The role of cyclophilins in splicing

Splicing is a process essential for the proper functioning of eukaryotic cells. It is a complicated and highly dynamic process, participated by large numbers of proteins which perform diverse functions, either directly within the splicing complex, or sometimes outside of it. Among the proteins, which play an important role in splicing, are cyclophilins. Cyclophilins probably cause conformational changes in the other components of splicing complex. They can also maintain them in the desire conformation thereby they contribute to the dynamics of the spliceosome. This work provides an overview of cyclophilins, which were confirmed to participate on pre-mRNA splicing, and summarizes suggestions of possible functions that these proteins may perform.

The protein p130Cas (human ortholog BCAR1) is a major substrate for phosphorylation by the Src family kinase and plays a central role in oncogenic transformation. Increased level of BCAR1 correlates with primary tumour growth and cancer progression. Localized to focal adhesion, p130Cas serves as a mechanosensor and mediates key interactions with the extracellular environment. The structure of p130Cas is crucial for its function, mainly the anchoring domains SH3 and CCH, together with the substrate domain which is extended when under tension. This Master's thesis presents a newly developer FRET mechanosensor based on the structure of p130Cas. The sensor utilizes the anchoring domains of p130Cas for proper localization to focal adhesions, where it can detect tension in living cells. Key words: p130CAS, FRET, focal adhesions, mechanosensing

Formování sestřihových snRNP v buněčném jádře

1 ABSTRACT There are many structures, suborganelles and bodies in the eukaryotic cell nucleus. These domains provide the nucleus with many specific functions. Nucleolus is specialized compartment serves to ribosomes assembly, nuclear speckles or Splicing Factors Compartment play an important role in RNA processing and best studied of them, Cajal bodies (CBs), are involved in snRNP maturation. However, non-membrane substructures are not unique for cell nucleus; processing bodies (P bodies) found in the cytoplasm are proposed to be important places in mRNA degradation pathway. This work is a compilation of four projects focused on non-membrane cellular bodies; namely, nuclear CBs and cytoplasmic P bodies. Both CBs and P bodies are dynamic structures that continuously exchange their components with surrounding environment. In addition to a widely accepted role of CBs in snRNP biogenesis, we show that the CB serves as a place where snRNPs are regenerated after each round of splicing. Thus, CBs are important nuclear compartment involved in snRNP recycling. To further characterize tri-snRNP assembly in CBs we applied kinetic experiments combined with mathematical modeling and created a kinetic model of tri- snRNP formation in the CB that determined kinetic parameters of tri-snRNP formation. The thesis deals with the work of Mgr. Jiřina Lhotská and her contribution to the drama in education in the South Bohemia County. The process of drama in education teaching and its basic elements are described in the theoretical part of the thesis. In the second part focuses on the character of Jiřina Lhotská, her career and contribution to the drama in education teaching. Furthermore, the practical part also contains interviews with individuals who profoundly influenced Jiřina Lhotská the pedagogue. The interviews with her colleagues and students with whom she has cooperated during various stages of her career (up until now) is also included therein. The lists of Jiřina Lhotská's creative activities, selected scripts of certain acts, as well as some photographs form part of multiple appendices.
