Viruses and cytosketelon of the cell nucleus
Cibulka, Jakub ; Forstová, Jitka (advisor) ; Šťovíček, Vratislav (referee)
The nuclear cytoskeleton (the nucleoskeleton) provides a structural integrity to the nucleus and is involved in number of key processes including transcription, chromatin remodelling and mRNA transport. The nucleoskeleton consists of nuclear lamins, nuclear actin and other proteins. Some viruses, which replicate themselves in the nucleus, use nuclear cytoskeleton in their life-cycle. On the other hand the nucleosketon may also represent a barrier for viral infection. Herpesviruses need nuclear actin for capsid assembly and transport, but they have to desintegrate the nuclear lamina in order to escape the nucleus. Nuclear actin also participates in the morphogenesis and probably nuclear export of baculovirus capsids. Some retroviruses transport their unspliced RNAs from the nucleus using nuclear actin and there is also some evidence of retrovirus-induced nuclear lamina disruption. In this work, I focus on the interactions of above-mentioned viruses with the nuclear cytoskeleton (namely nuclear actin and lamins).
Preparation of expression vectors and virus mutants for studies of the minor structural proteins of polyomaviruses.
Cibulka, Jakub ; Forstová, Jitka (advisor) ; Šroller, Vojtěch (referee)
Polyomaviruses are small non-enveloped DNA viruses infecting birds and mammals, including human. Their capsid consists of the major capsid protein, VP1, and two minor capsid proteins, VP2 and VP3. The VP2 and VP3 proteins are supposed to have an important function in the transport of viral genome into the cell nucleus, which is a key step to facilitate viral replication. VP2 and VP3 proteins of mouse polyomavirus and SV40 have an ability to bind and disrupt cellular membranes. This feature is believed to be involved in the transport of viral genome into the nucleus. Plasmids carrying genes of the minor capsid proteins of Merkel cell polyomavirus were prepared in order to produce and visualize these proteins in mammalian cells. These proteins are known to have very unusual sequences compared to other human polyomaviruses or related mouse polyomavirus. When produced alone, the minor capsid proteins of Merkel cell polyomavirus did not significantly interact with cellular membranes, unlike the minor proteins of the mouse polyomavirus. The second goal of this work was to prepare mouse polyomavirus mutants with deletion in hydrophobic domains of VP2 and VP3 proteins. These domains are likely responsible for the mentioned membrane interactions. Prepared mutants were non-infectious. The loss of infectivity was not...
Online Comparators in Insurance Business
Slavík, Jakub ; Ducháčková, Eva (advisor) ; Cibulka, Jakub (referee)
This bachelor thesis deals with problems of on-line insurance comparators. The theoretical part is devoted to the importance of insurance products, the pricing of insurance products, distribution channels of insurance products, on-line comparators and insurance products that can be arranged on them. Practical part deals with depth analysis of clients' awareness, behaviour and needs on the market of on-line comparators and the experience of clients with comparators, evaluates priorities, why they arrange insurance in this way and maps benefits to clients. The results of the questionnaire survey are formulated in the hypothesis evaluation. The conclusion summarizes its own findings and evaluation.

Factors affecting mortgage market
Mrázek, Michal ; Radová, Jarmila (advisor) ; Cibulka, Jakub (referee)
This bachelor thesis focuses on current issues regarding mortgage loans. The thesis aims to characterize the impacts of new regulations and selected macroeconomic factors on the mortgage market. The first part defines mortgage loans, including relevant terms connected to the field. Then, the impact of new legislature on banks and consumers in the mortgage business is researched. Furthermore, the thesis focuses on reccomendations of Czech national bank concerning indicators of LTV, DTI and DSTI. The second part characterizes selected macroeconomic factors and their impact on the mortgage market. The selected macroeconomic factors are gross domestic product, inflation, unemployment and interest rates of mortgage loans. Firstly, the impact of these factors is researched on quantity and sum of new mortgage loans. Then, the macroeconomic indicators are quantified and analyzed using regression analysis.
Analysis of the Motor third party liability on the Czech market
Janatová, Denisa ; Radová, Jarmila (advisor) ; Cibulka, Jakub (referee)
The aim of this bachelor thesis is to analyze offers of compulsory liability on the Czech market. The whole analysis is to compare the price offers of the third-party liability insurance at Partners with insurance companies and e-portals such as E-Insurance,, Compare24 and finally with the Mandatory portal. The first chapter describes one of the basic pillars of non-life insurance, namely liability insurance. There is also a breakdown of responsibility according to the three most important factors. The second chapter deals specifically with Motor third party liability insurance and the specific requirements of this insurance that every car owner should know. The third chapter briefly mentions the vehicle's accident insurance. The fourth chapter shows the percentage comparison and comparison of the individual parts of the non-life insurance market in the Czech Republic. At the same time, the comparison of the shares of all the insurers providing compulsory liability to the insurance policies. The fifth and final chapters analyze market prices for a predefined vehicle and person. The object is also to point out the differences in the accident insurance and to increase the total insurance premium due to the arrangement of supplementary insurance.
Analysis of the Creditworthiness of Vítkovice a.s.
Svatová, Ivana ; Cibulka, Jakub (advisor) ; Plíva, Rostislav (referee)
The aim of the bachelor thesis is to describe the methods used by the bank to determine the rating of client and to evaluate the creditworthiness of Vítkovice a.s. In the theoretical part of the thesis is defined credit risk and internal rating models of banks that are used in the assessment. Internal rating models vary from one bank to another. However, the basis for assessing the creditworthiness is the analysis of quantitative and qualitative indicators, which are explained in more detail in chapter three. The credit registers presented in chapter four also play an important role in assessing the client. In the practical part, the selected quantitative indicators are calculated. The results of the indicators are then compared with the sector. The conclusion of the work is a review of the company and a recommendation whether it is appropriate to provide the client with a loan.
The fundamental analysis of the company Pegas Nonwovens
Hladík, Josef ; Kábrt, Tomáš (advisor) ; Cibulka, Jakub (referee)
This bachelor thesis is dedicated to the fundamental analysis of the company Pegas Nonwovens. The purpose was to execute global, industrial and company fundamental analysis and to determine intrinsic value of the stock. Based on the previous, the aim was to suggest an investment recommendation. On global level I analyzed the relationship between the development of significant macroeconomic factors and the stock value. In the industrial part of the analysis I examined the characteristics of nonwovens industry, such as market structure or influence of regulatory authorities. In part dedicated to company fundamental analysis I dealt with calculation of intrinsic value by various methods. Based on the whole analysis and calculated intrinsic value I suggested an investment recommendation, according to which is the stock of Pegas Nonwovens underestimated and suitable for buying.
Comparison of crowdfunding regulation
