Adaptability of electrospun nanofibers for bone-tissue engineering

The field of bone tissue engineering focuses on developing biocompatible scaffolds, which can successfully mimic the extracellular matrix of human tissue and provide the growing bone cells with the necessary mechanical support and adequate stimulation for proper growth and proliferation. This bachelor thesis describes the basic principles of tissue engineering with an emphasis on the area of bone tissue regeneration, examines current technological challenges and discusses prospective materials and technologies for the design of a compatible tissue scaffold. Electrospun nanofibers, their characteristic properties and adaptability are discussed in more detail. Based on the theoretical findings from the research, a design of a suitable scaffold created from the selected material for following seeding of bone cells, and the planned production procedures are presented. According to the design, the tissue scaffold is manufactured and a series of fundamental research analyses are conducted on it in terms of its structural integrity, chemical composition, wettability, crystalline phase content and cell adhesion. The results of these analyses inform the conclusion regarding the tissue scaffold’s adaptability for bone tissue engineering.
Electrospinning of Modified Biopolymers for Medical Applications

Předkládaná disertační práce se zabývá přípravou a charakterizací nových biokompatibilních nanovláken s potenciální aplikací v medicíně. V této práci byl výběr jednotlivých složek pro přípravu nanovlákenného materiálu zvolen tak, aby vyhovoval nárokům tkáňového inženýrství. Literární rešerše shrnuje poznatky o elektrostatickém zvlákňování a o jeho parametrech. Dále se věnuje možnostem elektrostatického zvlákňování proteinů kolagenu a želatiny a jejich směsmi se syntetickými polymery a biopolymery a anorganickými plnivy. Teoretická část řeší také různé postupy síťování nanovláken vedoucí ke zlepšení jejich hydrolytické stability a mechanických vlastností. Poslední část je zaměřena na anorganické nanotrubky halloysitu (HNT), které získaly svou pozornost díky svým vynikajícím fyzikálním a biologickým vlastnostem. V experimentální části byly zpracovány dvě případové studie, z nichž každá se zabývá přípravou nanovlákenných biomateriálů s potenciální aplikací v medicíně. První studie je zaměřena na přípravu a charakterizaci nových hydrolyticky stabilních antibakteriálních želatinových nanovláken modifikovaných pomocí oxidované celulózy. Unikátní inhibiční účinky nanovláken byly testovány na kmenu bakterie Escherichia coli pomocí metody chemické bioluminiscence. Kultivované buňky lidského papilárního adenokacinomu plic prokázaly dobrou adhezi a proliferaci k povrchu nanovláken. Druhá část popisuje vliv zdroje a množství anorganických halloysitových nanotrubek na strukturu a vlastnosti amfifilních nanovláken ze směsi želatiny a syntetického polykaprolaktonu. Přídavek HNT zlepšil tepelnou stabilitu, mechanické vlastnosti (jak tuhost, tak prodloužení) a snížil krystalinitu nanovláken. HNT z různých zdrojů neměl vliv na chování buněk, ale mírně ovlivnil proliferaci a životaschopnost buněk na povrchu nanovláken.
Biomedical applications of polycaprolactone nanofibrous mats

The diploma thesis deals with the treatment of polycaprolactone (PCL) nanofibers. PCL fibers were subjected to the deposition of plasma amine polymers in a low pressure pulsed radiofrequency capacitively coupled discharge using cyclopropylamine monomer (CPA). Collagen as an extracellular matrix (ECM) protein was immobilized and cell proliferation on the modified nanofiber surface was monitored. Untreated PCL fibers were also subjected to the deposition of an antibacterial copper layer, and the fibers were characterized by scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and energy dispersive spectroscopy (EDX).
Wound dressing nanofibers mats fabricated from nanocomposite material

Boli pripravené kryty rán na bázi prírodných látok polyvinyl alcohol/ hyaluronan/ strieborné nanočastice (PVA/ HA/ Ag-NPs). Hyaluronan bol použitý ako redukčné a stabilizačné činidlo pre syntézu nanočastíc striebra. Pri príprave Ag-NPs boli testované viaceré parametre ako koncentrácia dusičnanu strieborného ako zdroja Ag-NPs (0,01; 0,1;0,5;1 M), koncentrácia kyseliny hyalurónovej (1,2 %) a jej rozdielna molekulová hmotnosť. Kryty rán z nanovlákien boli pripravené pomocou techniky electro-spinning z roztokov líšiacich sa pomerom PVA a HA/Ag-NPs (100; 90/10; 80/20; 60/40; 50/50). Vlastnosti nanokompozitu HA/Ag-NPs boli hodnotené pomocou TEM, reológie, DLS, XRD, UV/Vis spektroskopie a kryty rán boli charakterizované pomocou SEM, TGA, FTIR a ťahovej skúšky.
PREPARATION AND CHARACTERIZATION OF BIOPOLYMER-BASED NANOPARTICLES AND NANOFIBERS AND THEIR APPLICATION IN FOOD AND COSMETICS

