Variability and silencing of transgene expression in potato plants and in tobacco cell line BY-2

Conclusions In suspension cultures of tobacco BY-2 cell line derived from calli after transformation about 90 % of lines contained cells with various GFP fluorescence level after transformation. Newly introduced cloning method allowed obtaining nearly 50 % of clones with homogeneous GFP expression from primarily heterogeneous BY-2 lines. Heterogeneity of GFP expression in transgenic BY-2 lines had two causes - genetic (primary lines contained cells with different T-DNA insertions) and epigenetic one. Epigenetic heterogeneity of BY-2 lines was connected with transgene silencing, formation of stable epigenetic states early after transformation, and "permanent heterogeneity" with fluctuating levels in GFP expression. Reduction or silencing of transgene expression in potato was predominantly observed in lines with higher number of T-DNA insertions and with higher initial GFP expression. Silencing in potato always gradually affected both introduced genes. Silencing of GFP expression preceded (in months) loss of resistance to kanamycin (silencing of NPTII gene) in all monitored cases that indicates interconnections between silencing of both transgenes. Strategic Management in the Non-Profit Sector - Suggested Strategy for the Enterprise Leaders Platform

This work presents a case-study of application of the standard strategic framework to a specific non-profit organization in the Czech Republic. This organization is struggling with financial problems and with lack of competitive advantage. In the first part of the work, results of the external and internal audits are used to formulate a medium-term strategy for the organization. The process identified following key areas: strategic financial management, employee training and education and continuous improvement of quality of services. These areas are further developed into strategic goals and initiatives using a strategic management system Balanced Scorecard (BSC). Each perspective of BSC contains an example of the cause-effect relationship of selected initiatives. The output of the model is a matrix of scorecard measures which forms a basis for strategic management in the organization. Finally, early steps for the implementation of the Balanced Scorecard for a period of six months are identified. GBM is resistant to current treatment and the median survival of treated patients with this disease is around 15 months. The very poor prognosis of this disease has prompted the research of new therapeutic strategies. A promising direction is represented by drugs already registered for other indications, such as the anthelmintic flubendazole (FLU) with potential antitumour activity. The aim of this work was to study the effect of FLU after repeated low-dose treatment of GBM cells. The Influence of Neurotraining on Post-Concussion Syndrome in a Football Player - A Case Study

Title: Effect of neurotraining on post-concussion syndrome in a football player - a case report Aims: The aim of this study is to analyse the effectiveness of neurotraining for alleviating the symptoms of post-concussion syndrome using a case study of an adult football player after concussion. Methods: The case-control study used the test, re-test technique. The standardized SCAT5 questionnaire, neuroimaging system testing using optotype, prismatic flipper 6/12, Neurotracker, RightEye and Senaptec Sensory Station were used to assess changes in the effectiveness of the method. Stability measurements were performed using the Physics Toolbox Sensor Suite and, for cognitive function, the Stroop effect. Next, a neurodiagnostic was performed, based on which the proband was selected to perform neurotraining exercises for two months (visual system and mechanoreception were tested). Results: The proband improved in visual acuity by 20 %, vergence flexibility by 44 %, neurocognitive function by 44 %, reaction time by 29 %, depth of field binocularly by 12 %, visualization by 6 % and improved on the neurotracker by 0.3. Changes in nutritional status in patients with cystic fibrosis in the era of CFTR modulators

Cystic fibrosis is an inherited disease that primarily affects the respiratory system but also the gastrointestinal tract. Nutritional status is a significant factor in overall health and prevention of malnutrition is a key component of therapy. New modulator therapy contributes to major improvement in health status as well as quality and length of life. The aim of the bachelor thesis was to evaluate the development of nutritional status after initiating modulator therapy. The study assessed whether there was an increase in BMI, whether the treatment response was affected by the initial nutritional status, and whether the percentage of patients with target and better nutritional status increased. The research sample consisted of 86 patients from the Cystic Fibrosis Centre at the Motol University Hospital. The development of BMI was evaluated based on retrospective collection of anthropometric data. Data from 4 measurements were used: before treatment and then at 1, 3 and 6 months after starting treatment. Friedman test and comparison of median values were conducted to evaluate BMI development. Veganism and vegetarianism are among the dietary trends with increasing popularity. As one of the diets that excludes a certain food group, the risk of deficiencies of certain nutrients in the diet needs to be addressed. For children, these deficiencies may be even more risky. This thesis focuses on selected potentially risky nutrients whose levels have been investigated in the paediatric population. Aims: The aim of this thesis was to assess whether there are differences in blood levels of zinc, calcium and vitamin D between vegetarians, vegans and omnivores. A secondary aim was then also to compile recommendations for parents in the form of an educational leaflet with information on dietary information for vegetarianism and veganism with regard to zinc, calcium and vitamin D. Methodology: The data used in this thesis were collected as part of the first phase of the KOMPAS study. Whole families eating a homogeneous diet, either vegan, vegetarian or omnivorous participated in the study. An Analysis of Pathogenetic Processes Evolving in Comatose Survivors After an Out-of-Hospital Cardiac Arrest

In comatose out-of-hospital cardiac arrest victims, there is insufficient evidence for the timing of coronary angiography (CAG) in the absence of ST elevations on the baseline electrocardiogram, for the optimal body temperature during the temperature control pro- cedure, or for S100 protein levels and regional brain saturation values measured by near- infrared spectroscopy regarding neurological prognostication. The aim of this study was to compare the six-month survival and neurological outcome, evaluated by the Cerebral Performance Category (CPC) scale, in patients with- out ST-segment elevations indicated to immediate CAG and those initially treated conservatively, six-month CPC and acute complications according to the temperature target range of 34 - 36 řC versus 32 - 34 řC during therapeutic hypothermia, and S100 protein levels and regional brain saturation values with regard to the six-month CPC score. We analysed a prospective registry of 283 unconscious patients after an out-of- hospital cardiac arrest undergoing therapeutic hypothermia. Title: An analysis of training load in standard and Latin American ballroom dancing in 10-15 years old children Aims: The aim of this bachelor thesis is to study the intensity of physical activity of dancers during the performance of standard and Latin American dances. The analysis of the intensity of exercise will allow a deeper understanding of the effect of these dances on the human body and provide recommendations for an effective training process for dancers. Young dancers are often unaware that dance can be classified as an anaerobic sport with physiological demands similar to traditional anaerobic sports such as sprinting, swimming and weightlifting. The aim of this study is also to uncover the problems associated with the intensity of physical activity during the training of ballroom dances and to investigate the effect of these dances on the physical preparation of dancers. The research question focuses on the intensity of the dancers' physical exertion and the factors affecting this exertion. The results of the study could assist dance physiologists, physical therapists, and pediatricians in preventing growth-related injuries in young dancers.
