Inhibition of TET-1 protein by iron chelators

EN The objective of this work was to study novel synthetic iron chelators and to study their potential use in inhibition of TET1 (ten-eleven translocation methylcytosine dioxygenase 1) protein. Epigenetic mechanisms, such as hydroxymethylation of DNA, are promising target in many serious pathologies, including oncological disorders. In the presented study, we intended to discover novel inhibitors by screening small libraries of heterocyclic molecules, such as pyrrolo[3,2-b]pyrrole derivatives with hydrazide (compound 1) or hydrazone (compound 2-6) iron-binding group and hydrazone-based iron chelators (compound 7-10). Within the scope of this study, we used various analytical and biochemical techniques for the purpose of characterization of novel cancer therapies based on TET1 protein inhibition. The absorbance and the complexation of tested compounds with Fe(II) ions was studied by UV-Vis spectroscopy. Inhibition of TET1 protein was studied by fluorometric assay based on ELISA and further supported by microscale thermoporesis and in silico docking. The cytotoxicity of compounds on cancer cell lines was measured by MTT assay and intracellular distribution was determined by live-cell imaging. Utilization of light emitting diode lighting for cycle tracks

Kinetics of Inkjet Dyes Degradation

The stability of inkjet print is influenced by a lot of factors. Mutual effects of these factors accelerate the print degradation. The surrounding environment in image stability plays an important role, when the prints degrade especially by the light. The degradation of inkjet prints is presented as a decrease of dye or multiple dyes. It is necessary to know the dye concentration for the dye decrease prediction in the time. This dissertation thesis deals with the study of kinetics and changes in electron and molecular structure of digital photography prints after accelerated ageing tests. The study of resistance of inkjet prints was realized on one type of media using three different sets of inks. Changes in printed colours were measured and evaluated in calibration (by PLS calibration and least squares method). On the basis of calibration the dye decrease prediction of real samples in receiving layer was evaluated. Changes in electron and molecular structure were analysed on KBr pellets by FTIR an UV-Vis spectroscopy.
Printed Biosensor Based on Organic Electrochemical Transistor

Organické elektronické zariadenia sú vyvíjané ako vhodné riešenia senzorov pre bioelektroniku, a to najmä kvôli dobrej biokompatibilite organických polovodičov v nich použitých. Takzvané biosenzory dokážu premeniť elektrochemické procesy na elektronický signál. Matrica takýchto biosenzorov môže simultánne skenovať množstvo biologických vzoriek, alebo rôznych tkanív v živých systémoch. Aktívnou súčasťou zariadenia je organický elektrochemický tranzistor (OECT). V tejto práci je diskutovaný teoretický rámec fungovania takéhoto zariadenia, jeho elektrická charakterizácia, aplikácia v biosenzoroch na báze buniek, spôsoby výroby a aktuálnym stavom techniky v oblasti organickej elektroniky. Experimentálna časť obsahuje konkrétne výrobné postupy vývoja OECT zariadení, ktoré boli použité v našom laboratóriu. Hlavný dôraz sa kladie na schopnosť vyrobených zariadení detekovať reakciu a monitorovať stimuláciu elektrogenných buniek. Za týmto účelom boli vyvinuté matice mikroelektródových OECT zariadení založených na polovodivom polyméri PEDOT:PSS. Tieto boli vyrobené s využitím bežnými tlačiarenských techník (atramentová tlač a sieťotlač) spolu so štandardnými litografickými postupmi. Najnovšie nami vyvinuté zariadenia dosahujú najväčšieho zosílením signálu, g = 2,5 mS a časovú konštantu t = 0,15 s. Tieto zariadenia sú porovnateľné, často dokonca lepšie ako niektoré iné najmodernejšie a plne litograficky pripravené senzory.
Characterisation of the Physical Chemical Processes Using the Fractal and Harmonic Analysis

Existuje mnoho různých způsobů jak analyzovat disperzní systémy a fyzikálně chemické processy ke kterým v takových systémech dochází. Tato práce byla zaměřena na charakterizaci těchto procesů pomocí metod harmonické fraktální analýzy. Obrazová data sledovaných systémů byly analyzovány pomocí waveletové analýzy. V průběhu práce byly navrženy různé optimalizace samotné analýzy, převážně zaměřené na odstranění manuálních operací během analýzy a tyto optimalizace byly také inkorporovány do softérového vybavení pro Harmonickou Fraktální Analýzu HarFA, který je vyvíjen na Fakultě chemické, VUT Brno.
Study of Biological Material Attributes by using Image Analysis Methods

