Amylase gene variability in human populations

Amylase is a starch-digesting enzyme produced by the pancreas and salivary glands in humans. Genes for the salivary isoenzyme (AMY1), located on chromosome 1p21.1, show remarkable variability in their copy numbers (CNV). Their count is positively correlated with the final amount of the produced enzyme. This variability could be the result of positive selection during human evolution, depending on the amount of starch consumed by various populations. Starch pre-digested by salivary α-amylase is absorbed more effectively, resulting in a greater gain of energy in later stages of the metabolizing process. Thus, it could have been one of the factors for rapid brain growth in humans and, significantly later, even for the agricultural transition. It could belong to a group of other known subsistence-based evolutionary changes in the human genome, such as those resulting in lactase persistence or slow acetylation of xenobiotics. Nowadays, the number of copies and the amount of produced α-amylase can influence the glucose level in the blood and insulin production after consumption, as well as the incidence of certain civilization diseases, such as obesity or type II diabetes. Hypoglycaemia detection using smart devices

This work explores the possibility of using a smart device for hypoglycemia detection. Hypoglycemia is a dangerous condition in individuals with diabetes mellitus. The problem mainly occurs during sleep when a person is unable to react promptly. The aim of this study is to capture data to determine which symptoms (heart rate or electrodermal activity) are influenced by hypoglycemia. Data was collected during sleep using a smart device. Simultaneously, blood glucose levels were measured as a reference using a glucose monitoring sensor. The values were preprocessed, and subsequently, features were selected to identify hypoglycemia. Based on these selected features, a model for detecting hypoglycemia was created.
Nursing care of a paediatric patient with diabetes mellitus type I.

The bachelor thesis is focused on the care of children with type I diabetes mellitus by nuirses working in diabetes clinics. In the theoretical part, there are characteristics of diabetes mellitus, its classification causes, symptoms, diagnosis, complications, treatment, and also the specific needs of paediatric patients diagnosed with type I diabetes mellitus. The first aim of this bachelor thesis was to evaluete the current state of nursing care of paediatric patients with type I diabetes mellitus. The second aim was to find out and identify the specific needs of paediatric patients with type I diabetes mellitu. The third aim was to try to suggest improvements in nursing practices and strategies for paediatric patients with type I diabetes mellitus based on the findings. The pactical part of the bachelor thesis was elaborated by using a qualitative research method. We used a questionnaire method and a semi-structured interview technique to collect the data. The research file consisted of nine nurses working in the Children's Hospital in Brno in diabetes outpatient surgeries with different length of experience. After transcribing the interviews with the respondents verbatim, we analyzed the results by using open coding using the pencil and paper method, where we esstablished a total of seven categories: age of children with type I diabetes mellitus, initial symptoms of children with type I diabetes mellitus, education, violation of treatment regimen, improvement of treatment regimen, limitations of diabetes in personal life, and visit to the diabetes clinic. The results of the research are presented in the various tables and charts of the empirical part of this paper. The research shows that the most common age of children with type I diabetes mellitus is preschool age. Initial symptoms in children with type I diabetes mellitus were most commonly reported by the respondents as frequent urination, excessive thirst, abdominal pain, and lack of appetite. Only one respondent stated that she did not observe any symptoms. Insulin administration was the most frequently reported educational intervention. Within the treatment regimen violation, not applying insulin and improving the treatment regimen, educating the children resulted the most. Limitations of diabetes in personal life most of the respondents said that they do not have any limitations. According to the most frequent answers of the respondents, the visit to the diabetes clinic is regularly once in three months. We believe that the results of this bachelor thesis could be used as educational material in diabetes clinics for paediatric patients to extend information about proper diet and lifestyle.
Health assessment using smart devices

This thesis deals with the possibilities of non-invasive determination of blood glucose from photoplethysmographic signals. Elevated blood sugar is often associated with disease called diabetes mellitus. Diabetes is one of the world’s major chronic diseases. Untreated diabetes is often a cause of death. The aim of the work is to propose methods for glycemic classification and prediction. Two datasets have been created by recording the PPG signals using two smart devices (a smart wristband and a smartphone), along with their blood glucose levels measured in an invasive way. The PPG signals were preprocessed, and suitable features were extracted from them. Various machine-learning models for glycemic classification and prediction were created.
Mobile Application for Diabetes Monitoring

In the present time, all processes are automated. They are moving to smaller devices. Many people with diabetes, instead of prescribed diabetics journals, use mobile applications that help and simplify work with diabetes. This thesis analyzes and compares existing apps. After all, it describes draft of the application and its implementation. The result is verified by testers and publicly posted to production.
Detection and Recognition of Diabetes Disease Impacts to the Human Eye Retina

This bachelor's thesis deals with the design of algorithms for the recognition of a diabetes disease impacts to the human eye retina. Diabetic retinopathy is one of the most common diseases aecting the retina and its consequences lead to partial or complete weakness. The basis of the algorithm for detection is to create candidate areas from dierent viewpoints of image processing - computer vision and their subsequent analysis. Core components of the retina have impacts to detection results - optical disc and blood vessels, which need to be properly detected and subsequently excluded from processing. Testing the implemented application took place in 68 images selected from two databases. One of the possible uses of the proposed methods in the future is in combination with the retinal scanning device for the automatic detection of diabetes symptoms during the retinal screening process.

This thesis deals with accuracy measurement of personal glucometers. The theoretical part is focused on processing information about glucose and its role in organism, diabetes mellitus, procedures and devices used in self-monitoring of glucose. The next part is focused on preparation of practical part, especially describing available personal glucometer, testing its measuring accuracy, the calculation of necessary measurements to perform the experiment and a description of the reference sample for data measurement.
Prebiotic sweetener tagatose

This sweetener, D-tagatose, has numerous advantages, which would eventually be able to use in the food industry. It summarizes the properties of Dtagatose, physical and also chemical, the possibility of production from lactose or bacterial action on Dtalitol. The basic information about the properties of Dtagatosy also discusses the actual metabolism of Dtagatose, prebiotic properties and their effect in the gastrointestinal tract. Dtagatose is a proposed new sweetener, because the possibility can not be missed in certain applications tagatose in a different types of foods for example confectionery. And positive qualities in comparison with sucrose. This comparison is made to examples of disease associated with excessive use of conventional sweeteners sucrose. This paper also discusses possibilities Dtagatose determination in foods by different methods.
Fluorescenční sondy pro detekci sacharidů a glykovaných proteinů pomocí kapilární elektroforézy

Glycated albumin (gHSA) and glycated hemoglobin (HbA1c) represent important biomarkers assessing the progression of diabetes mellitus disease. The aim of this thesis was to develop and optimize a method for the simultaneous determination of these glycated proteins from human blood by capillary electrophoresis using the fluorescent probe ABAM (anthracene boronic acid methacrylate). Thanks to this new method, it was possible to selectively determine these two proteins with a resolution of 5.70 and the LOD of 11.9 µg/ml for gHSA and 26.5 µg/ml for HbA1c. Subsequently, the optimized method was successfully applied to a real human blood sample. The simultaneous determination of these biomarkers can help to better monitor dynamic changes during diabetes and enable its monitoring in cases, where HbA1c assessment is inappropriate for medical reasons (anemia, etc.).
