Atelocollagen separation and characterization

The aim of the presented bachelor thesis was to optimize separation of soluble atelocollagen out of commercially available bovine collagen and its characterization. Collagen went through an enzymatic digestion using pepsin as a protease and subsequently purified by filtration, centrifugation and dialysis. Obtained samples were characterized using infrared spectrometry, polyacrylamide gel electrophoresis and Hartree-Lowry assay for total protein determination. Suitable digestion time range to gain sufficient yields was decided from 60 – 70 hours at pepsin/collagen weight ratio 1:25. Highest yield (72.6 %) was obtained at mentioned ratio after 66 hours of reaction. Sufficient filtration, which was decisive for final product purity, was assured by combination of several filtration techniques with centrifugation. Quality of prepared product according to their molecular weight was characterized by gel electrophoresis. Solubility and its dependence on pH and NaCl concentration in solution were measured by Hartree – Lowry assay. Ratio between helical collagen and denaturated collagen was measured by infrared spectroscopy. Obtained spectra yet did not seem fully reliable and provided only a rough estimate. More accurate information about structure could be achieved by using circular dichroism spectroscopy.

Hlavním cílem této diplomové práce je navrhnout změnu motivačního systému ve firmě CRV CZ. Pozornost je věnována analýze současného motivačního systému a jeho nedostatkům. Na základě teorických východisek a dat sesbíraných z rozhovorů a dotazníkového šetření byly navrženy čtyři změny současného motivačního systému. Tyto návrhy zlepší motivaci zaměstnanců a jejich pracovní spokojenost, a tedy hospodářské výsledky celé firmy.
Modelling and Realization of Systems for Intelligent Houses

In my bachelors thesis I am focused on creating intelligent building control system Foxtrot. The main focus is on temperature control and security. The whole system is connected with an external sensor network. Between those two systems, the interface is created using Raspberry Pi and integrated web server.
Preparation and properties of silver nanoparticles on collagen matrix

Cílem předložené diplomové práce byla in-situ příprava stříbrných nanočástic na kolagenové matrici jako antibakteriálního povlaku a studie vlivu podmínek přípravy na vlastnosti nanočástic, zejména jejich velikost, tvar, homogenita jejich distribuce a antibakteriální aktivita. V rámci práce byla rovněž sledována kinetika redukce stříbrných nanočástic z dusičnanu stříbrného a vliv teploty na její průběh. Připravený materiál a jeho vlastnosti byly analyzovány pomocí různých technik. UV-VIS absorpčních vlastností stříbra bylo využito pro kinetické studie redukce a uvolňování nanočástic. Pomocí rastrovací elektronové mikroskopie byla vyhodnocena homogenita stříbrného povlaku a přibližná velikost částic a jejich aglomerátů. Velikostní distribuce nanočástic byla pak přesně stanovena pomocí dynamického rozptylu světla. Pomocí infračervené spektrometrie s Fourierovou transformací s technikou úplného zeslabeného odrazu byla sledována interakce stříbra s funkčními, zejména karboxylovými skupinami. Termogravimetricky byla stanovena tepelná stabilita a procentuální obsah stříbra v materiálu. Vliv AgNPs povlaku na 3D strukturu kolagenního scaffoldu a fázový kontrast pro 3D zobrazovací techniky byl zkoumán pomocí rentgenové výpočetní nanotomografie. V neposlední řadě byla také stanovena antibakteriální aktivita připraveného materiálu a její závislost na koncentraci stříbra.

The Bachelor Thesis is composed on the topic recruitment of employees by personal agency Manpower, Inc. On the basis of the estimation the processes recruitment and selection of employees in this agency, there are summarized the advantages and disadvantages, that brings the co-operation with the agency. The biggest advantages are the experiences and possibilities of employees and agency employment. Disadvantage is a high agency commission, which is compared with the costs of personal department in the company in the final part of thesis. During the last five years, the agency has added in all 3 494 employees for IBM. The company has already paid about 131 mil. CZK for these employees. In the case of recruitment by own employees the company would pay about 31 mil. CZK. The agency should reduce costs on selection of employees, for example by reduction of number of web portals, on which it advertises. Next the agency should improve the quality of the content of advertisement, where is missing the salary and description of company, for which the agency chooses the employees.
The Transformation of perception of vampire in Western culture from early modern period to present : The Assessment of marginal cultural phenomenon in terms of civilization theory of Norbert Elias

The thesis deals with the transformation of perception of vampire in Western culture from early modern period to present. The vampire in modern Western culture is very popular and has an unwavering place. Vampires are now attractive and basically not dangerous. The subject of this work is question whether it was so in previous centuries. Work present the change in appearance and character, both fictional character and "real vampires." Based on the analysis of selected fictional works (literary and visual) from the late 18th century, when the vampire came to Western fiction, work shows how gradually transforms the image that this supernatural beings attributes in West. An essential part of how culture perceives certain phenomenon is a belief in it. Therefore I am interested in how the West over time explaining this phenomenon. Work also introduces the Norbert Elias's civilization theory. Analysis of transformation in the perception of a vampire in Western culture follows it's tracks and the results are compared with it with the goal to give answer on question whether this phenomenon is turning into the terms of this theory.

Nemo tenetur se ipsum accusare principle Abstract This thesis deals with the concept and selected aspects of the nemo tenetur se ipsum accusare principle. This principle is one of the fundamental pillars of defence in criminal proceedings and is closely related to the right to a fair trial. The thesis focuses on the principle's theoretical starting points and attempts to outline problematic issues related to the interpretation of its content when applied in practice, particularly with regard to the permissible degree of coercion. The first chapter of the thesis deals with the historical development of the principle in both the continental and anglo-american legal systems. The second chapter contains an overview of how the principle is ensrished in international and national sources of law. The third chapter deals with the relationship between the privilege against self-incrimination and the right to remain silent based on the case law of the European Court of Human Rights and the US Supreme Court. The fourth chapter is devoted to the evidence, in particular the procedural consequences of an infringement in the proceedings, focusing on lawful and unlawful methods of coercion and the individual defects resulting therefrom.