The presented dissertation thesis deals with the polymer polyhydroxybutyrate and other biopolymers as a basic building block for the construction of micro- and nanoscopic structures and materials used in food and cosmetics. In the theoretical part, current literary review is prepared to introduce the basics of this application field. The practical part of the work is composed of three blocks developed during the doctoral study. In the first and most important part are summarized comments to the academic and patent outputs, where among the academic ones it is possible to find two peer-reviewed articles dealing with the electrostatic and wet spinning of PHB and properties of prepared materials. The patent outputs consist of several accepted and applied projects, which summarize results on both PHB spinning methods, but also on general approaches enabling the processing of PHB into forms enabling many applications in food and cosmetics. Second part was focused on the patented composition of the UV protection cream based on the prepared nanoscopic and micro- morphologies of PHB. The third block summarizes results focused predominantly on the electrostatic spinning of PHB and other biopolymers. Finally, a short chapter containing a brief description of projects that were in a way related to the dissertation topic, but rather practical development work in Central Tanzania and West Africa, which draw on knowledge and contacts gained during studies at FCH BUT Brno.
Electrospinning and characterization of fibrous structure for piezoapplications

The master’s thesis deals with the preparation and characterization of PVDF-based composite fibrous layers prepared by electrospinning. Ceramic particles TiO2, BaTiO3 and BCZT were used as fillers. The fibers were prepared containing 10, 20, 30, 40 and 50% of these particles and then their morphology, piezoactive phase representation and dielectric characterization were characterized. The effect of the presence of particles on the fiber diameter and on the crystallinity fraction of the polymer was shown. Particle size significantly influenced their distribution in the fibers. Piezoelectric particles further influenced the content of electroactive phases beta and gamma. The highest proportion of electroactive phases was determined in BaTiO3 filled fibers, which reached up to 98%. For the BaTiO3 filled fibers, the dielectric constant values were also measured, which reached lower values than expected due to the high porosity of the prepared fibers The as-prepared fibers can be considered promising not only for piezoapplications, therefore, optimization of their preparation and measurement of electrical characteristics will be further studied.
Piezoelectric nanofiber materials for wearable electronics

This thesis deals with the influence of production parameters on the morphology of PVDF nanofibers produced by electrospinning. Nanofibers are generally a widely used material, nanofibers are generally a widely used material, they can also have piezoelectric properties, which gives them a specific spectrum of use, eg in biosensors. The production of nanofibers was realized by electrospinning. A scanning electron microscope was used to examine the morphology and surface properties. The aim of this thesis is to compare the properties of PVDF nanofibers produced under different settings of production parameters.
Characterization of PVDF material in nanoscale resolution

Tato práce se zabývá charakterizací nanovláken na bázi polyvinylidenfluoridu. Zaměření práce je na piezoelektrické vlastnosti vlákna, které jsou studovány metodou piezoelektrické silové mikroskopie. Takto byly měřeny dva typy odlišných vzorků, které se lišily v parametrech výroby. Odlišnosti vláken v jejich fázovém složení byly také zkoumány za využití Ramanovy spektroskopie a infračervené spektroskopie s Fourierovou transformací. Chemická analýza povrchu a jeho stavu proběhla pomocí rentgenové fotoelektronové spektroskopie. Různé uspořádání nanovláken spolu s jejich průřezem bylo pozorováno rastrovacím elektronovým mikroskopem za využití fokusovaného iontového svazku. Rovněž byla zkoumána smáčivost a kontaktní úhel povrchu vzorků s demineralizovanou vodou. Bylo zjištěno, že vyšší rychlost otáček válce během procesu elektrostatického zvlákňování má velmi významný vliv na jejich uspořádání a tím i na parametry ovlivňující tvorbu piezoelektrického jevu a dalších materiálových vlastností.
Preparation and characterization of nanofiber wound dressings

The presented bachelor thesis deals with the preparation and characterization of nanofiber wound dressings. The materials used for the preparation of nanofibers were poly(3-hydroxybutyrate) and gelatin. Active substances, such as ampicillin and ibuprofen, were incorporated into the wound dressings. In the theoretical part of the work skin, wound healing, and nanofibers and their properties were described. This part also deals with the use of nanofibers as wound dressings. Additionally, it depicts materials suitable for the preparation of nanofiber wound dressings. Furthermore, this section contains a review focused on various methods of preparation of nanofibers, drugs that can be incorporated into the fibers and the subsequent determination of the safety of the prepared dressings. In the experimental part, nanofiber wound dressings containing ampicillin and ibuprofen were produced. The maximum possible amount of active substance feasible to be incorporated into the nanofibers has been determined. Subsequently, combined wound dressings were prepared to study the release rate of active substances into the model environments. The release in the time intervals was determined spectrophotometrically. Finally, antimicrobial activity assays were performed to test the effect of dressings containing ampicillin on selected strains of microbiirganism: S. Epidermidis, E. Coli and M. Luteus
Electrospun fibers based on PVDF and nylon

Polymer nanofibers used for the construction of triboelectric nanogenerator (TENG) and piezoelectric nanogenerator (PENG) are new and promising technologies for energy recovery. Thanks to the generation of electrical energy based on mechanical movement (deformation), these fibers can find application in the field of self-powered electronic devices. In this work, three nanofibrous structures of materials were prepared by electrostatic spinning: pure polyvinylidene fluoride (PVDF), pure polyamide-6 (PA6) and their mixed combination PVDF / PA6. Non-destructive analyzes such as Raman spectroscopy, FTIR, XPS and electron microscopy were used to study the properties of nanofibers. Analyzes confirmed the positive effect of electrostatic spinning of polymers on the support of the formation of highly polar crystalline -phase in PVDF and , -phase in PA6. The structure arrangement of the nanofibrous material and their defects were observed by scanning electron microscopy (SEM). Furthermore, the contact angle of the wettability of the liquid on the surface was measured for the materials, and the permittivity was measured to monitor the dielectric properties. The described results make the mixed material PVDF / PA6 very promising for further research in the field of nanogenerators and functional textiles.
