Development of active supplement with probiotics

Subject of this bachelor thesis was to investigate the antimicrobial activity of selected probiotic strains and determine how the probiotic growth is affected probiotic growth by prebiotics and essential oils. Probiotics are bacteria, which positively affect human health. They are mainly used in food industry as fermentation starter cultures, where they produce antimicrobial compounds that support resistance to pathogenetic bacteria during the fermentation process. In general, fermentation process is used to prolong the shelf-life of foodstuff. However, the applications of probiotics are very limited in food or other industry because probiotics are living organisms, so at least the industry tries to use their metabolic products. The method of encapsulation could support the usage of probiotics in other unusual ways. In experimental part of this thesis, the growth of probiotics was studied in both commercial MRS broth and in minimal broth, which could help with isolation of bioactive metabolic products such as lactic acid and bacteriocins. Additionally, the effect of prebiotic-inulin and essential oils from oregano and thyme on probiotics growth was investigated. In the second part of thesis, the content of lactic acid was analysed and antimicrobial activity of selected probiotic strains and effect of additives on activity was tested. Antimicrobial test discovered that prebiotics and essential oil from thyme enhanced the antimicrobial activity of tested samples. Also, the minimal broth was shown to be unsustainable for cultivation of used probiotics because selected strains exhibited small production of biomass and very low antimicrobial activity. In the third part, the probiotic strain was encapsulated with prebiotic and essential oil into alginate particles. Bacterial growth from the particles in MRS broth was investigated over period of 15 days. Results of this measurement were that probiotics were gradually released from particles over the period. Subsequently the potential use of these particles was proposed for further investigation.
Influence of some medicinal fungi and other prebiotics on growth and metabolism of probiotic bacteria

The bachelor thesis deals with the influence of medicinal mushrooms and other prebiotics on the growth and biological activity of probiotic bacteria. Selected genera of lactic acid bacteria Bifidobacterium and Lactobacillus can produce antimicrobial substances bacteriocins and their production depends on cultivation conditions and available nutrient sources. The theoretical review describes and characterizes probiotic bacteria, their metabolism and importance for human health. Similarly, the theoretical section focuses on prebiotics and their selected representatives, which are four species of medicinal fungi, the microscopic algae Chlorella, resistant starch, the cell biomass of the yeast Saccharomyces cerevisiae and dietary supplements such as inulin and psyllium. In the experimental part, extracts were prepared from these prebiotics by three different methods - boiling, macerating and hydrolysis. The obtained extracts were characterized in terms of macronutrient content such as carbohydrates and proteins. Based on the growth of four species of lactic acid bacteria cultured in standard medium and their production of antimicrobial substances, a strain of B. bifidum was selected for cultivation in the presence of prebiotic extracts. After determining the increase in probiotic biomass in the prebiotic extracts, the cultures were centrifuged, and the protein content of the bacterial supernatant was determined. The lactic acid content of the lyophilized probiotic extracts was determined by high-performance liquid chromatography, and finally, the antimicrobial activity was determined by broth dilution method and resazurin viability assay. The results of the antimicrobial tests were compared with the antimicrobial activity of lactic acid at the same concentration as that contained in the probiotic extracts. For the highest macronutrient content of the prebiotic extracts, extraction by boiling was the most suitable method. For insoluble polysaccharides such as resistant starch, hydrolysis might be a better option, but this requires subsequent treatment of the extract to provide suitable conditions for cultivation. The boiled extracts were the best for the growth of probiotic bacteria as well as their production of antimicrobial substances. The highest concentrations of protein and lactic acid were measured in these samples. The likely presence of bacteriocins was confirmed in all culture types. Almost all probiotic extracts showed antimicrobial activity against Staphylococcus epidermidis and Escherichia coli. The likely presence of bacteriocins was confirmed by the broth dilution method, for example in extracts containing chlorella algae or S. cerevisiae yeast, because these extracts were antimicrobial at lower concentrations than the lactic acid present.

