and standard method of theoretical description of semiconductor and metal nanocrystals. We will describe the luminescence properties of nanocrystals and the influence of the surrounding environment. In the se- cond part of the thesis we will follow up the construction of an apparatus for imaging luminescence spectroscopy intended for the measurement of individual nanoobjects in a liquid environment. Finally, we will study luminescence properties of organixally passi- vated metal clusters in a liquid environment with changes in temperature and excitation intensity. We compare the luminescence of gold nanocrystals with and without long po- lyethylene glycol chains bound on the surface. 1
Morphological and Genomic Profiling of Circulating Tumor Cells in Metastatic Colorectal Cancer

Colorectal cancer (CRC) is the third most common cancer worldwide; it is responsible for nearly 10% of all newly diagnosed cancers and is the second most cause of cancer related death in Europe. Biomarkers for therapy guidance, targeted therapy and survival prognosis are still limited. As CRC is a heterogeneous disease, different parts of the tumor might have varying molecular characteristics which may change during therapy or disease progression. Through solid biopsies and screenings, these local or temporal differences are impossible to monitor. To facilitate detection of these possible temporal changes, a regularly and non-invasively accessible biomarker is required for disease monitoring. Circulating tumor cells (CTCs) might represent such a biomarker as they have been shown to be fluid surrogates of the solid tumor. EpCAM positive CTCs have shown to be prognostic in CRC for survival, but their full potential has not yet been evaluated further. By using the High Definition Single Cell Analysis (HD-SCA) workflow, we were able to analyze the entire spectrum of CTCs and categorize them as the regular CTCs (HD-CTC), CTCs with a smaller nuclear area (CTC-Small), CTCs with low expression of epithelial marker cytokeratin (CTC-LowCK) and CTCs undergoing apoptosis and therefore releasing cell free DNA...
Twenty Eighty-Four

The project follows up on the undergraduate work on the issue of control and surveillance in public space. Tendencies of absolute transparency, well-arranged and controllable public premises, supposedly implying security, are present in most of the newly emerging complexes of buildings and urban planning strategies. Whether it is done consciously or as a result of current “trends” and requirements, which only a few question, the space is built to meet these demands for easy controllability. The second method of control, which has also emerged during the era of modernism, is the control of the indoor climate through "mechanical environmental management" or through the use of fossil fuel energy for heating, cooling and ventilation. Current requirements for the comfort of the indoor environment presuppose a constantly, fully and homogeneously tempered environment, regardless of the outdoor climate or the time of year. Both control regimes permeate all measures of the urban environment and have a direct impact on its organization, form and, above all, on the regimes of its usage. Developers and their commissioned architects repeat well-proven business models and forms, without any major focus on the context of the place, and incorporate this "logic of control" into their buildings and surroundings. Housing complexes are planned and built up as one unit with a rigid structure and only a minimal possibility of their later alteration. The domain of control through transparency penetrates from the public space to the entrance door of the apartment unit. Indoor, social control changes to total control of the interior climate of the apartment, where the boundaries between seasons and day and night are blurred. The proposal is investigating alternative ways of urban planning and architecture. The project deals with the relationship between building, its surroundings and applicable regulations, standards and energy efficiency, along with the idea of adaptive living.
Setup of Application-Computation On-Premise Mini-Cloud Based on Kubernetes

 Kubernetes je platforma na orchestráciu kontajnerov, na nasadenie a správu aplikácií v klastri. Cieľom tejto práce je porozumieť kubernetes a jeho komponentom a následne navrhnúť a sprevádzkovať optimálnu architektúru kubernetes klastru pre malý mini-cloud založený na kubernetes v areáli univerzity VUT. Táto bakalárska práca rozoberá základy kontajnerov, runtime kontajnerov, nástroje na orchestráciu kontajnerov, architektúru Kubernetes a jej komponenty a automatizačnú platformu Ansible. Ďalej obsahuje popis navrhnutej architektúry klastra, ktorá bude implementovaná. Príspevok tejto práce spočíva v návrhu architektúry kubernetes klastra, ktorý bude neskôr nasadený na pôde univerzity, pripravený na použitie.

Promotion of research and development by means of cluster brand building

The diploma thesis deals with promotion of research clusters by means of brand building. It focuses on a current situation of research clusters, research and development promotion and cluster branding. Primary data were collected by 2 qualitative research methods, focus group and in-depth interviews, which were carried out with representatives (n = 15) of 2 chosen target groups. The results are establishing research Stroke Brno cluster together with its branding strategy, cluster mapping and content analysis report summarizing the qualitative research results.

This thesis evaluates individual forms of cluster financing divided to public sources and private sources. Theoretical bases are applied to Network Security Monitoring Cluster, which represents an example of cluster. The first part of thesis is using descriptive method and it is formulated as literary research. The second part includes an analysis of Network Security Monitoring Cluster. The conclusion compares theoretical assumption of cluster financing and advice for clusters.