Within the dissertation thesis “Study of Biological Material Attributes by Using Image Analysis Methods”, attention is focused on monitoring of the application of image analysis methods, mostly a fractal analysis, in studying the properties of various yeast species. The thesis includes determining the number of yeast cells and vegetative propagation of yeast using fractal parameters – fractal measure D and fractal dimension K. Attention is also paid not only to the application of the existing image analysis methods, but also to their renovation. The obtained images were evaluated using the box counting method specified by implementation of wavelet transformation. To monitor yeast cells for a longer time, it was first necessary to prepare a suitable microscopic preparation. To distinguish live and dead cells, the following fluorescent dyes were used: acridine orange, fluorescein diacetate, FUN-1, and Calcofluor White M2R. The images of yeast cells were recorded using a still camera or a CCD camera and microscope. Clips of the same size were obtained from the acquired digital photographs and processed by the HarFA program developed at the Faculty of Chemistry, Brno University of Technology. On the results it is possible to see a change in the fractal dimension depending on time, i.e. on the change of a budding cell structure, or to determine the number and radius of yeast cells upon predefined conditions.
Study of Electric and Dielectric Properties of Ionic Liquids

Dizertační práce je zaměřena na studium elektrických a dielektrických vlastností iontových kapalin. Iontové kapaliny mohou nacházet uplatnění v široké škále aplikací, především pak v elektrotechnice. Teoretická část se věnuje popisu základních vlastností iontových kapalin a možností jejich uplatnění v kondenzátorech a elektrochemických senzorech plynů. Experimentální část se věnuje použitým metodám charakterizace vlastností iontových kapalin, jsou zde popsány teoretické poznatky o dielektrické spektroskopii včetně metod stanovení fyzikálních vlastností a vyhodnocení experimentálních dat. Experimentální část je rozdělena na dvě základní části. První se věnuje studiu iontových kapalin pro použití jako elektrolytu v kondenzátorech, druhá část se věnuje studiu vlastností připravených experimentálních senzorů na NO2, zejména vlivem vlastností iontových kapalin na sledované parametry senzoru.
Preparation of graphene quantum dots and study of their properties

Current methods for preparing graphene quantum dots (GQDs) rely on oxidation and reducing agents or require energy-intensive and technologically demanding preconditions. Applying microwave expansion and liquid phase exfoliation (LPE) in a sample of graphite powder enabled us to prepare GQDs that exhibit strong luminescence in the blue region of the visible spectrum. The proposed technique for synthesizing GQDs is energetically undemanding and does not necessitate additional chemical components.
Study of dielectrical properties of organic material thin films

The dissertation is focused on the study of electric and especially dielectric properties of thin film organic materials with their huge potential for optoelectronics and other industrial sectors. The theoretical part deals with the use of organic materials in organic photovoltaic cells, the methods of deposition techniques and characterization. The theoretical knowledge of the dielectric spectroscopy, including methods for determining the physical properties and evaluation of experimental data are also described. The experimental part is devoted to the study of small molecule organic solar cells with bulk heterojunction composed of electron donor molecule of DPP(TBFu)2 and electron acceptor fullerene derivatives, such as PC60BM, PC70BM and TC60BM. The experimental part is divided into two main parts. The first part deals with the study of processes occurring at the interface between the active layer and the contact; the second part is focused on transport processes inside the structure of photovoltaic cells and also contains a study of perovskite solar cells.
Study and mechanism of new-generation heterocyclic cytostatic with an emphasis on IL-6 andIL-8

EN The purpose of this study was to screen for novel synthetic inhibitors of interleukins IL-6 and IL-8, which are promising targets for cancer therapy. Small molecules are an attractive approach to inhibit these signalling pathways, which are known to be important for tumour cell growth and survival. The study aimed to discover new inhibitors of IL-6 and IL-8 signalling pathways by screening a small library of chemical derivatives, including π-expanded naphthalimide derivatives, Tröger's base, and pentamethinium derivatives. The structures of the compounds were designed based on in silico studies focusing on blocking the IL-6 and IL-8 signalling pathways. The study involved the use of various analytical and bioanalytical methods for the in vitro analysis of new potential anticancer drugs. The compounds were tested for cytotoxicity and their effect on cell growth using an MTT proliferation assay. Their antitumor activity was observed in NU/NU mice. The invasiveness of the cells was monitored using the wound healing assay. Live cell imaging was used to detect the distribution of the substances at the intracellular level. Other methods such as ELISA, microscale thermophoresis, and in silico predictions have been used to determine the selectivity of inhibitors of the signalling pathways IL-6 and IL-8....
