The aim of this thesis is to analyse the possibilities of influencing the antibiotic prophylaxis (AP) on the basis of those markers observed on patients that underwent the total hip arthroplasty (THA) and the total knee arthroplasty (TKA). Methods: Data was collected from a prospective study which began in March 2020 at the Department of Orthopedics at the University Hospital Hradec Králové. There were 118 patients from May 2021 to June 2022 older than 18 years with THA and TKA included in the analysis. The aim of this thesis was to analyze the administration of drugs to patients hospitalized by nurses in one of four studied health facilities in the South Bohemia region in the surgical, internal medicine and aftercare wards. Methods: This diploma thesis is part of an observational-interventional study, in years 2020- 2023. Observations took place in internal medicine, surgery and aftercare wards. Collection of data was held from 23th May to 26th May 2022 by a multidisciplinary team made up of a pharmacist and a nurse by direct observation method. Methods: This is a part of an extensive observational and intervention study, which is conducted between years 2020-2023. One of these measures is antibiotic prophylaxis (AP), thus the administration of an appropriate antibiotic or antibacterial chemotherapy (ATB) at the prescribed time, in sufficient dose and for sufficient duration to produce the desired effect. The aim of this study was to analyze the antibiotic administration in prophylaxis in relation to surgical procedures in a district type hospital and to compare these results with the hospital's recommended practice (DPNM), international recommended practices (ASHP-G) and a review of the latest scientific knowledge in this area (NVP). AIMS Aim of this work was to describe drug related problems occurred during drug administration in health care facility. METHODS The data was collected at three different departments (follow-up care, surgery and neurology) in one hospital in Olomouc region. The method of direct observation was chosen. The morning, midday and evening drug administration was observed. The observation was taken time 3 days in every department. Identification of a patient, hand hygiene and check of use were observed. Manipulation of oral tablets and capsules, oral liquids and eye drops and ointments were observed. Evaluation of compressibility of paracetamol

The objective of this work was to evaluate the compressibility of paracetamol by determination of compression and viscoelastic properties of the active substance and also to detect eventual changes in crystal modification of the substance during the compression process. The parameters of the compression equation were determined using tree-exponential equation developed at the Department of pharmaceutical technology. Mutual interactions between particles were also studied by stress relaxation test using compression forces 5, 10 and 15 kN, as well as energetic parameters of a force-distance record using compression forces from 5 to 40 kN. Tablets made in the last mentioned test were used as samples for DSC analysis to seek potential changes in the crystal structure of paracetamol when undergoing the compression process. The evaluation of the executed tests suggested that compression ability of paracetamol in basic form I is very poor and its plasticity when being compressed by commonly used pressures is about 50%, which is considered as very low. It was also found out that any crystal structure changes of the active substance did not occur during compression.

Title of diploma thesis: The assessment of inhibitory effects of selected targeted anticancer drugs on the activity of ABC drug eflux trasporters. Lung cancer is the leading cause of death within oncological diseases. Non-small cell lung carcinoma (NSCLC) accounts for about 85% of all lung cancer, and its major subtypes include adenocarcinoma and squamous cell carcinoma. In addition to surgery, radiotherapy and chemotherapy, the use of targeted low-molecular substances, which target tumor cells with higher specificity, has recently been used in treatment. The two main causes of death in cancer patients are the formation of metastases and the development of multidrug resistance (MDR). This may also be caused by overexpression of the efflux transporters. ATP-binding cassette (ABC) transporters are groups of transmembrane pumps that use energy in the form of ATP to transfer a wide range of substrates. In particular, P-glycoprotein (ABCB1), breast cancer-resistance protein (ABCG2) and multidrug resistance-associated protein 1 (ABCC1) are associated with MDR.