The thesis fetches the overview of achievable support for Czech small and medium enterprises, what can be used as a guide. The profit of this thesis the author brings in the analysis of the Philippines for potential Czech entrepreneurs. Cluster policy is one of the tool for the support of enterprises, what is in European context currently very actual. The analysis of Philippines is applied in a proposal of internationalization study for Czech cluster CREA Hydro&Energy to the Philippines. The internationalization study contains especially identification of an opportunities and a SWOT analysis. The added value of thesis is own experiences of the author achieved during internship in the Philippines. During the internship the author was collaborating with CREA Hydro&Energy on the European cluster association NATUREEF on project Go International.Formula clause:The thesis contains commercially exploitable information obtained and analyzed in collaboration with the cluster manager and financial support cluster CREA Hydro & Energy. Restrictions publications binds to chapters 5) Evaluation of the Philippines as a country suitable for Czech SMEs and 6) studies Internationalisation CREA Hydro & Energy in the Philippines. Secrecy takes two years after the thesis defense (2017-2019).
Předpoklady konkurenceschopnosti zemědělského podniku v komoditní vertikále víno

The aim of presented PhD. thesis is to specify the most important rules and conditions which are affecting a company operating in the first phase of the wine commodity chain and strengthen its competitiveness. First of all, the theoretical and methodological aspects of the issue have been formulated, tracking the whole process of the commodity flow. The assumptions are verified from theoretical view as well as statements answering the research questions and the dedicated hypotesis. The successful position is being created by utilization of the integration and contract processes particularly through an active membership in the wine industry clusters. Its briefly described changes of agriculture sector in contemporary agribusiness. This thesis is emphasizing role of globalization and its processes with a direct impact on the competitiveness of agriculture companies. Further on, the key factors of agribusiness, shaping of markets and comodity chains are being listed, those have influence the conditions of the above within last decades. Also, its describing key aspects of the comodity chain's competitiveness supported by contractation cooperation enhancing the whole wine sector. Consecutive part is dedicated to the history of wine industry in the Czech republic, transformation process of free competition after EU access with the related impacts and limits. There is also a chapter emphasizing the key role of the consumer playing in the whole commodity chain. Furthermore, the strongest forces of the industry which occured within last decades collateral to the long term factors are being specified. The analysis of the development of vineyards (wine growing) and wine sectors from the world, European and national view is presented there. To support the theoretical and methodological outlets, the conclusions were compared and applied to the selected middle sized companies operating in the first and second phase of the commodity (vertical) chain in the wine sector.
Effect of sub-micrometer structural features on rheology of polymer nanocomposites

Polymerní nanokompozity (PNCs) mají slibnou budoucnost jako lehké funkční materiály zpracovatelné aditivními výrobními technologiemi. Jejich rychlému rozšíření však brání silná závislost jejich užitných vlastností na prostorovém uspořádání nanočástic (NP). Schopnost řídit disperzi nanočástic je tak klíčovým předpokladem pro jejich uplatnění ve funkčních kompozitech. Tato práce zkoumá přípravu polymerních nanokompozitů v modelové sklotvorné polymerní matrici roztokovou metodou, technikou schopnou vytvářet prostorové uspořádání nanočástic řízené strukturními a kinetickými parametry přípravného procesu. Prezentované výsledky popisují rozdíly mezi změnami rheologického chování roztoku polystyrenu při oscilačním smyku s vysokou amplitudou (LAOS) vyvolanými nanočásticemi. Výsledky vedou k závěru, že vysoce-afinní OP-POSS nanočástice při nízkých koncentracích dobře interagují s PS a tvoří tuhé agregáty, zatímco nízko-afinní OM-POSS nanočástice za těchto podmínek neovlivňují deformační chování polymerních řetězců. Dále byla pozornost zaměřena na vliv použitého rozpouštědla na uspořádání nanočástic v SiO2/PMMA a SiO2/PS nanokompozitech, který je v literatuře prezentován jako parametr řídící prostorové uspořádání nanočástic v pevném stavu. Důraz byl kladen na kvalitativní rozdíly mezi „špatně dispergovanými“ shluky nanočástic, které byly na základě rheologie a strukturální analýzy (TEM, USAXS) identifikovány jako polymerními řetězci vázané nanočásticové klastry a dva typy agregátů, jeden termodynamického a druhý kinetického původu. Jednotlivé druhy agregátů se vyznačují odlišnými kinetikami vzniku a rozdílnými vlastnostmi jak mezi sebou, tak v porovnání s dispergovanými nanočásticemi. Pozorované typy disperze nanočástic byly kvantitativně posouzeny podle svých rheologických vlastností během roztokové přípravy, podle kterých byla vyhodnocena míra adsorpce polymeru na povrch nanočástic a atrakce ve vypuzeném objemu. Výsledky byly porovnány s teorií PRISM. Důležitost uspořádání nanočástic byla demonstrována na porovnání teplot skelných přechodů různých struktur při stejném chemickém složení.