Characterization of selected complex vitamin supplements

Modern society places great emphasis on a healthy lifestyle and balanced diet, leading to increased interest in products with high levels of vitamins, minerals, and other health-promoting substances. Consequently, there has been an increased interest in various dietary supplements, including so-called "greens" products. These supplements are designed to provide a concentrated source of nutrients from various green plants while also containing a high proportion of vitamins, minerals, antioxidants, and other bioactive compounds. The aim of this thesis is to characterize complex food supplements in terms of their composition and content of active ingredients to better understand their mechanisms of action and identify substances responsible for their health-promoting effects. The theoretical part of the work focuses on describing legislation of dietary supplements, including their characteristics and the evolution of interest in them over recent decades. Furthermore, the theoretical part addresses the description of basic components commonly found in supplements, such as vitamins, plants, algae, prebiotics, and probiotics. Finally, methods for determining these components and the most frequently used techniques for their analysis are described. In the experimental part, 12 different plant-based dietary supplements falling into the category known as "greens" were selected. The content of phenolic compounds, chlorophylls, carotenoids, and antioxidants were determined in these products. Additionally, the influence of these products on the growth of bacterial cultures and the metabolic activity of gram-negative and gram-positive bacteria was examined. Analysis of the vitamin C and B-group vitamins content in these supplements was also conducted. The test results showed that the examined supplements are a rich source of phenolic compounds, chlorophyll, antioxidants, and vitamin C. Furthermore, it was found that these products can be used as prebiotics in the diet. Based on the conducted tests, it was concluded that "greens" food supplements offer a wide range of health benefits and can be considered a useful contribution to a balanced diet and a healthy lifestyle. Practically, in all conducted tests, the products with a high matcha content achieved the best results, making this ingredient a key component for creating quality dietary supplements from plant-based ingredients.
Probiotics and prebiotics in the diet of the population. Effect on disease frequency.

Over the past decade, scientific research has focused on the intricate relationship between the human microbiome and the immune system. A particular emphasis in the last three years has been on investigating the impact of the microbiome on the susceptibility and severity of COVID- 19 infections. There is substantial evidence suggesting that the consumption of fermented foods, ranging from fermented vegetables and kefir to yogurt and kimchi, has a beneficial effect on the composition of the intestinal microbiome and, consequently, on the function of the immune system. This study, part of a bachelor's thesis, explores the complex association between regular consumption of fermented foods and the frequency of infectious diseases. Through the analysis of data obtained from a questionnaire, it was found that individuals who regularly incorporated fermented products into their diet exhibited a lower frequency of infectious diseases over the past two years. These results indicate that the consumption of probiotics and prebiotics may positively influence the immune system, reducing the risk of disease. Addition of probiotics to baby food products

This Diploma thesis deals with preparation of probiotic cultures Lactobacillus acidophilus, Lactobacillus casei and Bifidobacterium breve enriched with prebiotics meant for application in baby food products. Natural extracts from matcha, moringa, young beat, young barley, chlorella and spirulina were selected as prebiotics. The theoretical part is focused on probiotic bacteria, their biological effects and their effects on the child´s body. The experimental part deals with the cultivation of probiotic bacteria with plant extracts, monitoring their viability and stabilization in an encapsulated form. Mixtures of probiotic cells with prebiotics were encapsulated into alginate particles to increase stability. Some of the alginate particles were processed by freeze drying. Mixtures of probiotic cultures with plant extracts were subjected to model human digestion by the action of model digestive juices in unencapsulated, encapsulated and lyophilized form. Selected extracts of plant materials were characterized in terms of amount of total and reducing sugars, total phenolic substances, individual phenolic substances and antioxidant activity. Further, two baby commercial dietary supplements containing probiotics were selected, which were characterized in terms of cell number and viability. Probiotic products were also subjected to model digestion.
Biofilm formation in probiotic cultures and its application in pharmacy

