Overlaps of regulons of stress sigma factors of RNA polymerase in corynebacteria and rhodococci
Blumenstein, Jan ; Pátek, Miroslav (advisor) ; Hnilicová, Jarmila (referee) ; Ulrych, Aleš (referee)
Bacteria of the genera Rhodococcus and Corynebacterium are biotechnologically important microorganisms. This dissertation thesis focuses on the role of sigma (σ) factors of RNA polymerase in the regulation of gene expression in these bacteria. The C. glutamicum genome harbors 7 genes encoding σ factors, namely σA , σB , σC , σD , σE , σH and σM . With the exception of σM , these factors and their functions have been well described. The R. erythropolis genome encodes 21 putative genes for σ factors, but none of them has been analyzed in detail. The same holds for another biotechnologically useful bacterium, R. opacus. A group of genes whose expression is mediated by a single σ factor is called σ regulon. Overlaps of σ regulons are mainly caused by overlaps of recognition specificity of σ factors, i.e. when two or more σ factors recognize the same promoter. This thesis is focused on σ factors of group 4 (ECF) and their regulons: σD , σE , σH (C. glutamicum, R. erythropolis, R. opacus) and σM (C. glutamicum). We used RNA-sequencing, in vivo, in vitro and in silico methods (homology modeling and molecular dynamics simulations) to achieve the intended aims. The overlap of σD and σH regulons was demonstrated in the natural hybrid promoter Pcg0441. Using the combination of in vivo and in silico methods,...
Increase in muscle mass during remission in patients with Crohn's disease
Dittrichová, Jana ; Křížová, Jarmila (advisor) ; Šatný, Martin (referee)
Pacients with Crohn's disease lose a significant amount of muscle mass during the acute phase. The amount of muscle mass plays a crucial role in determining an individual's physical fitness, quality of life, and improves the prognosis of the disease. The theoretical part of the presented work focuses on the importance of muscle mass, its growth in relation to protein intake, the issues related to Crohn's disease, and the increase of muscle mass during remission. In the practical part of the study, the goal was to determine how many kilograms patients with Crohn's disease typically lose during the acute phase of the illness and compare it with the weight loss in patients with ulcerative colitis. The aim was also to verify the impact of protein intake education and recommendations for the frequency and type of exercise on muscle mass growth, increased muscle strength, and walking speed in patients with Crohn's disease. In the practical part, it was found that patients with Crohn's disease lose between 11.79 to 16.29 kg during the acute phase of the illness. In the study with education, each participant underwent two measurements with an interval of 4-6 weeks. The parameters examined included the amount of muscle mass, a 4-meter walking test, and handgrip strength. After the first measurement,...
Synthesis of microwave resonators
Šeděnka, Vladimír ; Dědková, Jarmila (referee) ; Raida, Zbyněk (advisor)
The thesis deals with microwave resonators synthesis in Matlab environment. Due to the multi-objective optimization, the shape of the resonátor is subsequently changed to fit requested frequency properties. Particle swarm optimization and frequency domain finite element method are used. Accuracy of the different number of elements was tested. Values of several optimization parameters were balanced to reach the maximal efficiency of the optimization.
The Students themselves - Student Center Poříčí
Dvořáková, Eva ; Bambuchová, Jarmila (referee) ; Sochor, Jan (advisor)
The proposed building responds to a chamfered corner of the solved plot by turning the mass of the dining hall on the ground floor and the mass of the student dormitory above it. Under the protruding upper mass arises covered main entrance to the building. The ground floor of the building is directly connected to the adjacent hotel Austerlitz. The higher levels are stepping back from the street front. The mass of the student dormitory respects here the current glass corner on the top floor of the building Austerlitz. The facades to the streets Táborského nábřeží and Vídeňská are covered with a wire mesh to enable the creepers grow on it. There will be gaps in the wire mesh in front of the windows to the student rooms. The student housing is designed as maisonettes. These two storey maisonettes are divided into several groups including 5 to 8 rooms. Each group has its own “living room” and kitchenette.

Production single parts from sheet metal
Hartelová, Karolína ; Matušková, Jarmila (referee) ; Žák, Ladislav (advisor)
The elaborate project present suggestion of technology of produktion pressed piece from engineering steel 11 375 in framework of engineering studies in branch 2307. Production series do 100 000 pieces. By virtue of literary study of surface forming work and calculation was devised procedural tool. The tool is solved by form usual rack fix on crank press LEN 40 (producer TOMA Industries Trnava), with traction 400 kN. Punch and stamping die are made from tool steel 19 436.3 (POLDI 2002) along graphic documentation.
Minimization number of measured data for magnetostatic field mapping
Csonka, Gábor ; Bartušek, Karel (referee) ; Dědková, Jarmila (advisor)
This work deals with questions of mapping statical magnetical field and methods analyses plumbed dates. There is mentioned solving of problems and interpretation of results. Work is centered on evaluation map of magnetic field in workspace of MR tomography from data measured on surface shere. Imposition of submission is proposition acceptable mathematical method and trought program MATLAB compile algorithm for interpretation map magnetic field. In work is further included calculation resulting values of coefficients for induction of magnetic field for selected parts of sphere. By force of admeasurement were experimentally verifyded values wich from the work proceed. Designed method is possible to use for calculation of coefficeints for magnetic induction at any point on sphere from booked up dates.

Phosphorus in sediments of small rivers
Zborovská, Jarmila ; Hrich, Karel (referee) ; Malá, Jitka (advisor)
This thesis deals with assessment of two small water courses Leskava and Troubsky brook. Streams are evaluated from two aspects: content of three forms of phosphorus TP, Panorg and Porg and hydromorphological assessment. Total phosphorus was determined by sample mineralization by H2SO4+H2O2. Pinorg and Porg were determined by a method published by Pardo. Hydromorphological assessment was done by methodology Králová at al.