The work was comprehensively focused on the development of adhesive forms of probiotics in the form of a biofilm on combined carriers with a prebiotic component. The second part dealed with the influence of food on the multiplication and survival of selected types of probiotic bacteria. Subsequently, the effect of individualized probiotic supplements on changes in the human intestinal microbiome was monitored. Suitable adherent probiotic strains for biofilm formation were selected and tested. Methods have been introduced and different variants of carriers for culturing and binding bacteria have been tested. In vitro experiments verified the stability of biofilm stucture and its resistance to low pH, bile and antibiotics in comparison with the planktonic cell form. The antimicrobial effect of probiotic strains in the form of a biofilm was studied. The cultivation of the multispecies biofilm on the combined carrier was optimized and the stability of the biofilm and the final viability of probiotic bacteria were confirmed. Furthermore, the influence of various foods and beverages on the viability of probiotic bacteria was evaluated with emphasis on the simulation of passage through the gastrointestinal tract. Both models, solutions with standardised concentrations of alcohol, sugar, salts, proteins or different pH and different types of real foods and beverages were tested. The effect of food and beverages was tested on monocultures of Lactobacillus acidophilus, Bifidobacterium breve and on probiotic capsules containing a mixed culture of probiotic microorganisms. The survival of probiotics in various food matrices in the simulated gastrointestinal tract was quantitatively different. We managed to define foods suitable for supporting the multiplication of probiotic bacteria. A separate part of the work was focused on the targeted modulation of the intestinal microbiome by individualized probiotics that were prepared on the basis of molecular biological analyzes of the intestinal microbiome aimed at detecting the percentage of lactobacilli, bifidobacteria and phylum Firmicutes and Bacteroidetes. Personalized probiotic supplementation confirmed the positive effect of this approach on microbiome changes, especially on the increase of the content of lactobacilli, bifidobacteria and the overall diversity of the microbiome.
Encapsulation of probiotics and prebiotics for use in nutritional products for children

The Diploma thesis deals with designing of probiotic dietary supplement for children with strains Lactobacillus acidophilus and Bifidobacterium breve and with prebiotics. Used prebiotics were Inulin, Chia fiber, Bamboo fiber, Chlorella + Spirulina and Yakon syrup. The theoretical part is focused on probiotics, prebiotics and their biological influence. In experimental part the possibilities of encapsulation into alginate particle and lyophilisation of probiotic cells were observed to find their good form to final nutritional product for children. Several types of probiotic with addition of prebiotics were tested in model conditions of gastrointestinal tract. It was found that addition of prebiotic highly increases viability of probiotic cells and their resistance to model conditions of gastrointestinal tract. In this case, the best prebiotic was found in Yakon syrup. The prebiotics were also characterised in terms of nutritional composition, amount of total and reducing sugars, oligosaccharides, proteins, lipids, polyphenols and chlorophyll were obtained. Finally, Chia fiber, Chlorella + Spirulina and Yakon syrup were chosen as prebiotics with best characterisation/properties. In conclusion, a dietary supplement with lyophilized alginate particles containing probiotic cells and with the most appropriate prebiotics were designed. Forms of the product were powder and gummy-bear.
Probiotics and prebiotics in food and other products

Theoretical part of this thesis focuses on present state and research of probiotics and prebiotics, their use for microflora modulation of host and their beneficial effects on the health of individuals. Furthermore thesis deals with efficiency of probiotics strains, which depends on the food matrix and other various factors. The experimental part focuses on the identification of chosen bacterial strain, which is contained in probiotics product. It is realized due the isolation of bacterial DNA by phenol extraction and by use of magnetic particles and subsequent analysis of obtained DNA by polymerase chain reaction (PCR).
Development of probiotic dietary supplement

The thesis deals with a theme of probiotic mikroorganisms which are aplicated as a dietary supplement. Health benefit on the host have been known for a long time, however, there might be some negative effects affecting a consumer. In connection with this observation defining requirements and evaluating of safety of probiotics in food was necessary. The new probiotic dietary supplement can not be expanded on the market without these processes. Every strain must be tested separately on each property. The ability of rezistance against conditions inside human gastrointestinal tract belongs to the most basic tests. The aim of the practical part is testing this ability of rezistance in vitro by simulated conditions. The model of gastrointestinal juices was designed according to the Czechoslovak codex. The form contributes to the higher rezistance of probiotics within implementing to the host. Also a dosage form of probiotics was involved to a in vitro testing.
